The preimplantation embryo is highly resilient to experimental manipulations. A specific manipulation that has revealed many clues to the developmental process is chimera production. Chimeras have been used to describe the importance of developmental characteristics of embryonic cells and how these characteristics are involved with developmental fate. These characteristics have been monopolized in the production of interspecific chimeras and the production of transgenic animals. A simplified procedure for cDNA and genomic library construction using nonpalindromic oligonucleotide adaptors 总被引:2,自引:0,他引:2
The properties and characteristics of oligonucleotide adaptors for use in a simplified procedure for the construction of cDNA and genomic DNA libraries are described. The adaptors are suitable for joining to blunt ended cDNA or sheared genomic DNA, and then to the cohesive ends of restriction sites in vectors. Each adaptor consists of two oligonucleotides with complementary but nonpalindromic sequences that include an internal restriction site, a 5' phosphorylated blunt end, and an overlapping or staggered 5' hydroxylated end corresponding to a restriction endonuclease site in a vector of choice. Ligation of the blunt end to high molecular weight target DNA proceeds efficiently and there is no tandem concatenation of the adaptor. Insertion into the appropriate vector only requires ligation of the cohesive ends. There is no requirement for methylation, restriction enzyme cleavage, G-C tailing, or denaturation after ligation of the adaptor to the target DNA, all characteristics of other procedures. 相似文献

OBJECTIVES--To determine the symptomatic and urodynamic outcome of elective prostatectomy and to establish whether the outcome is influenced or can be predicted by preoperative urodynamic measurements. DESIGN--Prospective non-randomised study with follow up at a mean of 11 months after operation. Most men were assessed jointly by a urologist and a general practitioner. SETTING--Department of urology in a teaching hospital serving a large district population. PATIENTS--253 Men listed for elective prostatectomy because of symptoms and low urinary flow rates (less than 15 ml/s) and excluding those already on a waiting list or with acute urinary retention, clinically apparent prostatic cancer, and neurological or cerebrovascular disease; 217 (86%) were followed up. INTERVENTION--Elective prostatectomy. MAIN OUTCOME MEASURE--Classification on the basis of relief of symptoms assessed by patients and urologist and general practitioner and of symptom scores obtained by questionnaire. RESULTS--Of the 217 men followed up, 171 (79%) had a satisfactory subjective review and 155 (72%) had a satisfactory review and also low symptom scores. An unsatisfactory outcome was associated with preoperative symptoms of urge incontinence, small prostatic size and resected weight, low voiding pressures, and low urethral resistance. Preoperative maximum urinary flow rates did not predict outcome. Men with poor outcome could be classified into two groups: those with irritative symptoms who were more likely before operation to have had urge incontinence and detrusor instability and men with symptoms of poor urinary flow who were more likely before operation to have had a small prostate, low voiding pressures, and low urethral resistance. In patients in the second group flow rates or voiding pressures improved little after operation. Men with stable detrusors and either low urethral resistance or low voiding pressures were less likely to do well after prostatectomy, but despite these associations preoperative urodynamic measurements were unable to predict outcome accurately. CONCLUSIONS--Prostatectomy was satisfactory in relieving symptoms and improving urodynamic measurements in most men, but even in those with classic symptoms and low urinary flow rates a substantial minority experienced little improvement afterwards and urodynamic measurements did not accurately predict outcome in individual patients. 相似文献

Conclusion It should be clear by now the extent to which many features of Thorpe's interpretation of animal behavior and of the animal mind rested, at bottom, not simply on conventional scientific proofs but on interpretive inferences, which in turn rested on a willingress to make extensions of human experience to animals. This, in turn, rested on his view of evolution and his view of reality. And these were governed by his natural theology, which was the fundamental stratum of his intellectual experience.Contrary to the scientific ethos, which restricts theory choice to scientific issues alone, Thorpe's career suggests that the actual reasons for theory choice among scientists often are not limited to science, but are multiple and may sometimes be difficult to discover. It is largely because Thorpe took a public part in the natural theology enterprise that we can know something about his religious beliefs and so can see their probable influence on his scientific decisions. Similar beliefs of other scientists are sometimes harder to get at. Most may be practically beyond discovery, for the ethos of science has discouraged public professions of personal belief in relation to scientific work.101 Yet does it seem plausible that, for example, the restriction of self-consciousness to humans by some scientists is a purely scientific decision?102 Surely not, any more than that the strong influence of natural theology on Thorpe's thought means that he was not a good scientist. His natural theology may have led him into incautious enthusiasms regarding the animal mind — such as the potential if unrealizable linguistic ability of chimpanzees — through a bias in favor of the continuity of emergents in a progressive evolutionary system, just as it led him to advocate animal consciousness long before the recent upsurge of interest, but the scientific integrity of his work overall is unimpeachable. And yet, that work is not comprehensible historically as science alone. Personal philosophy must not be discounted in writing the history of recent science. This somewhat obvious conclusion (obvious to historians of science) needs emphasis, for we are still prone to think that the sciences of our own time provide their own internal dynamic that is in itself sufficient to account for their content and development. 相似文献

Longosu-chus is characterised by the possession of seven dentary teeth, a dentary excluded from the ventral margin of the lateral mandibular fenestra and by the angular, spinose lateral scutes throughout the dorsal region with lateral horns which have faceted sides.Longosuchus is found in mid-late Carnian strata in Texas, New Mexico and North Carolina, USA. Summary Stable transformed Black Mexican Sweet (BMS) maize callus was recovered from suspension culture cells bombarded with plasmid DNA that conferred resistance to the herbicide bialaphos. Suspension culture cells were bombarded with a mixture of two plasmids. One plasmid contained a selectable marker gene, bar, which encoded phosphinothricin acetyl transferase (PAT), and the other plasmid encoded a screenable marker for -glucuronidase (GUS). Bombarded cells were selected on medium containing the herbicide bialaphos, which is cleaved in plant cells to yield phosphinothricin (PPT), an inhibitor of glutamine synthetase. The bialaphos-resistant callus contained the bar gene and expressed PAT as assayed by PPT inactivation. Transformants that expressed high levels of PAT grew more rapidly on increasing concentrations of bialaphos than transformants expressing low levels of PAT. Fifty percent of the bialaphos-resistant transformants tested (8 of 16) expressed the nonselected gene encoding GUS. 相似文献

The uv-visible spectra of 7,8-didemethyl-8-hydroxy-5-deazaflavin-5'-phosphoryllactyl glutamate (coenzyme F420), a naturally occurring 5-deazaflavin derivative, in three different buffers changed with a rise in temperature; the effect on the extinction coefficient at 420 nm (epsilon 420) was as follows: In phosphate-buffered solutions at pH less than 7.5, the epsilon 420 increased (at pH 5.0 for a temperature shift from 15 to 60 degrees C, delta epsilon 420 was +87%), but between pH 7.5 and 8, epsilon 420 changed very little. At pH greater than 8.0 in phosphate- or borate-buffered solutions, epsilon 420 decreased slightly. In morpholineethanesulfonic acid (Mes)-buffered F420 solutions at pH 5 and 5.5, epsilon 420 changed very little, whereas at pH 6-8, the epsilon 420 decreased. Absorbance of F420 at 401 nm in phosphate buffer at pH 5 to 9 was not significantly affected by temperature. The cis/trans isomerization of the peptide bond preceding proline residues in proteins can limit the rate at which a protein folds to its native conformation. Mutagenic analyses of dihydrofolate reductase (DHFR) from Escherichia coli show that this isomerization reaction can be intramolecularly catalyzed by a side chain from an amino acid which is distant in sequence but adjacent in the native conformation. The guanidinium NH2 nitrogen of Arg 44 forms one hydrogen bond to the imide nitrogen and a second to the carbonyl oxygen of Pro 66 in wild-type DHFR. Replacement of Arg 44 with Leu results in a change of the nature of the two slow steps in refolding from being limited by the acquisition of secondary and/or tertiary structure to being limited by isomerization. The simultaneous replacement of Pro 66 with Ala (i.e., the Leu 44/Ala 66 double mutant) eliminates this isomerization reaction and once again makes protein folding the limiting process. Apparently, one or both of the hydrogen bonds between Arg 44 and Pro 66 accelerate the isomerization of the Gln 65-Pro 66 peptide bond. The replacement of Arg 44 with Leu affects the kinetics of the slow folding reactions in a fashion which indicates that the crucial hydrogen bonds form in the transition states for the rate-limiting steps in folding. 相似文献

Oviposition by the diamondback moth, Plutella xylostella (Lepidoptera: Plutellidae), on substrates treated with host stimuli (cabbage homogenate or sinigrin) and/or waxes (paraffin or a mixture of 10 single chain n-alkanes) was quantified using continuous observations and endpoint bioassays. Paraffin or an n-alkane mixture applied over cabbage homogenate or sinigrin caused an increase in oviposition compared to that on any single stimulus in choice tests. Sinigrin alone at 10–5 M to 10–2 M is an ovipositional stimulant; addition of alkane over sinigrin made all sinigrin concentrations (10–6 M to 10–2 M) significantly more stimulatory than controls. Waxes alone do not stimulate oviposition. In choice tests, insect movement between sinigrin/alkane treatment combinations was random, however, once encountered, visit duration was significantly longer on sinigrin with alkane than on sites treated with either stimulus alone. Given the ubiquity of waxes on plant surfaces and the interaction between waxes and host-specific chemical stimuli, waxes should be included when considering factors that significantly influence herbivore host acceptance. 相似文献

We propose a pair of population genetic models for a modifier-of-imprinting locus for which different genotypes imprint different proportions of an imprintable target locus in their gametes. The two models examine the situations in which imprinting is advantageous or disadvantageous, and we discuss three cases for which the modifier is respectively partially dominant, dominant, or recessive. The models predict the stable equilibrium frequencies of the mutant modifier and functionally diploid individuals in a large population in terms of up to four parameters: the mutation rate at the modifier locus, V; the selection coefficient against the disadvantageous phenotype, s; the proportion of unimprinted eggs produced by homozygotes for the mutant modifier, θ, and, in the partially dominant models, the dominance parameter, k. The equilibrium frequency of the mutant phenotypes is shown to be approximately twice that of standard Mendelian models: 2V/s or 4V/s when the modifier is recessive or dominant, respectively. Mathematical equivalences between these and nonimprinting models are noted. 相似文献
