Competitive interactions between coinfecting genotypes of the same pathogen can impose selection on virulence, but the direction of this selection depends on the mechanisms behind the interactions. Here, we investigate how host immune responses contribute to competition between clones in mixed infections of the rodent malaria parasite Plasmodium chabaudi. We studied single and mixed infections of a virulent and an avirulent clone and compared the extent of competition in immunodeficient and immunocompetent mice (nude mice and T cell-reconstituted nude mice, respectively). In immunocompetent mice, the avirulent clone suffered more from competition than did the virulent clone. The competitive suppression of the avirulent clone was alleviated in immunodeficient mice. Moreover, the relative density of the avirulent clone in mixed infections was higher in immunodeficient than in immunocompetent mice. We conclude that immune-mediated interactions contributed to competitive suppression of the avirulent clone, although other mechanisms, presumably competition for resources such as red blood cells, must also be important. Because only the avirulent clone suffered from immune-mediated competition, this mechanism should contribute to selection for increased virulence in mixed infections in this host-parasite system. As far as we are aware, this is the first direct experimental evidence of immune-mediated apparent competition in any host-parasite system.  相似文献

The discovery of proteins that recognize membrane curvature created a paradigm shift by suggesting that membrane shape may act as a cue for protein localization that is independent of lipid or protein composition. Here we review recent data on membrane curvature sensing by three structurally unrelated motifs: BAR domains, amphipathic helices and membrane-anchored proteins. We discuss the conclusion that the curvature of the BAR dimer is not responsible for sensing and that the sensing properties of all three motifs can be rationalized by the physicochemical properties of the curved membrane itself. We thus anticipate that membrane curvature will promote the redistribution of proteins that are anchored in membranes through any type of hydrophobic moiety, a thesis that broadens tremendously the implications of membrane curvature for protein sorting, trafficking and signaling in cell biology.  相似文献

We report a novel approach to quantitatively determine complete size distributions of surface-bound objects using fluorescence microscopy. We measure the integrated intensity of single particles and relate it to their size by taking into account the object geometry and the illumination profile of the microscope, here a confocal laser scanning microscope. Polydisperse (as well as monodisperse) size distributions containing objects both below and above the optical resolution of the microscope are recorded and analyzed. The data is collected online within minutes, which allows the user to correlate the size of an object with the response from any given fluorescence-based biochemical assay. We measured the mean diameter of extruded fluorescently labeled lipid vesicles using the proposed method, dynamic light scattering, and cryogenic transmission electron microscopy. The three techniques were in excellent agreement, measuring the same values within 7-9%. Furthermore we demonstrated here, for the first time that we know of, the ability to determine the full size distribution of polydisperse samples of nonextruded lipid vesicles. Knowledge of the vesicle size distribution before and after extrusion allowed us to propose an empirical model to account for the effect of extrusion on the complete size distribution of vesicle samples.  相似文献

The phenology ofAsphodelus aestivus Brot. is described by means of a phenological model which has been formulated to fit skewed phenological data. Based on the model parameters the timing of different phenophases of biomass accumulation were determined. The biomass oscillations of leaves, inflorescence stalks and tubers were found to be synchronized with the predictable seasonal climatic changes. In addition, the plant seems to respond to minor random climatic variations. The emergence of leaves and inflorescence stalks depends on stored material in the tubers while leaf and inflorescence stalk elongation as well as flowering depends on current production. The storage part of the tubers seems to be a regulating structure, which is responsible for the synchronization ofA. aestivus productivity with the seasonality of the Mediterranean climate.A. aestivus is considered to be a Competitor Ruderal and Stress tolerant (C-R-S) strategist which may explain the wide distribution of this plant over the Mediterranean Basin.  相似文献

Habitat templets are graphical-qualitative models which describe the development of life-history strategies in specific environmental conditions. In the context of the previous models of life-history strategies, life-history theorists focused on the density-dependent factors as the factors determining life-history strategies. With the use of habitat templets, the focus is oriented towards the environmental causal factors, considering density-dependent phenomena as by-products of the environmental impact. This implies an important shift in causality as well as in the worldview of life-history theorists: population is not considered as a closed system isolated from the environment. The object of study is the organism-in-its-environment, as a complex multilevel system. This shift has also methodological consequences: Life-history theory combines holistic and reductionistic insights, using a variety of heuristic models. This imposes a new conception of generality as well as of the structure of scientific theories.  相似文献

The effect of the nematofauna on the microbiology and soil nitrogen status was studied in 6 major European grassland types (Northern tundra (Abisko, Sweden), Atlantic heath (Otterburn, UK), wet grassland (Wageningen, Netherlands), semi-natural temperate grassland (Linden, Germany), East European steppe (Pusztaszer, Hungary) and Mediterranean garigue (Mt. Vermion, Greece). To extend the range of temperature and humidity experienced locally during the investigation period, soil microclimates were manipulated, and at each site 14 plots were established representing selected combinations of 6 temperature and 6 moisture levels. The investigated soils divided into two groups: mineral grassland soils that were precipitation fed (garigue, wet grassland, seminatural grassland, steppe), and wet organic soils that were groundwater fed (heath, tundra). Effects of the nematofauna on the microflora were found in the mineral soils, where correlations among nematode metabolic activity as calculated from a metabolic model, and microbial activity parameters as indicated by Biolog and ergosterol measurements, were significantly positive. Correlations with bacterial activity were stronger and more consistent. Microbial parameters, in turn, were significantly correlated with the size of the soil nitrogen pools NH4, NO3, and Norganic. Furthermore, model results suggested that there were remarkable direct effects of nematodes on soil nitrogen status. Calculated monthly nematode excretion contributed temporarily up to 27% of soluble soil nitrogen, depending on the site and the microclimate. No significant correlation among nematodes and microbial parameters, or nitrogen pools, were found in the wet organic soils. The data show that the nematofauna can under favourable conditions affect soil nitrogen status in mineral grassland soils both directly by excretion of N, and indirectly by regulating microbial activity. This suggests that the differences in nitrogen availability observed in such natural grasslands partly reflect differences in the activity of their indigenous nematofauna. Penaeid and carid communities were assessed in St Lucia over a wide range of saline conditions, water level conditions and mouth states, including a six-month marine connection and various connections to the Mfolozi Estuary. Samples were collected biannually in spring and autumn from November 2004 to May 2012 using seine nets. Seven penaeid and seven carid species were recorded, of which Palaemon pacificus, P. peringueyi, Penaeus indicus and Metapenaeus monoceros were the most abundant. The decline from 2004 to 2007 in both species recorded and densities of freshwater, estuarine and marine species during the initial closed period was linked to increased salinity, lack of recruitment and reduced estuarine surface area. Increased wave action and high seas following Cyclone Gamede opened the mouth in March 2007 after a 57-month closed period, resulting in recruitment of marine species, predominantly P. indicus, followed by a decline in densities after reclosure in August 2007. Increases in marine and freshwater species were evident after Mfolozi flood connections during 2008, 2009 and 2010. Highest densities were recorded after breaching in 2007 and after flood connections, highlighting the importance of a marine link to maintain the recruitment of penaeid species.  相似文献

We monitored the action of phospholipase A2 (PLA2) on L- and D-dipalmitoyl-phosphatidylcholine (DPPC) Langmuir monolayers by mounting a Langmuir-trough on a wide-field fluorescence microscope with single molecule sensitivity. This made it possible to directly visualize the activity and diffusion behavior of single PLA2 molecules in a heterogeneous lipid environment during active hydrolysis. The experiments showed that enzyme molecules adsorbed and interacted almost exclusively with the fluid region of the DPPC monolayers. Domains of gel state L-DPPC were degraded exclusively from the gel-fluid interface where the buildup of negatively charged hydrolysis products, fatty acid salts, led to changes in the mobility of PLA2. The mobility of individual enzymes on the monolayers was characterized by single particle tracking. Diffusion coefficients of enzymes adsorbed to the fluid interface were between 3.2 μm2/s on the L-DPPC and 4.9 μm2/s on the D-DPPC monolayers. In regions enriched with hydrolysis products, the diffusion dropped to ≈0.2 μm2/s. In addition, slower normal and anomalous diffusion modes were seen at the L-DPPC gel domain boundaries where hydrolysis took place. The average residence times of the enzyme in the fluid regions of the monolayer and on the product domain were between ≈30 and 220 ms. At the gel domains it was below the experimental time resolution, i.e., enzymes were simply reflected from the gel domains back into solution.  相似文献
