In this work we unmask a novel downregulation mechanism of the uric acid/xanthine transporter UapA, the prototype member of the ubiquitous Nucleobase-Ascorbate Transporter family, directly related to its function. In the presence of substrates, UapA is endocytosed, sorted into the multivesicular body pathway and degraded in vacuoles. Substrate-induced endocytosis, unlike ammonium-induced turnover, is absolutely dependent on UapA activity and several lines of evidence showed that the signal for increased endocytosis is the actual translocation of substrates through the UapA protein. The use of several UapA functional mutants with altered kinetics and specificity has further shown that transport-dependent UapA endocytosis occurs through a mechanism, which senses subtle conformational changes associated with the transport cycle. We also show that distinct mechanisms of UapA endocytosis necessitate ubiquitination of a single Lys residue (K572) by HulARsp5. Finally, we demonstrate that in the presence of substrates, non-functional UapA versions can be endocytosed in trans if expressed in the simultaneous presence of active UapA versions, even if the latter cannot be endocytosed themselves.  相似文献

When comparing the causal effect of peritoneal dialysis (PD) and hemodialysis (HD) treatment on lowering mortality in renal patients, using observational data, it is necessary to adjust for different forms of confounding and informative censoring. Both the type of dialysis treatment that is started with and mortality are affected by baseline covariates. Longitudinal and baseline variables can affect both the probability of switching from one type of dialysis to the other, and mortality. Longitudinal and baseline variables can also affect the probability of receiving a kidney transplant, possibly causing informative censoring. Adjusting for longitudinal variables by including them as covariates in a regression model potentially causes bias, for instance by losing a possible indirect effect of dialysis on mortality via these longitudinal variables. Instead, we fitted a marginal structural model (MSM) to estimate the causal effect of dialysis type, adjusted for confounding and informative censoring. We used the MSM to compare the hazard of death as well as cumulative survival between the potential treatment trajectories "always PD" and "always HD" over time, conditional on age and diabetes mellitus status. We used inverse probability weighting (IPW) to fit the MSM.  相似文献

Hypoxia and acidosis are common features of the tumor microenvironment. Under hypoxic conditions increased lactic acid secretion together with carbonic acid production contributes to a high acid load in the tumor. Multiple isoforms of carbonic anhydrase (CA) (EC 4.2.1.1) are expressed in normal and neoplastic tissues. The expression of membrane-bound CAs, such as CA IX and CA XII is increased in hypoxic tumors and is tightly controlled by oxygen levels via the hypoxia-inducible factor (HIF). In this article we have reviewed recent findings that implicate the hypoxia-inducible CAs in pH homeostasis and in controlling cell viability and tumor growth. We then presented some preliminary, original in vitro data that point to the implication of hypoxia-inducible CAs in cell migration. In addition, we reviewed the clinical data that exploit the immunohistochemical detection of the hypoxia-inducible CAs as a marker for patient prognosis in different types of cancer and finally, we mentioned briefly approaches to therapeutic inhibition of CA.  相似文献

Nonaspanins constitute a family of proteins, also called TM9SF, characterized by a large non-cytoplasmic domain and nine putative transmembrane domains. This family is highly conserved through evolution and comprises three members in Saccharomyces cerevisiae, Dictyostelium discoideum, and Drosophila melanogaster, and four members are reported in mammals (TM9SF1–TM9SF4). Genetic studies in Dictyostelium and Drosophila have shown that TM9SF members are required for adhesion and phagocytosis in innate immune response, furthermore, human TM9SF1 plays a role in the regulation of autophagy and human TM9SF4 in tumor cannibalism. Here we report that the zebrafish genome encodes five members of this family, TM9SF1–TM9SF5, which show high level of sequence conservation with the previously reported members. Expression analysis in zebrafish showed that all members are maternally expressed and continue to be present throughout embryogenesis to adults. Gene expression could not be regulated by pathogen-associated molecular patterns such as LPS, CpG, or Poly I:C. By bioinformatic analyses of 80 TM9SF protein sequences from yeast, plants, and animals, we confirmed a very conserved protein structure. An evolutionary conserved immunoreceptor tyrosine-based inhibition motif has been detected in the cytoplasmic domain between transmembrane domain (TM) 7 and TM8 in TM9SF1, TM9SF2, TM9SF4 and TM9SF5, and at the extreme C-terminal end of TM9SF4. Finally, a conserved TRAF2 binding domain could also be predicted in the cytoplasmic regions of TM9SF2, TM9SF3, TM9SF4, and TM9SF5. This confirms the hypothesis that TM9SF proteins may play a regulatory role in a specific and ancient cellular mechanism that is involved in innate immunity.  相似文献

Background

Alleles of apolipoprotein E (APOE) are the major genetic risk factor for late onset Alzheimer's Disease (LOAD). Recently, an APOE splice variant that retains intron 3 (APOE-I3) was identified. To gain insight into the possible role of this isoform in LOAD, we quantified its expression in a cohort of 56 human brain specimens by using quantitative RT-PCR.

Results

We found that APOE-I3 generally represents a low percentage (< 0.5%) of overall APOE expression. However, in one specimen, the proportion of APOE-I3 was increased about ~13 fold. This specimen was unique in the cohort for possessing the minor allele of an intron 3 single nucleotide polymorphism (SNP), rs12982192. Additionally, an allelic expression imbalance study indicated that the rs12982192 minor allele was associated with increased APOE-I3 expression.

Conclusions

Overall, we interpret our results as suggesting that APOE-I3 represents a minor portion of APOE expression and that rs12982192 is associated with APOE intron 3 retention. Since the minor allele of this SNP is on the same haplotype as the minor allele of rs429358, which defines the APOE4 allele, we speculate that rs12982192 may reflect a modest loss of mRNA encoding functional APOE4.  相似文献

The Indian mock strawberry [Duchesnea indica (Andrews) Focke] is an invasive plant in several regions of central Europe and Germany. In order to explore its competitive ability, we compared it with the native woodland strawberry (Fragaria vesca L.) by growing it alone as well as in intra- or inter-specific competition in a pot experiment under greenhouse conditions. Nutrient solution was added several times at two nitrogen (N) levels. One addition involved 15N labelling to determine whether the competition of both plant species depends on their ability to acquire N from soil. Duchesnea had a higher biomass production than Fragaria when grown in nutrient-rich soil, both in competition and as a solitary plant. Under N-poor conditions, root interference could change this superiority due to limited soil space. After 65 days of growth, total plant dry weight, total N content and 15N content in the plant tissues were determined. The results show that the predominance of Duchesnea in biomass production was confirmed at high, but not at low N availability. The assimilate partitioning strategy of Duchesnea differs from that of Fragaria: the former generally had a higher shoot-to-root ratio. The N content in shoots and roots was affected only by N addition but not by competition or species. Duchesnea allocated more N to the leaves, Fragaria to the roots. The amount of 15N taken up was nearly equal for both species. In relation to root biomass, Duchesnea had a higher specific uptake rate at low N addition because of the higher root biomass in Fragaria. The roots of Fragaria and Duchesnea did not affect each other when grown together. We conclude that the invasive potential of Duchesnea is only poorly related to the N uptake rate or to better root competition for N. In N-rich environments, however, Duchesnea is highly competitive because of the preferred investment in shoot biomass. Therefore, environments with increased N deposition, i.e. from anthropogenic sources, could promote the invasive potential of Duchesnea.  相似文献

The aim of this study was to explore and compare the impacts of GS on the activities of pharmaceutical companies representing mainly original or generic pharmaceutical products in Finland.
