6-Gingerol ( 1 ) is one of the major components in ginger and developing new synthetic methodologies could bring semisynthetic analogs with improved therapeutic properties. Towards this, multigram scale isolation of 6-gingerol with excellent purity was optimized using a simple and robust extraction, followed by column purification. Synthesis of 6-gingerdione, 7 from 6-gingerol was then achieved through selective –OTBDMS protection, DMP oxidation and deprotection reaction sequence for the first time. Compounds 1 , 7 and 8 (dehydrozingerone) exhibited excellent cell-free antioxidant properties in DPPH, ABTS, superoxide radical scavenging assay and H2O2 assay at 10–50 μM concentrations. The hemolytic study suggests that up to 50 μM, all three compounds did not exhibit toxicity to human erythrocytes. When H2O2 treated zebrafish larvae groups (96hpf) were exposed to compounds 1 , 7 and 8 , it increases the SOD (19, 19.1 and 18.7 U/mg protein), CAT (18.1, 16.5, and 15.8 μmol/mg levels and decreases the lipid peroxidation level (13, 15 and 18 nmol/mg protein), respectively. In vivo ROS levels and degree of cell death were studied using DCFDA and Acridine orange assays. Compounds 1 , 7 and 8 decreases the ROS and cell death level significantly. Taken together, compounds 1 , 7 and 8 exhibit excellent antioxidant properties, counteract H2O2 induced oxidative stress, reduces cell death in zebrafish larvae.  相似文献

Temperature and humidity are the two main factors that influence the rate of oviposition and the fecundity of Apanteles angaleti Muesebeck. At 90% relative humidity the same number of eggs is laid at 20, 25, 30 and 35°. Low humidities reduce oviposition.
Zusammenfassung Temperatur und Luftfeuchtigkeit sind die beiden Hauptfaktoren, welche die Eiablagerate und Fruchtbarkeit von Apanteles angaleti Muesebeck beeinflussen. Bei 90% relativer Luftfeuchtigkeit wird bei 20, 25, 30 und 35°C die gleiche Anzahl Eier abgelegt. Niedere Luftfeuchtigkeit verringert die Eiablage. Bei der Untersuchung des Lichteinflusses auf die Anzahl der produzierten Eier konnte kein signifikanter Unterschied gefunden werden.

A Scopulariopsis brevicaulis poultry farm isolate was chosen to study factors influencing keratinase production. The parameters were optimized by factorial design. The highest enzyme production by this fungus was obtained at pH 7.5, a temperature of 30 degrees C and a growth period of 5 weeks. The production of the enzyme was enhanced when the culture medium was supplemented with glucose (1%), sodium nitrate (2%), feather (1.5%) and CaCl(2) (1 mM). According to the responses from the experimental design, the effects of each variable were calculated, and the interactions between them were determined. The experimental values were found to be in accordance with the predicted values, the correlation coefficient is 0.9978.  相似文献

Summary Lipofuscin pigment formation and distribution in the Mes. N.5 neurons, trigeminal and spinal ganglia of male Wistar rats of 2, 14, 32 and 49 months as an indication of aging has been investigated. These intraneuronal pigment granules are found as early as 2 months in all the cells, and continue to accumulate in all the cells in varying amounts until the first year of life. The different rate at which lipofuscin accumulates probably shows the difference in the maturation of the functionally related cells. At later stages the obvious findings are complex pigment body formation and localization of the pigment bodies either at one pole as seen in the Mes. N. 5 neurons or arranged submembranously parallel to the long axis of the cells in the ganglia. The vacuolated lipofuscin pigment bodies are bound by a double limiting membrane and among the vacuoles are found tubular membranous structures resembling residual mitochondrial substructures. These findings suggest a mitochondrial origin of lipofuscin, rather than a lysosomal. The intracellular pigment bodies seen in the perineuronal satellite cells of peripheral ganglia appear to be signs of removal of lipofuscin from the ganglion cells. Acknowledgements. Human vaults are the largest cytoplasmic ribonucleoprotein and are overexpressed in cancer cells. Vaults reportedly function in the extrusion of xenobiotics from the nuclei of resistant cells, but the interactions of xenobiotics with the vault-associated proteins or non-coding RNAs have never been directly observed. In the present study, we show that vault RNAs (vRNAs), specifically the hvg-1 and hvg-2 RNAs, bind to a chemotherapeutic compound, mitoxantrone. Using an in-line probing assay (spontaneous transesterification of RNA linkages), we have identified the mitoxantrone binding region within the vRNAs. In addition, we analyzed the interactions between vRNAs and mitoxantrone in the cellular milieu, using an in vitro translation inhibition assay. Taken together, our results clearly suggest that vRNAs have the ability to bind certain chemotherapeutic compounds and these interactions may play an important role in vault function, by participating in the export of toxic compounds.  相似文献

OBJECTIVE--To determine the prevalence and predictors of smoking in urban India. DESIGN--Cross sectional. SETTING--Delhi, urban India, 1985-6. SUBJECTS--Random sample of 13,558 men and women aged 25-64 years. MAIN OUTCOME MEASURES--Smoking prevalence; subjects who were currently smoking and who had smoked > or = 100 cigarettes or beedis or chuttas in their lifetime were defined as smokers. RESULTS--45% (95% confidence interval 43.8 to 46.2) of men and 7% (6.4 to 7.6) of women were smokers. Education was the strongest predictor of smoking, and men with no education were 1.8 (1.5 to 2.0) times more likely to be smokers than those with college education, and women with no education were 3.7 (2.9 to 4.8) times more likely. Among smokers, 52.6% of men and 4.9% of women smoked only cigarettes while the others also smoked beedi or chutta. Compared with cigarette smokers, people smoking beedi or chutta were more likely to be older and married; have lower education, manual occupations, incomes, and body mass index; and not drink alcohol or take part in leisure exercise. CONCLUSION--There are two subpopulations of smokers in urban India, and the prevention strategy required for each may be different. The educated, white collar cigarette smoker in India might respond to measures that make non-smoking fashionable, while the less educated, low income people who smoke beedi or chutta may need strategies aimed at socioeconomic improvement.  相似文献
