Polyadenylated mRNA extracted from cytoplasm of measles virus-infected Vero cells was translated in a cell-free system. Three of the polypeptides obtained corresponded to nucleocapsid protein, phosphoprotein, and membrane protein of measles virions. A fourth polypeptide, present in measles virus-infected cells, could be generated by addition of Vero cytoplasmic extract and was identified as a cleavage product of the nucleocapsid protein.  相似文献

Previously, we reportedthat red blood cells (RBCs) of rabbits and humans release ATP inresponse to mechanical deformation and that this release of ATPrequires the activity of the cystic fibrosis transmembrane conductanceregulator (CFTR). It was reported that cAMP, acting through acAMP-dependent protein kinase, PKA, is an activator of CFTR. Here weinvestigate the hypothesis that cAMP stimulates ATP release from RBCs.Incubation of human and rabbit RBCs with the direct activator ofadenylyl cyclase, forskolin (10 or 100 µM), with IBMX (100 µM),resulted in ATP release and increases in intracellular cAMP. Inaddition, epinephrine (1 µM), a receptor-mediated activator ofadenylyl cyclase, stimulated ATP release from rabbit RBCs. Moreover,incubation of human and rabbit RBCs with an active cAMP analog[adenosine 3'5'-cyclic monophosphorothioate Sp-isomer (Sp-cAMP, 100 µM)] resulted in ATP release. In contrast, forskolin and Sp-cAMPwere without effect on dog RBCs, cells known not to release ATP inresponse to deformation. When rabbit RBCs were incubated with theinactive cAMP analog and inhibitor of PKA activity, adenosine3',5'-cyclic monophosphorothioate Rp-isomer (100 µM),deformation-induced ATP release was attenuated. These results areconsistent with the hypothesis that adenylyl cyclase and cAMP arecomponents of a signal-transduction pathway relating RBC deformation toATP release from human and rabbit RBCs.

The Zebrafish Information Network, ZFIN, is a WWW community resource of zebrafish genetic, genomic and developmental research information (http://zfin.org). ZFIN provides an anatomical atlas and dictionary, developmental staging criteria, research methods, pathology information and a link to the ZFIN relational database (http://zfin. org/ZFIN/). The database, built on a relational, object-oriented model, provides integrated information about mutants, genes, genetic markers, mapping panels, publications and contact information for the zebrafish research community. The database is populated with curated published data, user submitted data and large dataset uploads. A broad range of data types including text, images, graphical representations and genetic maps supports the data. ZFIN incorporates links to other genomic resources that provide sequence and ortholog data. Zebrafish nomenclature guidelines and an automated registration mechanism for new names are provided. Extensive usability testing has resulted in an easy to learn and use forms interface with complex searching capabilities.  相似文献
