RNA synthesis in ovarian follicles of Drosophila melanogaster was studied by methods which eliminate experimentally induced alterations in gene expression. Gel electrophoresis of follicular RNA, labeled after injection of precursors into females, revealed qualitative and quantitative differences in synthesis during the course of oogenesis. A highly heterogeneous group of poly(A)-containing RNAs is produced during much of the course of follicular development. However, post-vitellogenic stages synthesize a small number of stage-specific poly(A)-containing RNAs. During this period, RNA synthesis is known to take place primarily in the follicle cells, which are engaged in the production of the endochorion and exochorion. Two intense bands of nonmitochondrial poly(A)+ RNA are labeled between stage 11 and early stage 13. The synthesis of a more heterogeneous group of very small poly(A)-containing RNAs characterizes the last part of oogenesis, stages 13 and 14. Evidence is presented to show that these RNAs are specifically localized in the follicle cells of the egg chamber. We propose that they represent mRNAs for chorion proteins. In situ hybridization of preparations of late stage poly(A)-containing RNA to salivary gland chromosomes revealed two major sites of complementarity, 7E11 and 12E, as well as several minor sites. Experiments in which RNAs were separated on gels prior to hybridization in situ suggested that both the major stage 12-specific RNA bands contained molecules which were complementary to DNA in the 7E11 region. It is particularly interesting that this site is within a small chromosomal interval known to contain the gene ocelliless. Females homozygous for ocelliless have been shown to produce structurally abnormal chorions (Johnson and King, 1974).  相似文献

In a previous paper (Yang et al., Biophys. J. 75:641-645, 1998), we showed a simple, efficient method of recording the diffraction patterns of supramolecular peptide assemblies in membranes where the samples were prepared in the form of oriented multilayers. Here we develop a method of analysis based on the diffraction theory of two-dimensional liquids. Gramicidin was used as a prototype model because its pore structure in membrane in known. At full hydration, the diffraction patterns of alamethicin and magainin are similar to gramicidin except in the scale of q (the momentum transfer of scattering), clearly indicating that both alamethicin and magainin form pores in membranes but of different sizes. When the hydration of the multilayer samples was decreased while the bilayers were still fluid, the in-plane positions of the membrane pores became correlated from one bilayer to the next. We believe that this is a new manifestation of the hydration force. The effect is most prominent in magainin patterns, which are used to demonstrate the method of analysis. When magainin samples were further dehydrated or cooled, the liquid-like diffraction turned into crystal-like patterns. This discovery points to the possibility of investigating the supramolecular structures with high-order diffraction.  相似文献

Understanding how stem-cell proliferation is controlled to maintain adult tissues is of fundamental importance. Drosophila oogenesis provides an attractive system to study this issue since cell production in the ovary depends on small populations of observable germ-line and somatic stem cells. By controlling the amount of protein-rich nutrients in the diet, we established conditions under which the rate of egg production varied 60-fold. Using a cell-lineage labeling system, we found that both germ-line and somatic stem cells, as well as their progeny, adjust their proliferation rates in response to nutrition. However, the number of active stem cells does not appear to change. Proliferation rates varied fourfold; the remaining 15-fold difference in egg production resulted from different frequencies of cell death at two precise developmental points: (1) the region 2a/2b transition within the germarium, and (2) stage 8 egg chambers that are entering vitellogenesis. To initiate a genetic analysis of these changes in cell proliferation and apoptosis, we show that ovarian cells require an intact insulin pathway to fully upregulate their rate of cycling in response to a protein-rich diet and to enter vitellogenesis.  相似文献

目的:了解铅锌镉联合染毒对大鼠血液系统的影响及营养干预对其损伤的修复作用。方法:选择SPF级初断乳Wistar大鼠72只,随机分为对照组、染毒组和干预组,分别采用生理盐水、铅锌镉联合染毒液及染毒后以营养干预液灌胃28天和56天之后,检测其血液系统中五元素和血细胞的指标。结果:染毒组较对照组大鼠血铜、血锌含量高,血钙含量低于对照组,差异均有统计学意义(P〈0.05);染毒组血铜含量高于干预组,血钙含量低于干预组,差异均有统计学意义(P〈0.05);干预组红细胞(RBC)计数、血红蛋白(Hb)、血细胞比容(HCT)均高于染毒组,差异均有统计学意义(P〈0.05);对照组白细胞(WBC)计数高于染毒组、干预组,差异均有统计学意义(P〈0.05)。结论:铅镉对大鼠血铜、血钙、血锌水平有影响;综合营养干预对重金属元素造成的血液系统损伤有明显的拮抗作用,对血液系统有一定的保护及修复作用。  相似文献

The Drosophila ovariole tip produces new ovarian follicles on a 12-hour cycle by controlling niche-based germline and follicle stem cell divisions and nurturing their developing daughters. Static images provide a thumbnail view of folliculogenesis but imperfectly capture the dynamic cellular interactions that underlie follicle production. We describe a live-imaging culture system that supports normal ovarian stem cell activity, cyst movement and intercellular interaction over 14 hours, which is long enough to visualize all the steps of follicle generation. Our results show that live imaging has unique potential to address diverse aspects of stem cell biology and gametogenesis. Stem cells in cultured tissue respond to insulin and orient their mitotic spindles. Somatic escort cells, the glial-like partners of early germ cells, do not adhere to and migrate along with germline stem cell daughters as previously proposed. Instead, dynamic, microtubule-rich cell membranes pass cysts from one escort cell to the next. Additionally, escort cells are not replenished by the regular division of escort stem cells as previously suggested. Rather, escort cells remain quiescent and divide only to maintain a constant germ cell:escort cell ratio.  相似文献

Background  

Minor alleles of the human dopamine receptor polymorphisms, DRD2/TaqI A and DRD4/48 bp, are related to decreased functioning and/or numbers of their respective receptors and have been shown to be correlated with body mass, height and food craving. In addition, the 7R minor allele of the DRD4 gene is at a higher frequency in nomadic compared to sedentary populations. Here we examine polymorphisms in the DRD2 and DRD4 genes with respect to body mass index (BMI) and height among men in two populations of Ariaal pastoralists, one recently settled (n = 87) and the other still nomadic (n = 65). The Ariaal live in northern Kenya, are chronically undernourished and are divided socially among age-sets.  相似文献
