The EGF receptor in waved-2 mice contains a point mutation that renders it kinase-deficient. We investigated how the waved-2 mutation affects the internalization and endosomal trafficking of the receptor in vivo in response to EGF. When the waved-2 mice were injected with EGF, there was approximately 50% less tyrosine phosphorylation detected in whole-liver homogenate compared to wild-type background mice. Although EGF increased the EGF receptor levels in the early and late liver endosomal fractions of waved-2 mice, its trafficking was delayed compared to wild-type mice. Ubiquitination of the EGF receptor may affect its endosomal sorting. We found that a similar amount of EGF receptor was immunoprecipitated from the endosomal fractions of EGF-treated waved-2 and wild-type with anti-ubiquitin antibody. These results demonstrate that the waved-2 EGF receptor can become ubiquitinated and can be trafficked to the late endosomes, although it appears that its kinase deficiency delays this process.  相似文献

Sexual selection is thought to be a powerful diversifying force, based on large ornamental differences between sexually dimorphic species. This assumes that unornamented phenotypes represent evolution without sexual selection. If sexual selection is more powerful than other forms of selection, then two effects would be: rapid divergence of sexually selected traits and a correlation between these divergence rates and variance in mating success in the ornamented sex. I tested for these effects in grouse (Tetraonidae). For three species pairs, within and among polygynous clades, male courtship characters had significantly greater divergence than other characters. This was most pronounced for two species in Tympanuchus. In the Eurasian polygynous clade, relative courtship divergence gradually increased with nucleotide divergence, suggesting a less dramatic acceleration. Increase in relative courtship divergence was associated with mating systems having higher variance in male mating success. These results suggest that sexual selection has accelerated courtship evolution among grouse, although the microevolutionary details appear to vary among clades.  相似文献

Female rats were subjected to superior cervical ganglionectomy (Gx), blinding and anosmia (BAs) or combined procedures (BAsGx). Onset and growth of dimethylbenz(a)anthracene (DBMA)-induced mammary tumors was studied in these animals and compared to tumorigenesis in intact control rats. Carcinostatic effects were present in all surgically altered animals, as evidenced by a trend toward reduced tumor incidence, reduced final tumor mass, and a significant reduction in mean number of tumors in Gx and BAsGx rats, and increased regression of tumors in BAs rats compared to intact group. Reduced tumorigenesis was paralleled by a trend toward either an increase (BAs) or a decrease (Gx and BAsGx) in the activity of pineal hydroxyindole-O-methyltransferase (HIOMT) compared to intact group. In addition, BAs and BAsGx animals showed a significant reduction in body weight. These results suggest that Gx reduces mammary tumorigenesis in both sighted and BAs rats. They further confirm the findings of others on reduced mammary tumorigenesis in BAs rats. Possible involvement of multiple carcinostatic mechanisms in different animal models is discussed.  相似文献

Alpine lakes receive a large fraction of their nutrients from atmospheric sources and are consequently sensitive to variations in both the amount and chemistry of atmospheric deposition. In this study we explored the spatial changes in lake water chemistry and biology along a gradient of dust deposition in the Wind River Range, Wyoming. Regional differences were explored using the variation in bulk deposition, lake water, sediment, and bedrock geochemistry and catchment characteristics. Dust deposition rates in the Southwestern region averaged 3.34 g m?2 year?1, approximately three times higher than deposition rates in the Northwestern region (average 1.06 g m?2 year?1). Dust-P deposition rates ranged from 87 µg P m2 day?1 in the Northwestern region to 276 µg P m2 day?1 in the Southwestern region. Subalpine and alpine lakes in the Southwestern region had greater total phosphorus (TP) concentrations (5–13 µg L?1) and greater sediment phosphorus (SP) concentrations (2–5 mg g?1) than similar lakes elsewhere in the region (1–8 µg L?1 TP, 0.5–2 mg g?1 SP). Lake phosphorus concentrations were related to dissolved organic carbon (DOC) across vegetation gradients, but related to the percent of bare rock, catchment area to lake area, and catchment steepness across dust deposition gradients. Modern phytoplankton and zooplankton biomasses were two orders of magnitude greater in the Southwest than in the Northwest, and alpine lakes in the Southwest had a unique diatom species assemblage with relatively higher concentrations of Asterionella formosa, Pseudostaurosira pseudoconstruens, and Pseudostaurosira brevistriata. These results suggests that catchment controls on P export to lakes (i.e. DOC) are overridden in dominantly bare rock basins where poor soils cannot effectively retain dust deposited P.  相似文献

In a paper by Zimmermann and colleagues in this issue of Arthritis Research & Therapy, results of extended laboratory research with the drug combination of prednisolone and dipyridamole are reported. There seems to be a boost and extension of the glucocorticoid effect by the combination, without a clear increase of adverse effects, potentially allowing the application of lower dosages. However, laboratory models are not patients and the glucocorticoid mechanisms leading to effects and adverse effects are manifold. The next required step will be to demonstrate the improved therapeutic window in patients in adequate comparative clinical trials, assessing predefined beneficial effects and adverse effects in a standardized way.  相似文献

Microgravity offers unique advantages for the cultivation of mammalian tissues because the lack of gravity-induced sedimentation supports three-dimensional growth in batch culture in aqueous medium. Bioreactors that simulate microgravity but operate in unit gravity provide conditions that permit human epithelial cells to grow to densities approaching 107 cells/ml on microcarriers in suspension, in masses up to 1 cm in diameter, and under conditions of low shear stress. While useful for many different applications in tissue culture, this culture system is especially useful for the analysis of the microenvironment in which host matrix and cells interact with infiltrating tumor cells. Growth in the microgravity-based bioreactor has supported morphological differentiation of human colon carcinoma cells when cultured with normal human stromal cells. Furthermore, these co-cultures produced factors that stimulated goblet cell production in normal colon cells in an in vivo bioassay. Early experiments also suggest that the microgravity environment will not alter the ability of epithelial cells to recognize and associate with each other and with constituents of basement membrane and extracellular matrix. These findings suggest that cells grown in bioreactors that simulate aspects of microgravity or under actual microgravity conditions will produce tissues and substances in sufficient quantity and at high enough concentration to promote characterization of molecules that control differentiation and neoplastic transformation. © 1993 Wiley-Liss, Inc.  相似文献
