Anionic vesicles containing acidic phospholipids are known complement activators. To clarify which negative physicochemical electrostatic charges on vesicles and structural specificities of acidic lipids are critical to complement activation, the electrostatic properties and activity to complement of two anionic vesicles modified with a carboxylic acid derivative or a conventional acidic phospholipid were compared. Electrophoretic mobility measurements indicated that the negative zeta potential and the electrostatic interactivity of these two anionic vesicles were equal at pH 7.4. However, the infusion of vesicles containing acidic phospholipid induced significant complement activation, while vesicles containing the carboxylic acid derivative failed to activate complement. These results indicate that the negative charge on the surface of vesicles is not critical for the activation complement, suggesting that complement activation is specific to the structure of acidic groups. This finding is likely to be important to the design of anionic biointerfaces and may support the promising medical applications of this anionic vesicle modified with a carboxylic acid derivative.  相似文献

19F NMR has recently emerged as an efficient, sensitive tool for analyzing protein binding to small molecules, and surface plasmon resonance (SPR) is also a popular tool for this purpose. Herein a combination of 19F NMR and SPR was used to find novel binders to the ATP-binding pocket of MAP kinase extracellular regulated kinase 2 (ERK2) by fragment screening with an original fluorinated-fragment library. The 19F NMR screening yielded a high primary hit rate of binders to the ERK2 ATP-binding pocket compared with the rate for the SPR screening. Hit compounds were evaluated and categorized according to their ability to bind to different binding sites in the ATP-binding pocket. The binding manner was characterized by using isothermal titration calorimetry and docking simulation. Combining 19F NMR with other biophysical methods allows the identification of multiple types of hit compounds, thereby increasing opportunities for drug design using preferred fragments.  相似文献

Many structures and molecules closely related to those involved in the specific process of immunoglobulin (Ig) hypermutation existed before the appearance of primordial Ig genes. Consequently, these structures can be found even in animals and organisms distinct from vertebrates; likewise, homologues of hypermutation enzymes are present in a broad range of species, from bacteria to mammals. Our analysis, based predominantly on primary structure, demonstrates the existence of molecules similar to Ig domains, variable Ig domains (IGv), and antigen receptors (AR) in unicellular organisms, nonvertebrate metazoans, and nonvertebrate Coelomata, respectively. In addition, we deal here with some important structural properties of CDR1-like segments of the selected sponge adhesion molecule GCSAMS exhibiting chimerical Ig domain similarities, and demonstrate the occurrence of conserved regions corresponding to Ohno's modern intact primordial building block in the C-terminal part of IGv-related segments of nonvertebrate origin. The results of our analysis are also discussed with respect to the possible phylogeny of molecules preceding the hypothetical common antigen receptor ancestor.  相似文献

The paper deals with the health status, production and structure of autochthonous beech stands in the eastern part of the Krkonoše Mts. (Czech Republic). The region was affected by increased air pollution load due to long-range SO2 transmission from the late 1970s to the early 1990s. In 1980, five research plots were established in long unmanaged beech stands at an optimum stage. Tree coordinates and initial biometrical measurements were made in 1980 and repeated in 2005. The defoliation of individual trees was monitored every year (ICP Forests principles). The actual stand development is compared with the model development worked out by the Sibyla growth simulator in variants with a model of natural mortality and with entering a real mortality of trees. Differences between the stand characteristics of actual and simulated development of forest stands suggest some production losses. The reason to the losses can be seen in the long-term impact of air pollution in the first decade of monitoring and its after-effects on soil quality and worsening health condition of stands. The anticipated acceleration in the onset of the phase of stand disintegration under the influence of air pollution load was not demonstrated.  相似文献

The authors investigated hourly total plasma tryptophan concentrations over a 24 hour period in 3 patients with bipolar depression during 2 types of treatment: a phase advance process and conventional antidepressant therapy. Both treatment modalities increased the 24 hour mean tryptophan levels and the amplitude of circadian tryptophan concentrations. By contrast, in a previous study, moderately decreased tryptophan levels and a blunted amplitude in the circadian rhythm were observed in these same patients in a depressive phase.  相似文献

The inactivation of four micromycete species by action of non-thermal plasma was followed. Two sources of plasma were compared, namely, positive corona discharge and dielectric barrier discharge. The corona discharge appeared as suitable for fungal spore inactivation in water suspension, whereas the barrier discharge inactivated spores on the surface of cultivation agar. Cladosporium sphaerospermum was the most sensitive, being inactivated within 10 min of exposure to plasma, whereas Aspergillus oryzae displayed decrease in viable cell count only, the complete inactivation was not achieved even after 40 min of exposure. Intermediate sensitivity was found for Alternaria sp. and Byssochlamys nivea. The significant delay of growth was observed for all fungi after exposure to sublethal dose of plasma, but we failed to express this effect quantitatively.  相似文献

Five days of suitable continuous light induced flowering in the majority ofChenopodium murale L. ecotype 197 plants as early as at the phase of the first pair of leaves. At the time of initiation of the 2nd to 4th pairs of leaves the capacity of plants to flower was reduced, the number of flowering plants being significantly lower under the same inductive light treatment. The capacity to flower increased again at the phase of the 5th and the 6th pairs of leaves. Inductive light treatment brought about a marked growth activation of organs present before induction, shoot apex elongation, precocious formation of new leaves and activation of axillary meristems. The course of these changes in plants of different age is demonstrated. The terminal flower developed during 5 short days following inductive light treatment. The paper shows similarities and differences between long-daymutale L. ecotype 197 and short-day C.rubrum L. ecotype 374 grown under practically uniform conditions.  相似文献

A method of cultivation and effectiveness of different light sources and light regimes in photoperiodic induction of flowering in non-rosette long-day plantChenopodium murale L. ecotype 197 are described. Under the described conditions of cultivation 5 days, of continuous light produced by incandescent bulbs (TESLA 74 3x40 W, red 4.9 μWcm-2nn-1, far-red 7.4 μWcn-2nm-1, blue 0.25 μW cm-2nn-1) induced flowering in the majority of plants.  相似文献
