Removal of polychlorinated biphenyls (PCBs) from contaminated sediments is a priority due to accumulation in the food chain. Recent success with reduction of PCB bioavailability due to adsorption onto activated carbon led to the recognition of in situ treatment as a remediation approach. In this study, reduced bioavailability and subsequent break-down of PCBs in dehalorespiring biofilms was investigated using Dehalobium chlorocoercia DF1. DF1 formed a patchy biofilm ranging in thickness from 3.9 to 6.7?µm (average 4.6?±?0.87?µm), while the biofilm coverage varied from 5.5% (sand) to 20.2% (activated carbon), indicating a preference for sorptive materials. Quantification of DF1 biofilm bacteria showed 1.2–15.3?×?109 bacteria per gram of material. After 22?days, coal activated carbon, bone biochar, polyoxymethylene, and sand microcosms had dechlorinated 73%, 93%, 100%, and 83%, respectively. These results show that a biofilm-based inoculum for bioaugmentation of PCBs in sediment can be an efficient approach.  相似文献

Gangjee et al. recently reported a novel series of 2-amino-4-methyl-5-phenylethyl substituted-7-benzyl-pyrrolo[2,3-d]pyrimidines, some of which exhibited two digit nanomolar antitumor and antimitotic activity and were not subject to P-glycoprotein (Pgp) or multidrug resistance protein 1 (MRP1) mediated tumor resistance (unlike the Vinca alkaloids and taxanes). Some of these compounds, in addition to their antitumor activity, had the ability to reverse the Pgp-mediated resistance to clinically used antimitotic agents. This report consists of an attempt to optimize the various activities of the parent compounds by synthetic variations of the phenyl ring of the 5-phenylethyl side chain. The target compounds were synthesized via a nine-step synthesis involving a Sonogashira reaction. The substituted phenylacetylenes as coupling partners were in turn synthesized from unactivated aryl bromides or iodides. The target compounds exhibited moderate cytotoxicity against MCF-7 tumor cells. However, most of these compounds showed improved cytotoxicity against the resistant NCI/ADR and MCF-7/VP. This study afforded an analog which reversed both Pgp-mediated as well as MRP1-mediated resistance to clinically used antimitotic agents, along with its own antimitotic mediated antitumor activity. In addition, in the NCI-60 cell line panel one of the compounds inhibited the growth of MDA-MD-435 breast cancer cell line at submicromolar concentration.  相似文献

Rhetinotheca tetrasolenata gen. et sp. n. is described from a Herrin #6 coal ball from Illinois. It consists of an ellipsoidal cluster of small synangia bearing spores of the Monoletes type. Although the synangia are partially connected to each other by sparse sterile tissue, evidence indicates that the synangia are immature, and it is presumed that they separated and spread apart at maturity. A small portion of a protostelic axis is present near the center of the cluster. Individual synangia consistently contain four sporangial tubes and measure 2.0–3.6 mm long by 0.7–1.2 mm in diam. A conspicuous central columella is present. When compared with compression forms, Rhetinotheca tetrasolenata compares most favorably with Aulacotheca iowensis. On this basis, arguments are advanced disputing the classical concept of whittleseyan fructifications. It is contended that none of them possessed a hollow central cavity as depicted in most reconstructions.  相似文献

This article is part of a Special Issue "Neuroendocrine-Immune Axis in Health and Disease." Microglia and astrocytes are the primary immune cells within the central nervous system. Microglia influence processes including neural development, synaptic plasticity and cognition; while their activation and production of immune molecules can induce stereotyped sickness behaviors or pathologies including cognitive dysfunction. Given their role in health and disease, we propose that glia may also be a critical link in understanding the etiology of many neuropsychiatric disorders that present with a strong sex-bias in their symptoms or prevalence. Specifically, males are more likely to be diagnosed with disorders that have distinct developmental origins such as autism or schizophrenia. In contrast, females are more likely to be diagnosed with disorders that present later in life, after the onset of adolescence, such as depression and anxiety disorders. In this review we will summarize the evidence suggesting that sex differences in the colonization and function of glia within the normal developing brain may contribute to distinct windows of vulnerability between males and females. We will also highlight the current gaps in our knowledge as well as the future directions and considerations of research aimed at understanding the link between neuroimmune function and sex differences in mental health disorders.  相似文献

The four WNK (with no lysine (K)) protein kinases affect ion balance and contain an unusual protein kinase domain due to the unique placement of the active site lysine. Mutations in two WNKs cause a heritable form of ion imbalance culminating in hypertension. WNK1 activates the serum- and glucocorticoid-induced protein kinase SGK1; the mechanism is noncatalytic. SGK1 increases membrane expression of the epithelial sodium channel (ENaC) and sodium reabsorption via phosphorylation and sequestering of the E3 ubiquitin ligase neural precursor cell expressed, developmentally down-regulated 4-2 (Nedd4-2), which otherwise promotes ENaC endocytosis. Questions remain about the intrinsic abilities of WNK family members to regulate this pathway. We find that expression of the N termini of all four WNKs results in modest to strong activation of SGK1. In reconstitution experiments in the same cell line all four WNKs also increase sodium current blocked by the ENaC inhibitor amiloride. The N termini of the WNKs also have the capacity to interact with SGK1. More detailed analysis of activation by WNK4 suggests mechanisms in common with WNK1. Further evidence for the importance of WNK1 in this process comes from the ability of Nedd4-2 to bind to WNK1 and the finding that endogenous SGK1 has reduced activity if WNK1 is knocked down by small interfering RNA.  相似文献

The objective of this study was to examine the effects of rest interval length on perceived exertion and during 3 sets of 10 inertial knee extension repetitions. Thirty healthy men (n = 15) and women (n = 15) volunteers were randomly assigned to 1 of 3 groups (1-, 2-, or 3-minute rest interval length) following the establishment of each subject's 1 repetition maximum (1RM) for inertial knee extension exercise. Subjects in each group performed 3 sets of 10 repetitions at 70% of a theoretical 10RM (based on each subject's 1RM), with a 1-, 2-, or 3-minute rest interval between each set. Perceived exertion was recorded, via the Borg category-ratio scale, from each subject after each repetition of each set. The results demonstrated no significant rest interval length effect on perceived exertion across the 3 sets of 10 repetitions. The results revealed a significantly higher perceived exertion value following the first repetition in set 3 as compared to sets 2 and 1 in all groups. The increase in perceived exertion within each set, as described by the slope, was found to be significantly lowest in set 1, as compared to sets 2 and 3. The major findings of this study demonstrate that perceived exertion significantly increases in a similar manner across 3 sets of 10 knee extension repetitions, despite rest interval lengths of 1-3 minutes.  相似文献
