Aberrant Nuclear Factor-κappaB (NF-κB) activation due to rapid IκBα turnover and high basal IκBα kinase (IKK) activity has been frequently observed in prostate cancer. Apigenin, a naturally occurring plant flavone, exhibits anti-proliferative, anti-inflammatory and anti-carcinogenic activities by inhibiting NF-κB pathway, through a mechanism not fully understood. We found that apigenin feeding in microgram doses (bioavailable in humans) inhibited prostate tumorigenesis in TRAMP mice by interfering with NF-κB signaling. Apigenin feeding to TRAMP mice (20 and 50 μg/mouse/day, 6 days/week for 20 weeks) exhibited significant decrease in tumor volumes of the prostate and completely abolished metastasis, which correlated with inhibition of NF-κB activation and binding to the DNA. Apigenin intake blocked phosphorylation and degradation of IκBα by inhibiting IKK activation, which in turn led to suppression of NF-κB activation. The expression of NF-κB-regulated gene products involved in proliferation (cyclin D1, and COX-2), anti-apoptosis (Bcl-2 and Bcl-xL), and angiogenesis (vascular endothelial growth factor) were also downregulated after apigenin feeding. These events correlated with the induction of apoptosis in tumor cells, as evident by increased cleaved caspase-3 labeling index in the dorsolateral prostate. Our results provide convincing evidence that apigenin inhibits IKK activation and restores the expression of IκBα, preventing it’s phosphorylation in a fashion similar to that elicited by IKK and proteasomal inhibitors through suppression of NF-κB signaling pathway. 相似文献

Brucella species include important zoonotic pathogens that have a substantial impact on both agriculture and human health throughout the world. Brucellae are thought of as “stealth pathogens” that escape recognition by the host innate immune response, modulate the acquired immune response, and evade intracellular destruction. We analyzed the genome sequences of members of the family Brucellaceae to assess its evolutionary history from likely free-living soil-based progenitors into highly successful intracellular pathogens. Phylogenetic analysis split the genus into two groups: recently identified and early-dividing “atypical” strains and a highly conserved “classical” core clade containing the major pathogenic species. Lateral gene transfer events brought unique genomic regions into Brucella that differentiated them from Ochrobactrum and allowed the stepwise acquisition of virulence factors that include a type IV secretion system, a perosamine-based O antigen, and systems for sequestering metal ions that are absent in progenitors. Subsequent radiation within the core Brucella resulted in lineages that appear to have evolved within their preferred mammalian hosts, restricting their virulence to become stealth pathogens capable of causing long-term chronic infections. 相似文献

Japanese encephalitis (JE) is one of the leading causes of acute encephalopathy affecting children and adolescents in the
tropics. Optimization of media was carried out for enhanced production of recombinant JE virus envelope domain III (EDIII)
protein in Escherichia coli. Furthermore, batch and fed-batch cultivation process in E. coli was also developed in optimized medium. Expression of this protein in E. coli was induced with 1 mM isopropyl-β-thiogalactoside and yielded an insoluble protein aggregating to form inclusion bodies.
The inclusion bodies were solubilized in 8 M urea, and the protein was purified under denaturing conditions using Ni-NTA affinity
chromatography. After fed-batch cultivation, the recombinant E. coli resulted in cell dry weight and purified protein about 36.45 g l−1 and 720 mg l−1 of culture, respectively. The purity of the recombinant JE virus EDIII protein was checked by sodium dodecyl sulfate polyacrylamide
gel electrophoresis analysis, and reactivity of this protein was determined by Western blotting and ELISA with JE virus-infected
human serum samples. These results establish the application of this protein to be used for the diagnosis of JE virus infection
or for further studies in vaccine development. This process may also be suitable for the high-yield production of other recombinant

Self-organization is a key condition to the success of community-based conservation initiatives, including those recognized
by the Equator Initiative of the UNDP. This paper contributes to emerging scholarship that focuses on community-based conservation
in South Asia and in particular examines self-organization strategies within a small-scale community-based conservation initiative
in a cross-cultural setting to further understanding about how such initiatives originate, sustain and grow. This is achieved
through a case study of the Baripada Forest Protection Initiative in India by utilizing in-depth interviews and focus groups.
In addition to certain often-cited strategies for self-organization, the Baripada initiative included unique features of self-organization
such as village community design, implementation and adaptation of rules for local natural resources use and conservation,
little need for financial support, and significant mobilization of human resources. These strategies, along with emerging
social learning opportunities (e.g., a community plant diversity register) inspired by the Baripada initiative, inform and
enrich the criteria for designing and evaluating conservation and development initiatives, irrespective of their scale. 相似文献

Background
Subcutaneous zygomycosis is a chronic infection caused by fungus of the order Entomophthorales. It can have varying presentations and presents in the nose and face area with gradually progressing subcutaneous swelling that may be difficult to diagnose unless a strong suspicion of fungal involvement is maintained. We present a case of subcutaneous zygomycosis in a 35-year-old male patient, resident of a North Indian state. The patient was diagnosed to be suffering from subcutaneous zygomycosis, the causative agent being Basidiobolus ranarum identified on culture and lactophenol cotton blue mount preparation. Unusual fungal agents that exist environmentally as saprophytes can often lead to opportunistic infections. Hyalohyphomycosis
is a group of fungal infections caused by fungi characterized by hyaline septate hyphae and can infect both immunocompetent
as well as immunocompromised patients. Many a times it becomes difficult to distinguish a pathogenic and a contaminant fungus,
because many such agents can assume clinical significance depending on circumstances. Subcutaneous and invasive fungal infection
due to the emerging hyalohyphomycotic fungus, Acremonium, has drawn the attention of clinicians and microbiologists, as a potential pathogen in patients with and without underlying
risk factors. Generally considered to be minimally invasive in the past, genus Acremonium has been responsible for eumycotic mycetomas and focal infections in otherwise healthy individuals. It has also been increasingly
implicated in systemic fungal diseases. The management with different antifungals in various clinical situations has been

Chikungunya is one of the most important emerging arboviral infections of public health significance. Due to lack of a licensed
vaccine, rapid diagnosis plays an important role in early management of patients. In this study, a QC-RT–PCR assay was developed
to quantify Chikungunya virus (CHIKV) RNA by targeting the conserved region of E1 gene. A competitor molecule containing an
internal insertion was generated, which provided a stringent control of the quantification process. The introduction of 10-fold
serially diluted competitor in each reaction was further used to determine sensitivity. The applicability of this assay for
quantification of CHIKV RNA was evaluated with human clinical samples, and the results were compared with real-time quantitative
RT–PCR. The sensitivity of this assay was estimated to be 100 RNA copies per reaction with a dynamic detection range of 102 to 1010 copies. Specificity was confirmed using closely related alpha and flaviviruses. The comparison of QC-RT–PCR result with real-time
RT–PCR revealed 100% concordance for the detection of CHIKV in clinical samples. These findings demonstrated that the reported
assay is convenient, sensitive and accurate method and has the potential usefulness for clinical diagnosis due to simultaneous

AimsThe aim of the present study is to investigate the effect of curcumin on cerebral blood flow (CBF), memory impairment, oxidative stress and cholinergic dysfunction in intracerebral (IC) streptozotocin (STZ) induced memory impairment in mice.Main methodsMemory impairment was induced by STZ (0.5 mg/kg, IC) administered twice with an interval of 48 h in mice. Memory function was assessed by Morris water maze and passive avoidance test. CBF was measured by Laser Doppler Flowmetry (LDF). To study the preventive effect, curcumin (10, 20 and 50 mg/kg, PO) was administered for 21 days starting from the first dose of STZ. In another set of experiment, curcumin was administered for 7 days from 19th day after confirming STZ induced dementia to observe its therapeutic effect. Biochemical parameters of oxidative stress and cholinergic function were estimated in brain on day 21.Key findingsThe major finding of this study is that STZ (IC) caused a significant reduction in CBF along with memory impairment, cholinergic dysfunction and enhanced oxidative stress. Curcumin dose dependently improved CBF in STZ treated mice together with amelioration of memory impairment both in preventive and therapeutic manner.SignificanceThe present study clearly demonstrates the beneficial effects of curcumin, the dietary staple of India, on CBF, memory and oxidative stress which can be exploited for dementia associated with age related vascular and neurodegenerative disorders. 相似文献
