Female meiosis in the onion fly and cabbage root fly (Diptera: Anthomyiidae).

The sequence of female meiosis was investigated in two populations of the cabbage root fly (Delia radicum) and three populations of the onion fly (D. antiqua). In contrast with the completely achiasmate males, both species showed high levels of recombination in females. However, significant differences in chiasma frequency occurred between individuals within populations and between the populations. It was not uncommon to find aneuploidy of the X chromosomes. The autosomes occasionally showed asynapsis or desynapsis, but normal disjunction of univalents was facilitated by distance pairing.     

Determining the influence of structure on hybridization using oligonucleotide arrays.

We have studied the effects of structure on nucleic acid heteroduplex formation by analyzing hybridization of tRNAphe to a complete set of complementary oligonucleotides, ranging from single nucleotides to dodecanucleotides. The analysis points to features in tRNA that determine heteroduplex yield. All heteroduplexes that give high yield include both double-stranded stems as well as single-stranded regions. Bases in the single-stranded regions are stacked onto the stems, and heteroduplexes terminate at potential interfaces for coaxial stacking. Heteroduplex formation is disfavored by sharp turns or a lack of helical order in single-stranded regions, competition from bases displaced from a stem, and stable tertiary interactions. The study is relevant to duplex formation on oligonucleotide microarrays and to antisense technologies.     

Biology of cloned cytotoxic T lymphocytes specific for lymphocytic choriomeningitis virus. V. Recognition is restricted to gene products encoded by the viral S RNA segment

Lymphocytic choriomeningitis virus (LCMV) Armstrong (ARM) strain-specific, H-2d-restricted CTL effectively lyse syngeneic targets infected by LCMV ARM, but show reduced killing of LCMV Pasteur (PAST) strain-infected H-2d cells. We have reassorted the two RNA segments, large (L) and small (S), of LCMV ARM and PAST to generate LCMV with genotypes of L ARM/S PAST and L PAST/S ARM. By using these reassortants and both LCMV primary CTL and CTL clones, we report that the induction, recognition, and lysis of LCMV-specific CTL depend on the S RNA segment and the genes it encodes.     

Comparison of the Pulmonary Bactericidal Capacity of Mice and Rats Against Strains of Pseudomonas aeruginosa

After aerosol deposition of Pseudomonas aeruginosa strains in mice and rats, completely different patterns of pulmonary bacterial clearance were observed in each animal species.
