Actual,potential and speculative applications of seaweed cellular biotechnology: some specific comments on Gelidium 总被引:2,自引:0,他引:2
Cellular biotechnology is a promising application in the propagation and selection of superior strains of seaweeds. Although axenic cultures, organogenetic tissue cultures, vegetative micro-propagation, callus induction and high yields of agar from calli have been described for several species of Gelidium, a number of basic problems remain to be solved. These include standardized methods for obtaining axenic cultures, identification of requirements for organic nutrients, PGR's, cellular disorganization and reorganization, somaclonal variation and somatic incompatibilities. Future progress in seaweed biotechnology will depend on the resolution of many of these problems. 相似文献

The possible action of melatonin and 5-methoxytryptophol, two pineal indoles, upon castrated male rat sexual behaviour with three testosterone dose levels was studied. Both indoles reduced the number of mounts and intromissions. 5-Methoxytryptophol, in addition, decreased the number of ejaculations. Megamelus scutellaris Berg (Delphacidae) and Taosa (Cuernavaca) longula Remes Lenicov (Dictyopharidae) are specialist planthoppers that feed and reproduce on the invasive aquatic weed, Eichhornia crassipes (Martius) Solms-Laubach (Pontederiaceae). They overlap geographically in several regions of South America and may, therefore, interact and compete for food and microhabitat. Preliminary observations indicated that both species do not feed on the same part of the plant. We hypothesized that they partition the resource; hence, we studied (1) the feeding mechanism at the tissue level and (2) the spatial distribution of both species on the water hyacinth plant. Salivary sheaths were detected through histological sections of plant tissues using light microscopy. The location of either planthopper species on the plant was recorded when in the presence or absence of the other species. Both species produced true salivary sheaths, mostly branched (M. scutellaris: 82%; T. longula: 84%), ending in phloem (M. scutellaris: 56%; T. longula: 52%), and xylem tissues (M. scutellaris: 24%; T. longula: 28%). They resided on different parts of the water hyacinth plant even when they did not coexist; nymphs of T. longula occurred primarily on the back side of the leaf laminas, while nymphs of M. scutellaris occupied the basal zone of the petioles. This study shows that these planthoppers complement each other and could be used in combination as control agents for water hyacinth. Analysis of the role of interleukins in T cell ontogeny in vitro indicates that the regulation of T cell development involves interleukins (ILs) as well as thymic hormones (THs). In order to assess their respective roles in T lymphocyte development in vivo, chemically thymectomized mice were treated with ILs and THs. After 2 days of hydrocortisone treatment, aged mice showed acute thymic involution (weight was less than 30% of control) and reduced spleen size (less than 80% of control) with progressive recovery to 8 days. After 2 days of hydrocortisone treatment, adult mice were injected for 5 days with mixed buffy coat interleukins (BC-IL; 50 units IL2 equivalence), purified IL2 (50 units), rIL1 beta (4 ng), and thymosin fraction V (TF5; 100 micrograms). The animals were sacrificed and spleens and thymuses were analyzed for weight, cellularity, T cell number, subsets, and function as determined by proliferative responses to concanavalin A and ILs. BC-IL treatment increased the recovery of spleen and thymus weights and cellularity with corresponding augmentation of number and function of T lymphocytes; neither IL1 or IL2 or their combination had this effect. TF5 had no effect alone but strongly potentiated the effect of BC-IL on T lymphocyte function. These data indicate that BC-IL in combination with thymic peptides potently promotes T lymphocyte development. The combination may be therapeutically relevant for immunorestoration. 相似文献

Secreted protein, acidic and rich in cysteines (SPARC) is a secreted protein associated with increased aggressiveness of different human cancer types. In order to identify downstream mediators of SPARC activity, we performed a 2-DE proteomic analysis of human melanoma cells following antisense-mediated downregulation of SPARC expression. We found 23/504 differential spots, 15 of which were identified by peptide fingerprinting analysis. Three of the differential proteins (N-cadherin (N-CAD), clusterin (CLU), and HSP27) were validated by immunoblotting, confirming decreased levels of N-CAD and CLU and increased amounts of HSP27 in conditioned media of cells with diminished SPARC expression. Furthermore, transient knock down of SPARC expression in melanoma cells following adenoviral-mediated transfer of antisense RNA confirmed these changes. We next developed two different RNAs against SPARC that were able to inhibit in vivo melanoma cell growth. Immunoblotting of the secreted fraction of RNAi-transfected melanoma cells confirmed that downregulation of SPARC expression promoted decreased levels of N-CAD and CLU and increased secretion of HSP27. Transient re-expression of SPARC in SPARC-downregulated cells reverted extracellular N-CAD, CLU, and HSP27 to levels similar to those in the control. These results constitute the first evidence that SPARC, N-CAD, CLU, and HSP27 converge in a unique molecular network in melanoma cells. 相似文献
