The ecosystem engineer onuphid polychaete Diopatra biscayensis has a continuous population in the Bay of Biscay from the Cantabria coast in Spain to southern Brittany in France. A group of disjunct populations also are found in the English Channel, separated from the Biscay population by more than 400 coastal kilometers. It remains unclear whether D. biscayensis is native to the Bay of Biscay; it is also debated whether the disjunct populations in the English Channel are relics of a formerly continuous population, or the product of recent introductions through aquaculture. Here, we use climate hindcasts to explore hypotheses about the D. biscayensis historical distribution in Europe. If D. biscayensis is native, its range would have been restricted to southern Iberia and the Mediterranean during the Last Glacial Maximum (21,000 BP). However, the species is completely absent from both regions today, further supporting its interpretation as a non‐native species. If it was historically present in Europe, the climate hindcasts are congruent with range contraction in the Last Glacial Maximum (21,000 BP), expansion in the Mid‐Holocene Warm Period (6000 BP), and contraction again in the past 1000 years (850–1850), prior to the first reports of D. biscayensis on the Spanish and French Atlantic coasts. However, the simulations do not support there being climatic refugia along the English Channel coast that would account for the existence of relic populations. Taken together, the evidence suggests that D. biscayensis has been introduced to the Bay of Biscay, and that disjunct populations in the English Channel are the result of recent transport through human activities, perhaps aquaculture.  相似文献

Sabellaria alveolata is a tube-building gregarious polychaete that constructs large biogenic reefs. In macrotidal shellfish ecosystems, this species competes for food with cultivated suspension-feeders. The suspension-feeding activity and clearance rate of S. alveolata were investigated in response to changes in seston concentration. A flow-through system was designed to study 225 cm2 reef blocks with more than 500 individuals. The experimental conditions were characterized by increasing concentrations of suspended particulate matter ranging from 6.5 to 153.8 mg L1, while the organic content of the diet (microalgae Skeletonema costatum) decreased inversely from 49 to 9%, to mimic the dilution of organic matter by inorganic particles, characteristic of tidal resuspension. We showed that the clearance rate exponentially decreased in relation to an increase in SPM concentration. Clearance rate was estimated at 5.3 10− 3 L h− 1 or 0.93 L h− 1 g− 1 (dry weight) for the lowest seston concentration (SPM = 6.5 mg L− 1) and reached the asymptote at CR = 1.97 10− 3 L h− 1 or 0.35 L h− 1 g− 1 (dry weight) when SPM exceeded 45 mg L− 1. Using picture analyses of polychaete movements, we showed that, paradoxically, an increase in SPM concentration did not adversely affect the feeding activity of S. alveolata since the number of filtering individuals remained stable from SPM = 6.5 to 55.5 mg L− 1. These values were applied at the scale of the bay of Mont-Saint-Michel (France) to demonstrate that the filtration pressure of large populations of wild suspension-feeders should not be underestimated when the carrying capacity has to be assessed in the context of increasing bivalve cultures.  相似文献

A new family of mGlu receptor orthosteric ligands called APTCs was designed and synthesized using a parallel chemistry approach. Amongst 65 molecules tested on mGlu4, mGlu6 and mGlu8 subtypes, (2S,4S)-4-amino-1-[(E)-3-carboxyacryloyl]pyrrolidine-2,4-dicarboxylic acid (8a06-FP0429) has been shown to be a full mGlu4 agonist and a partial mGlu8 agonist. In addition, 8a06 was shown to be selective versus group I and II mGlu subtypes. A possible explanation for this efficacy difference is proposed by docking experiment performed with molecular model of the receptor.  相似文献

Background

Malaria, an Anopheles-borne parasitic disease, remains a major global health problem causing illness and death that disproportionately affects developing countries. Despite the incidence of malaria, which remains one of the most severe infections of human populations, there is no licensed vaccine against this life-threatening disease. In this context, we decided to explore the expression of Plasmodium vaccine antigens fused to the granule bound starch synthase (GBSS), the major protein associated to the starch matrix in all starch-accumulating plants and algae such as Chlamydomonas reinhardtii.

Methods and Findings

We describe the development of genetically engineered starch granules containing plasmodial vaccine candidate antigens produced in the unicellular green algae Chlamydomonas reinhardtii. We show that the C-terminal domains of proteins from the rodent Plasmodium species, Plasmodium berghei Apical Major Antigen AMA1, or Major Surface Protein MSP1 fused to the algal granule bound starch synthase (GBSS) are efficiently expressed and bound to the polysaccharide matrix. Mice were either immunized intraperitoneally with the engineered starch particles and Freund adjuvant, or fed with the engineered particles co-delivered with the mucosal adjuvant, and challenged intraperitoneally with a lethal inoculum of P. Berghei. Both experimental strategies led to a significantly reduced parasitemia with an extension of life span including complete cure for intraperitoneal delivery as assessed by negative blood thin smears. In the case of the starch bound P. falciparum GBSS-MSP1 fusion protein, the immune sera or purified immunoglobulin G of mice immunized with the corresponding starch strongly inhibited in vitro the intra-erythrocytic asexual development of the most human deadly plasmodial species.

Conclusion

This novel system paves the way for the production of clinically relevant plasmodial antigens as algal starch-based particles designated herein as amylosomes, demonstrating that efficient production of edible vaccines can be genetically produced in Chlamydomonas.  相似文献

The brain response to auditory novelty comprises two main EEG components: an early mismatch negativity and a late P300. Whereas the former has been proposed to reflect a prediction error, the latter is often associated with working memory updating. Interestingly, these two proposals predict fundamentally different dynamics: prediction errors are thought to propagate serially through several distinct brain areas, while working memory supposes that activity is sustained over time within a stable set of brain areas. Here we test this temporal dissociation by showing how the generalization of brain activity patterns across time can characterize the dynamics of the underlying neural processes. This method is applied to magnetoencephalography (MEG) recordings acquired from healthy participants who were presented with two types of auditory novelty. Following our predictions, the results show that the mismatch evoked by a local novelty leads to the sequential recruitment of distinct and short-lived patterns of brain activity. In sharp contrast, the global novelty evoked by an unexpected sequence of five sounds elicits a sustained state of brain activity that lasts for several hundreds of milliseconds. The present results highlight how MEG combined with multivariate pattern analyses can characterize the dynamics of human cortical processes.  相似文献

In an inundated Mexican forest, 89 out of 92 myrmecophytic tank bromeliads (Aechmea bracteata) housed an associated ant colony: 13 sheltered Azteca serica, 43 Dolichoderus bispinosus, and 33 Neoponera villosa. Ant presence has a positive impact on the diversity of the aquatic macroinvertebrate communities (n = 30 bromeliads studied). A Principal Component Analysis (PCA) showed that the presence and the species of ant are not correlated to bromeliad size, quantity of water, number of wells, filtered organic matter or incident radiation. The PCA and a generalized linear model showed that the presence of Azteca serica differed from the presence of the other two ant species or no ants in its effects on the aquatic invertebrate community (more predators). Therefore, both ant presence and species of ant affect the composition of the aquatic macroinvertebrate communities in the tanks of Abracteata, likely due to ant deposition of feces and other waste in these tanks.  相似文献

Statistical coupling analysis (SCA) is a method for analyzing multiple sequence alignments that was used to identify groups of coevolving residues termed “sectors”. The method applies spectral analysis to a matrix obtained by combining correlation information with sequence conservation. It has been asserted that the protein sectors identified by SCA are functionally significant, with different sectors controlling different biochemical properties of the protein. Here we reconsider the available experimental data and note that it involves almost exclusively proteins with a single sector. We show that in this case sequence conservation is the dominating factor in SCA, and can alone be used to make statistically equivalent functional predictions. Therefore, we suggest shifting the experimental focus to proteins for which SCA identifies several sectors. Correlations in protein alignments, which have been shown to be informative in a number of independent studies, would then be less dominated by sequence conservation.  相似文献
