Plasma membrane phosphatidic acid phosphohydrolase (PAPH) plays an important role in signal transduction by converting phosphatidic acid to diacylglycerol. PAPH-2, a Mg2+-independent, detergent-dependent enzyme involved in cellular signal transduction, is reportedly absent from the plasma membranes of neutrophilic leukocytes, a cell that responds to metabolic stimulation with abundant phospholipase -dependent diacylglycerol generation. The present study was designed to resolve this discrepancy, focusing on the influence of cellular disruption techniques, detergenta availability and cation sensitivity on the apparent distribution of PAPH in neutrophil sub-cellular fractions. The results clearly indicate the presence of two distinct types of PAPH within the particulate and cytosolic fractions of disrupted cells. Unlike the cytosolic enzyme, the particulate enzyme was not potentiated by magnesium and was strongly detergent-dependent. The soluble and particulate enzymes displayed dissimilar pH profiles. Separation of neutrophil particulate material into fractions rich in plasma membranes, specific granules and azurophilic granules by high speed discontinuous density gradient centrifugation revealed that the majority of the particulate activity was confined to plasma membranes. This activity was not inhibited by pretreatment with n-ethyl-maleimide in concentrations as high as 25 mM. PAPH activity recovered in the cytosolic fraction of disrupted neutrophils was almost completely inhibited by 5.0 mM n-ethylmaleimide. We conclude that resting neutrophils possess n-ethylmaleimide-resistant PAPH (type 2) within their plasma membranes. This enzyme may markedly influence the kinetics of cell activation by metabolizing second messengers generated as a result of activation of plasma membrane phospholipase D.  相似文献

Most estimates of regional and global soil carbon stocks are based on extrapolations of mean soil C contents for broad categories of soil or vegetation types. Uncertainties exist in both the estimates of mean soil C contents and the area over which each mean should be extrapolated. Geographic information systems now permit spatially referenced estimates of soil C at finer scales of resolution than were previously practical. We compared estimates of total soil C stocks of the state of Maine using three methods: (1) multiplying the area of the state by published means of soil C for temperate forests and for Spodosols; (2) calculating areas of inclusions of soil taxa in the 1:5,000,000 FAO/UNESCO Soils Map of the World and multiplying those areas by selected mean carbon contents; and (3) calculating soil C for each soil series and map unit in the 1:250,000 State Soil Geographic Data Base (STATSGO) and summing these estimates for the entire state. The STATSGO estimate of total soil C was between 23% and 49% higher than the common coarse scale extrapolations, primarily because STATSGO included data on Histosols, which cover less than 5% of the area of the state, but which constitute over one-third of the soil C. Spodosols cover about 65% of the state, but contribute less than 39% of the soil C. Estimates of total soil C in Maine based on the FAO map agreed within 8% of the STATSGO estimate for one possible matching of FAO soil taxa with data on soil C, but another plausible matching overestimated soil C stocks. We also compared estimates from the 1:250,000 STATSGO database and from the 1:20,000 Soil Survey Geographic Data Base (SSURGO) for a 7.5 minute quadrangle within the state. SSURGO indicated 13% less total soil C than did STATSGO, largely because the attribute data on depths of soil horizons in SSURGO are more specific for this locality. Despite localized differences, the STATSGO database offers promise of scaling up county soil survey data to regional scales because it includes attribute data and estimates of areal coverage of C-rich inclusions within map units. The spatially referenced data also permit examination of covariation of soil C stocks with soil properties thought to affect stabilization of soil C. Clay content was a poor predictor of soil C in Maine, but drainage class covaried significantly with soil C across the state.  相似文献

RNA has been hypothesized to have preceded proteins as the major catalysts of the biosphere, yet there are only a very limited number of chemical reactions that are known to be catalyzed by modern RNA. Cofactors are used by the majority of protein enzymes to supply additional functional groups to the active site. RNA should also be able to utilize some of these same cofactors to extend its own catalytic potential. We describe here how it could be possible to use selection — amplification from a population of random RNA to obtain a coenzyme A mediated RNA transacylase. Exploitation of some of the sulphur chemistry mediated by coenzyme A could have significantly expanded a prebiotic RNA directed metabolism.  相似文献

Summary A method was developed for the introduction of plasmids into Clostridium botulinum by electroporation. A 4.4 kb plasmid vector, pGK12, which contains genes for resistance to erythromycin (Emr) and chloramphenicol (Cmr) was electroporated into C. botulinum type A (Hall A). The highest transformation efficiency was obtained using midlog phase cells, 10% PEG 8000 as the electroporation solution, and 2.5 kV field strength. The transformation efficiency was highest (103 transformants/g of DNA) when 1 g of plasmid DNA and 4 × 108 CFU/ml of recipient cells were used. Plasmid DNA recovered from the transformants was indistinguishable from that introduced on the basis of restriction enzyme digestion and agarose gel electrophoresis.  相似文献

In spinal cord injury, sexual reactions have a common dimension with ablebodied mankind, but also particular features corresponding to particular neurological syndromes. The third dimension which has to be taken into account is the singular one, every man, paraplegic or able-bodied, being unique. When the level of the lesion is above T10, genito-sexual responses to stimulations have a reflex functioning. In such cases, erotic tricks must be used to complete psycho-physiological reactions. On the contrary, technical devices are ill-tolerated both by the paraplegic man and by his spouse. Incomplete lesions in T12-L1 have to psychogenic responses to stimulations, i.e. partial erection and/or semen emission due to the neurophysiological mecanisms of desire. In the complete lesions of the conus medullaris or of the cauda equina, men become impotent and can no longer produce semen. In such cases, technical devices and drugs directly injected in the corpus cavernosum are technically efficacious but psychologically ill-tolerated. Facing his new sexual responses, the paraplegic man and his spouse have to adjuste themselves to the new circumstances. The way of adjustment is similar to a mourning process. One has to pay attention to let every paraplegic live every period of adjustment on his own, and at the same time one must be present to help him and answer his questioning.  相似文献

Abstract: We previously showed that gangliosides inhibit DNA synthesis in Swiss 3T3 cells stimulated with platelet-derived growth factor (PDGF) in a dose-responsive manner. This correlated with the inhibitory effects of several gangliosides (except GM3) on tyrosine phosphorylation of the PDGF receptor (PDGFR). [35S]Methionine-labeled Swiss 3T3 cells were incubated either with or without gangliosides and stimulated with PDGF, and proteins were cross-linked with bis(sulfosuccinimidyl) suberate. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed that two protein bands (170 and 350 kDa) were specifically immunoprecipitated with an anti-PDGFR antibody. Using both Swiss 3T3 and human glioma U-1242MG cells, western blots with anti-PDGFR and anti-phosphotyrosine antibodies confirmed that these bands were the PDGFR monomer and dimer, respectively, and that phosphotyrosine was present in these bands only after cells were stimulated with PDGF. Of the gangliosides tested, GM1, GM2, GD1a, GD1b, GD3, and GT1b, but not GM3, inhibited the formation of the 350-kDa band. These results demonstrate that all gangliosides tested, except GM3, probably inhibit PDGF-mediated growth by preventing dimerization of PDGFR monomers. Loss of more complex gangliosides in human gliomas would permit unregulated activation of the PDGFR, contributing to uncontrolled growth stimulation. We propose that ganglioside inhibition of receptor dimerization is a novel mechanism for regulating and coordinating several trophic factor-mediated cell functions.  相似文献

The ability of the marine microorganism Desulfuromonas acetoxidans to reduce Fe(III) was investigated because of its close phylogenetic relationship with the freshwater dissimilatory Fe(III) reducer Geobacter metallireducens. Washed cell suspensions of the type strain of D. acetoxidans reduced soluble Fe(III)-citrate and Fe(III) complexed with nitriloacetic acid. The c-type cytochrome(s) of D. acetoxidans was oxidized by Fe(III)-citrate and Mn(IV)-oxalate, as well as by two electron acceptors known to support growth, colloidal sulfur and malate. D. acetoxidans grew in defined anoxic, bicarbonate-buffered medium with acetate as the sole electron donor and poorly crystalline Fe(III) or Mn(IV) as the sole electron acceptor. Magnetite (Fe3O4) and siderite (FeCO3) were the major end products of Fe(III) reduction, whereas rhodochrosite (MnCO3) was the end product of Mn(IV) reduction. Ethanol, propanol, pyruvate, and butanol also served as electron donors for Fe(III) reduction. In contrast to D. acetoxidans, G. metallireducens could only grow in freshwater medium and it did not conserve energy to support growth from colloidal S0 reduction. D. acetoxidans is the first marine microorganism shown to conserve energy to support growth by coupling the complete oxidation of organic compounds to the reduction of Fe(III) or Mn(IV). Thus, D. acetoxidans provides a model enzymatic mechanism for Fe(III) or Mn(IV) oxidation of organic compounds in marine and estuarine sediments. These findings demonstrate that 16S rRNA phylogenetic analyses can suggest previously unrecognized metabolic capabilities of microorganisms.  相似文献

Mobilization of a genetically engineered IncQ plasmid, pSKTG, was studied in vitro and in sterile and nonsterile soils. In biparental and triparental filter matings, the mobilization frequencies of pSKTG were identical, and the plasmid was mobilized only in the presence of self-transmissible plasmid RP4p. In sterile soil, mobilization was probably limited by reduced cell-to-cell contact, since the frequencies of mobilization were approximately 100-fold lower than the frequencies in the filter matings. The transfer frequency of pSKTG in sterile soil when RP4p was present in the same strain was about 100-fold higher than the transfer frequency when RP4p was present in a separate strain. In studies in natural soil, pSKTG was also found to be transferred to indigenous bacteria. However, natural mobilization by genetic elements present in the indigenous soil microflora could not be detected. In vitro studies of natural transfer suggested that such genetic elements occur in soil bacteria.  相似文献
