Spirochaeta coccoides Dröge et al. 2006 is a member of the genus Spirochaeta Ehrenberg 1835, one of the oldest named genera within the Bacteria. S. coccoides is an obligately anaerobic, Gram-negative, non-motile, spherical bacterium that was isolated from the hindgut contents of the termite Neotermes castaneus. The species is of interest because it may play an important role in the digestion of breakdown products from cellulose and hemicellulose in the termite gut. Here we provide a taxonomic re-evaluation for strain SPN1T, and based on physiological and genomic characteristics, we propose its reclassification as a novel species in the genus Sphaerochaeta, a recently published sister group of the Spirochaeta. The 2,227,296 bp long genome of strain SPN1T with its 1,866 protein-coding and 58 RNA genes is a part of the Genomic

The objective of the study was to compare growth performance, carcass characteristics, meat quality and fatty acid composition of the adipose tissue of group-penned barrows, immunocastrated boars and entire males. Furthermore, the effect of housing of entire males on the aforementioned parameters was evaluated. At 55.2 days of age, 52 Swiss Large White pigs were blocked by litter and assigned by BW to four experimental groups: barrows (C), immunocastrated boars (IC), entire males (EMG) reared in group pens and entire males (EMP) reared in individual pens. In experiment 1, the effects of the method of castration were investigated (experimental groups C, IC and EMG). In experiment 2, the effects of housing on entire male pigs were evaluated (experimental groups EMG and EMP). All pigs had ad libitum access to standard diets from weaning to 107 kg BW. The two vaccinations (Improvac®) were applied to the IC pigs at an average BW of 22.6 and 73.0 kg. In experiment 1, average daily gain (ADG) did not (P > 0.05) differ among the experimental groups. However, EMG consumed less feed and had a better feed-conversion ratio than C (P < 0.001 for each). For IC, intermediate values were observed, which differed (P < 0.001) from EMG and C. Lean meat percentage decreased (P < 0.05) from EMG to IC, and from IC to C. The androstenone and skatole levels were higher (P < 0.05) in the adipose tissue of EMG than IC and C. Shear force values were higher (P < 0.01) in the longissimus muscle of C and EMG, compared to IC. The concentration of saturated fatty acid in the adipose tissue increased (P < 0.001) from EMG to IC, and from IC to C pigs, and that of polyunsaturated fatty acid decreased (P < 0.001). In experiment 2, ADG did not (P > 0.05) differ between EMP and EMG. However, EMP pigs consumed more feed than EMG pigs and had a poorer feed efficiency (P < 0.01 for each). In conclusion, EMG pigs had a better feed efficiency than IC pigs and their carcasses were leaner, but the risk of boar tainted pork was elevated. Group-housing negatively affected average daily feed intake but not ADG of entire males. The value of the metazoan parasites of the anchoveta Engraulis ringens as biological tags for stock discrimination is assessed. Three hundred and fifty-nine specimens, obtained from six landing ports in northern and central Chile (Arica, Antofagasta, Caldera, Coquimbo, Valparaiso and Talcahuano) covering a latitudinal gradient of c . 18°, were analysed. Six metazoan parasite species (three ectoparasites and three endoparasites) were obtained. A correspondence analysis suggested that anchoveta from northern and central Chile are comprised of two stocks. Identification of stocks was largely based on qualitative differences (absence in southern localities of the monogenean Pseudanthocotyloides heterocotyle , a parasite proper of engraulids, and the copepod Caligus sp.) and quantitative differences in the prevalence of infection for the remaining species. At present, information on the effects of ultraviolet radiation (UVR) on structure and diversity of polar, in particular Arctic,
benthic communities is scarce. It is unclear whether and to what extent communities of different successional age are susceptible
to UVR and whether UVR effects known to be detrimental at the species level can be buffered at the community level. In a subtidal
field study on Spitsbergen (Norwegian Arctic), we investigated the potential effect of distinct UVR regimes on macrobenthic
communities of different successional ages, grown on ceramic tiles. Total taxon cover, taxon composition, evenness, and richness
were assessed after experimental exposure of 4 and 8 weeks. Overall, 17 algal and invertebrate taxa were encountered in the
study and diatoms dominated the communities regardless of successional age or radiation treatment. UVR effects were dependent
on both exposure time and community age. We did not find overall detrimental UVB effects. In contrast, abundance of several
species increased in UVR-exposed communities. Especially, UVA seemed to have a beneficial effect in that several green and
brown algal taxa increased in abundance (e.g. Ulothrix flacca, Chlorophyta, and Desmarestia sp., Phaeophyceae). In general, UVR effects depended on species composition and thus on successional age of communities,
with later successional communities likely to be able to buffer and alleviate possible negative effects of UVR at species
level. Overall, the presented study provides a first insight into the complex role UVR plays in structuring Arctic epibenthic

We studied the pollination and reproductive success in continuous and fragmented populations of Lapageria rosea, a self-compatible plant endemic to temperate forests of Chile. Pollinator abundance, visitation rates, flower abundance,
nectar volume and concentration, pollen germination and fruit and seed production, were compared between continuous forest
of 145 ha and four forest fragments of 6, 3, 3, and 1 ha respectively, surrounded by mature pine plantations of Pinus radiata. Flower abundance was lower in three out of four forest fragments relative to continuous forest. Nectar volume and sugar
concentration did not differ between flowers in the two habitats. Pollinators of L. rosea, the hummingbird Sephanoides sephaniodes and bumblebee Bombus dahlbomii were less abundant and visited flowers of L. rosea at lower rates in fragments than in continuous forest. In addition, in vitro rates of pollen germination were lower for flowers in forest fragments. The number of seeds per fruit was also lower in forest
fragments. We suggest that fragmentation affects the reproductive success of L. rosea, lowering the total numbers of seeds produced and possibly compromising long term persistence of fragmented populations. 相似文献

Blood monocyte-derived macrophages invading the alveolus encounter pulmonary surfactant, a phospholipoprotein complex that changes composition during lung development. We tested the hypothesis that characteristic phosphatidylcholine (PC) components differentially influence macrophage phenotype and function, as determined by phagocytosis of green fluorescent protein-labeled Escherichia coli and alphaCD3-induced T cell proliferation. Human macrophages were exposed to surfactant (Curosurf(R)), to two of its characteristic phosphadidylcholine (PC) components (dipalmitoyl-PC and palmitoylmyristoyl-PC), and to a ubiquituous PC (palmitoyloleoyl-PC) as control. Interaction of Curosurf and PC species with macrophages was assessed using Lissaminetrade mark-dihexadecanoyl-phosphoethanolamine-labeled liposomes. Curosurf and both saturated surfactant PC species downregulated CD14 expression and upregulated CD206. HLA-DR and CD80 were upregulated by Curosurf and palmitoylmyristoyl-PC, whereas dipalmitoyl-PC showed no effect. The latter upregulated TLR2 and TLR4 expression, whereas Curosurf and palmitoylmyristoyl-PC had no effect. PC species tested were incorporated in comparable amounts by macrophages. Curosurf and PC species inhibited phagocytosis of E. coli. Scavenger receptor CD36, CD68, SR-A, and LOX-1 mRNA expression was upregulated by Curosurf, whereas PC species only upregulated SR-A. Curosurf and palmitoylmyristoyl-PC inhibited alphaCD3-induced T cell proliferation by 50%, whereas dipalmitoyl-PC and palmitoyloleoyl-PC showed no effect. The maintenance of genomic stability relies on the spindle assembly checkpoint (SAC), which ensures accurate chromosome segregation by delaying the onset of anaphase until all chromosomes are properly bioriented and attached to the mitotic spindle. BUB1 and BUBR1 kinases are central for this process and by interacting with Blinkin, link the SAC with the kinetochore, the macromolecular assembly that connects microtubules with centromeric DNA. Here, we identify the Blinkin motif critical for interaction with BUBR1, define the stoichiometry and affinity of the interaction, and present a 2.2 ? resolution crystal structure of the complex. The structure defines an unanticipated BUBR1 region responsible for the interaction and reveals a novel Blinkin motif that undergoes a disorder-to-order transition upon ligand binding. We also show that substitution of several BUBR1 residues engaged in binding Blinkin leads to defects in the SAC, thus providing the first molecular details of the recognition mechanism underlying kinetochore-SAC signaling. 相似文献

Environmental stress affects species richness and diversity in communities, but the precise form of the relationship is unclear.
We tested an environmental stress model (ESM) that predicts a unimodal pattern for total richness and diversity in local communities
across the full stress gradient where a regional biota can occur. In 2008, we measured richness and diversity (considering
all macrobenthic species) across the entire intertidal range on three rocky shores on Helgoland Island, Germany. Intertidal
elevation is known to be positively related to abiotic stress. Since Helgoland is between the northern and southern biogeographic
boundaries for the cold-temperate NE Atlantic intertidal biota, it exhibits low stress levels for this biota at low elevations
and high stress at high elevations because of long (>6 h) emersion times. Thus, we predicted a unimodal trend for richness
and diversity across elevation. On all three shores, richness increased from high to middle elevations, but remained similar
between middle and low elevations. Diversity followed the same trend on one shore and different trends (although also non-unimodal)
on the other two. Evenness explained the trend differences between richness and diversity. Overall, our study yielded little
support for the ESM. Reasons for richness and diversity not decreasing at low elevations may be related to influences of mostly
subtidal species, Helgoland’s intertidal range, or sampling resolution. Our study also suggests that the ESM must be developed
further to differentiate between richness and diversity. We offer recommendations to improve future ESM research using intertidal

In Escherichia coli the codons CGG, AGG, UGG or GGG (NGG codons) but not GGN or GNG (where N is non-G) are associated with low expression of a reporter gene, if located at positions +2 to +5. Induction of a lacZ reporter gene with any one of the NGG codons at position +2 to +5 does not influence growth of a normal strain, but growth of a strain with a defective peptidyl-tRNA hydrolase (Pth) enzyme is inhibited. The same codons, if placed at position +7, did not give this effect. Other codons, such as CGU and AGA, at location +2 to +5, did not give any growth inhibition of either the wild-type or the mutant strain. The inhibitory effect on the pth mutant strain by NGG codons at location +5 was suppressed by overexpression of the Pth enzyme from a plasmid. However, the overexpression of cognate tRNAs for AGG or GGG did not rescue from the growth inhibition associated with these codons early in the induced model gene. The data suggest that the NGG codons trigger peptidyl-tRNA drop-off if located at early coding positions in mRNA, thereby strongly reducing gene expression. This does not happen if these codons are located further down in the mRNA at position +7, or later. 相似文献
