We have investigated the potential of the GTP synthesis pathways as chemotherapeutic targets in the human pathogen Cryptococcus neoformans, a common cause of fatal fungal meningoencephalitis. We find that de novo GTP biosynthesis, but not the alternate salvage pathway, is critical to cryptococcal dissemination and survival in vivo. Loss of inosine monophosphate dehydrogenase (IMPDH) in the de novo pathway results in slow growth and virulence factor defects, while loss of the cognate phosphoribosyltransferase in the salvage pathway yielded no phenotypes. Further, the Cryptococcus species complex displays variable sensitivity to the IMPDH inhibitor mycophenolic acid, and we uncover a rare drug-resistant subtype of C. gattii that suggests an adaptive response to microbial IMPDH inhibitors in its environmental niche. We report the structural and functional characterization of IMPDH from Cryptococcus, revealing insights into the basis for drug resistance and suggesting strategies for the development of fungal-specific inhibitors. The crystal structure reveals the position of the IMPDH moveable flap and catalytic arginine in the open conformation for the first time, plus unique, exploitable differences in the highly conserved active site. Treatment with mycophenolic acid led to significantly increased survival times in a nematode model, validating de novo GTP biosynthesis as an antifungal target in Cryptococcus.  相似文献

为阐明陕西省植被净初级生产力(NPP)变化的整体状况,应用以遥感观测数据驱动的GLO-PEM模式模拟估计的NPP数据,对陕西省1981~2000年间的地表植被净初级生产力的空间分布及时间序列变化进行了综合分析.研究结果表明:(1)20年来,陕西省年NPP变化范围为687~858 g/(m2?a),平均值为749 g/(m2?a);(2)陕西省年平均NPP在波动中缓慢上升,NPP年增长幅度为3.248 2 g/(m2?a);(3)陕西省年平均NPP分布总体呈现从南到北递减的趋势,长城沿线风沙区NPP在200~400 g/(m2?a)之间,是陕西省NPP最低的地区;关中中东部NPP较高,大部在800 g/(m2?a)以上,NPP在1 200 g/(m2?a)以上的面积约占1/3,是全省NPP的高值区;(4)陕西省NPP基本不变(NPP变化百分率在-10%~10%之间)的面积占54.7%,NPP变化百分率增加10%以上的面积占40.5%,NPP变化百分率降低10%以下的面积仅占4.8%;(5)占陕西省总面积的77.3%的区域NPP变化不显著(P>0.05),NPP增加极显著(P<0.01)的区域占8.3%,增加显著(P<0.05)的区域占13.8%,NPP降低显著(P<0.05)和降低极显著(P<0.01)占0.7%.  相似文献

Group A Streptococcus (GAS) is a human pathogen that has the potential to cause invasive disease by binding and activating human plasmin(ogen). Streptococcal surface enolase (SEN) is an octameric α-enolase that is localized at the GAS cell surface. In addition to its glycolytic role inside the cell, SEN functions as a receptor for plasmin(ogen) on the bacterial surface, but the understanding of the molecular basis of plasmin(ogen) binding is limited. In this study, we determined the crystal and solution structures of GAS SEN and characterized the increased plasminogen binding by two SEN mutants. The plasminogen binding ability of SENK312A and SENK362A is ~2- and ~3.4-fold greater than for the wild-type protein. A combination of thermal stability assays, native mass spectrometry and X-ray crystallography approaches shows that increased plasminogen binding ability correlates with decreased stability of the octamer. We propose that decreased stability of the octameric structure facilitates the access of plasmin(ogen) to its binding sites, leading to more efficient plasmin(ogen) binding and activation.  相似文献

The possibility of conscious experiences of emotions in non-humananimals has been much less explored than that of conscious experiencesassociated with carrying out complex cognitive tasks. However,no great cognitive powers are needed to feel hunger or painand it may be that the capacity to feel emotions is widespreadin the animal kingdom. Since plants can show surprisingly sophisticated"choice" and "decision-making" mechanisms and yet we would notwish to imply that they are conscious, attribution of emotionsto animals has to be done with care. Breeding for wheat varieties resistant to Stagonospora nodorum blotch (SNB) is the most sustainable strategy for controlling the disease. In order to map quantitative trait loci (QTLs) for SNB resistance we analysed 204 recombinant inbred lines of the cross between the winter wheat (Triticum aestivum L.) variety Forno and the winter spelt (Triticum spelta L.) variety Oberkulmer. We determined the level of resistance of adult plants to leaf blotch (SNL) and glume blotch (SNG) as well as morphological traits for 2 years after artificial inoculation with S. nodorum. Using composite interval mapping and LOD > 3.7, we detected ten QTLs for SNG blotch resistance (six inherited from the susceptible parent Forno) and 11 QTLs for SNL resistance (four inherited from Forno) across 2 years. Both resistance traits were moderately correlated (r = 0.52) and had only one common QTL. For SNL resistance, seven QTLs were not associated with QTLs for morphological traits. Among them, QSnl.eth-2D, QSnl.eth-4B and QSnl.eth-7B3 had major effects (R(2) > 13%) and were potential candidates for marker-assisted selection. For SNG, the major QTL on chromosome 5A, explaining 36% of the phenotypic variance for resistance, was associated with the q locus conferring the spelt morphology (long lax ear, long culm and hard glumes). Only QSng.eth-1BS, which explained 7% of the variance for resistance to SNG blotch, was not associated with QTLs for morphological traits. The consequences for breeding programmes are discussed.  相似文献

SpyCEP is a Streptococcus pyogenes protease that cleaves CXCL8/IL‐8 and its activity is associated with human invasive disease severity. We investigated the role of SpyCEP in S. pyogenes necrotizing fasciitis and respiratory tract infection in mice using isogenic strains differing only in SpyCEP expression. SpyCEP cleaved human CXCL1, 2, 6 and 8 plus murine CXCL1 and 2 at a structurally conserved site. Mice were infected in thigh muscle with a strain of S. pyogenes that expresses a high level of SpyCEP, or with an isogenic non‐SpyCEP expressing strain. SpyCEP expression by S. pyogenes hindered bacterial clearance from muscle, and enhanced bacterial spread, associated with cleavage of murine chemoattractant CXCL1. Mice were then infected with Lactococcus lactis strains that differed only in SpyCEP expression. In contrast to the parent L. lactis strain (lacks SpyCEP), which was avirulent when administered intramuscularly, infection with a strain that expressed SpyCEP heterologously led to dramatic systemic illness within 24 h, failure to clear bacteria from muscle and marked dissemination to other organs. In the upper airways, SpyCEP expression was required for survival of L. lactis but not S. pyogenes. However, dissemination of S. pyogenes to the lung was SpyCEP‐dependent and was associated with evidence of chemokine cleavage. Taken together, the studies provide clear evidence that SpyCEP is necessary and sufficient for systemic bacterial dissemination from a soft tissue focus in this model and also underlies dissemination in the respiratory tract.  相似文献

IntroductionPrevalence of an abnormal Papanicolaou smear was significantly increased in lupus patients in cross-sectional studies, associated with a higher prevalence of high-risk human papillomavirus (HPV) infection. The nucleic acid-specific Toll-like receptors (TLRs) locate at the endolysosomal compartments and trigger the induction of cytokines for the innate immune response. This study evaluated whether abnormal host innate immune response in lupus patients may enhance HPV persistence.MethodsProtein levels of TLRs 3, 7, 8 and 9 in cervical epithelial cells of lupus patients and controls with or without HPV infection were assessed using flow cytometry. Characteristics associated with the differential expression of TLRs in systemic lupus erythematosus (SLE) were elucidated. The effect and interferon-stimulated genes (ISGs) (ISG15 and Mx-1) gene expressions were then measured in oncogenic HeLa (HPV18), CaSki (HPV) and C33A (HPV negative) cell lines using flow cytometry and quantitative real-time PCR. Ex vivo productions of cytokines and interferon-gamma (IFN-γ) upon TLR ligands stimulations were subsequently measured using cytometric bead array and ELISA.ResultsFor subjects with HPV infection, levels of TLR3 and TLR7 were significantly lower in lupus patients compared with controls. Significantly decreased TLRs 7, 8 and 9 levels were observed in HPV-negative SLE compared to healthy controls. For SLE with and without HPV infection, TLR7 and 9 levels were significantly lower in infected SLE than those in HPV-negative patients. Independent explanatory variables associated with down-regulation of TLR7 level included HPV infection and a higher cumulative dose of prednisolone; while a higher cumulative dose of hydroxychloroquine and HPV infection were associated with down-regulation of TLR9 level. In cervical cell lines, TLRs 3, 7, 8, 9 protein levels and antiviral ISG15 and Mx-1 gene expressions were inhibited in two oncogenic HPV types. Functional data showed that the induction of pro-inflammatory cytokines by TLR ligands (R837, ssRNA and ODN2395) was greatly impaired in CaSki and HeLa than C33A cells.ConclusionsIn conclusion, prednisolone and TLR antagonist (hydroxychloroquine) may down-regulate protein levels of TLR7 and TLR9 in lupus patients, thereby decreasing the innate immune response against HPV infection. Upon infection, HPV further down-regulate TLR7 and 9 levels for viral persistence. Furthermore, reduction of nucleic acid-sensing TLRs 7, 8 and 9 in carcinogenic HPVs ensures that the expression of inducible pro-inflammatory cytokines is minimized to prevent the expression of antiviral ISGs (ISG15 and Mx-1) on a biologically relevant antiviral response.  相似文献

Summary We examined how predation by vespid wasps,Polistes dominulus andP. fuscatus, affected the behavior, growth rate and survivorship of aggregated caterpillars ofHemileuca lucina (Saturniidae). Although these larvae can exhibit a variety of defense and escape behaviors, in general larvae reacted to wasp attacks by clinging to the hostplant. Neighboring larvae in the aggregation responded by leaving the feeding site and moving to the interior or base of the plant. To determine wheter wasp attack affected the behavior and growth of the caterpillars that escaped, a field experiment was conducted with treatments of: 1) larvae exposed to wasps, 2) larvae protected from wasps, and 3) larvae protected from wasps but with the attack of wasps simulated (=harassment). Over just one instar, protected larvae gained significantly more weight than the harassed larvae, which in turn weighed significantly more than the larvae that escaped the wasps. The behavior of attacked and harassed larvae differed from that of the protected larvae; the disturbed larvae often fed in smaller groups and in shaded portions of the plant where only mature leaves were available. A laboratory experiment showed that at 35° C (daytime temperature) larvae had significantly higher relative growth rates and significantly shorter instar duration than larvae reared at 25° C. Our results suggest that wasps, in addition to killing caterpillars, indirectly affect larval fitness by slowing larval growth, at least in part by forcing larvae into cooler microhabitats where leaves are of lower quality.  相似文献
