Novel interaction between Apc5p and Rsp5p in an intracellular signaling pathway in Saccharomyces cerevisiae

The ubiquitin-targeting pathway is evolutionarily conserved and critical for many cellular functions. Recently, we discovered a role for two ubiquitin-protein ligases (E3s), Rsp5p and the Apc5p subunit of the anaphase-promoting complex (APC), in mitotic chromatin assembly in Saccharomyces cerevisiae. In the present study, we investigated whether Rsp5p and Apc5p interact in an intracellular pathway regulating chromatin remodeling. Our genetic studies strongly suggest that Rsp5p and Apc5p do interact and that Rsp5p acts upstream of Apc5p. Since E3 enzymes typically require the action of a ubiquitin-conjugating enzyme (E2), we screened E2 mutants for chromatin assembly defects, which resulted in the identification of Cdc34p and Ubc7p. Cdc34p is the E2 component of the SCF (Skp1p/Cdc53p/F-box protein). Therefore, we analyzed additional SCF mutants for chromatin assembly defects. Defective chromatin assembly extracts generated from strains harboring a mutation in the Cdc53p SCF subunit or a nondegradable SCF target, Sic1(Deltaphos), confirmed that the SCF was involved in mitotic chromatin assembly. Furthermore, we demonstrated that Ubc7p physically and genetically interacts with Rsp5p, suggesting that Ubc7p acts as an E2 for Rsp5p. However, rsp5CA and Deltaubc7 mutations had opposite genetic effects on apc5CA and cdc34-2 phenotypes. Therefore, the antagonistic interplay between Deltaubc7 and rsp5CA, with respect to cdc34-2 and apc5CA, indicates that the outcome of Rsp5p's interaction with Cdc34p and Apc5p may depend on the E2 interacting with Rsp5p. 相似文献

In total 52 samples of Sahiwal (19), Tharparkar (17), and Gir (16) were genotyped by using BovineHD SNP chip to analyze minor allele frequency (MAF), genetic diversity, and linkage disequilibrium among these cattle. The common SNPs of BovineHD and 54K SNP Chips were also extracted and evaluated for their performance. Only 40%?50% SNPs of these arrays was found informative for genetic analysis in these cattle breeds. The overall mean of MAF for SNPs of BovineHD SNPChip was 0.248?±?0.006, 0.241?±?0.007, and 0.242?±?0.009 in Sahiwal, Tharparkar and Gir, respectively, while that for 54K SNPs was on lower side. The average Reynold’s genetic distance between breeds ranged from 0.042 to 0.055 based on BovineHD Beadchip, and from 0.052 to 0.084 based on 54K SNP Chip. The estimates of genetic diversity based on HD and 54K chips were almost same and, hence, low density chip seems to be good enough to decipher genetic diversity of these cattle breeds. The linkage disequilibrium started decaying (r2?0.2) at 140?kb inter-marker distance and, hence, a 20K low density customized SNP array from HD chip could be designed for genomic selection in these cattle else the 54K Bead Chip as such will be useful. 相似文献

Intracellular rod-like inclusions were found in a high percentage of inhibited fourth-stage larvae of H. contortus in sheep with naturally acquired infections. Artificial infections showed inclusions occurred in developing as well as inhibited fourth-stage larvae. The presence of these structures was associated with degenerative changes of the parasites and serial worm counts showed that larvae with large numbers of inclusions failed to persist within the host.Large refractive hexagonal crystals were observed in the gut lumen of fourth-stage and adult O. ostertagi acquired by previously worm-free lambs grazed on cattle pastures. The crystals tended to accumulate in the posterior gut region where they may cause gut blockage and lead to early mortality of the parasites in the abnormal host. Rates of turnover and routes of excretion of 1-14C-ascorbic acid were compared among laboratory rabbits and two species of arboreal marsupials. Ringtail possums (Pseudocheirus peregrinus) exhibited higher blood ascorbate levels than either brushtail possums (Trichosurus vulpecula) or rabbits, as well as greater pool sizes, shorter half-times and higher total entry rates of ascorbic acid. These differences were due partly to the high ascorbic acid content of Eucalyptus foliage, the ringtail's diet; when the dietary contribution of ascorbate was taken into account, endogenous entry rates (i.e. 'synthesis') were not significantly different among species. The main excretory route of 14C from 1-14C-ascorbate was expired CO2 in all three species. Estimates to total pool size of ascorbate by isotope dilution were higher than those by tissue analysis. This was considered to be due to excretion of 14C during equilibration of injected 1-14C-ascorbate with the body pool. 相似文献
