Antimicrobial surfaces are one approach to prevent biofilms in the food industry. The aim of this study was to investigate the effect of poly((tert-butyl-amino)-methyl-styrene) (poly(TBAMS)) incorporated into linear low-density polyethylene (LLDPE) on the formation of mono- and mixed-species biofilms. The biofilm on untreated and treated LLDPE was determined after 48 and 168 h. The comparison of the results indicated that the ability of Listeria monocytogenes to form biofilms was completely suppressed by poly(TBAMS) (Δ168 h 3.2 log10 cfu cm?2) and colonization of Staphylococcus aureus and Escherichia coli was significantly delayed, but no effect on Pseudomonas fluorescens was observed. The results of dual-species biofilms showed complex interactions between the microorganisms, but comparable effects on the individual bacteria by poly(TBAMS) were identified. Antimicrobial treatment with poly(TBAMS) shows great potential to prevent biofilms on polymeric surfaces. However, a further development of the material is necessary to reduce the colonization of strong biofilm formers.  相似文献

The arbuscular mycorrhizal (AM) symbiosis is a key plant–microbe interaction in sustainable functioning ecosystems. Increasing anthropogenic disturbance poses a threat to AM fungal communities worldwide, but there is little empirical evidence about its potential negative consequences. In this global study, we sequenced AM fungal DNA in soil samples collected from pairs of natural (undisturbed) and anthropogenic (disturbed) plots in two ecosystem types (10 naturally wooded and six naturally unwooded ecosystems). We found that ecosystem type had stronger directional effects than anthropogenic disturbance on AM fungal alpha and beta diversity. However, disturbance increased alpha and beta diversity at sites where natural diversity was low and decreased diversity at sites where natural diversity was high. Cultured AM fungal taxa were more prevalent in anthropogenic than natural plots, probably due to their efficient colonization strategies and ability to recover from disturbance. We conclude that anthropogenic disturbance does not have a consistent directional effect on AM fungal diversity; rather, disturbance equalizes levels of diversity at large scales and causes changes in community functional structure.  相似文献

Explanations of the pattern of species have traditionally relied on small-scale, local processes occurring in ecological time. Differences in species richness have associated with different mechanisms avoiding competition, such as spatiotemporal heterogeneity (weaker competitors may find a more favourable place or time) or environmental stress (competition is assumed to be less intensive under difficult conditions). More recently, large-scale process have been taken into account, raising such questions as: which plant species may potentially grow in a certain community? Are evolutionary processes and species dispersal responsible for the differences between communities? The species-pool theory attempts to answer these general questions, and information about species pools is needed for the design of experiments where the number of species in a community is manipulated.  相似文献

Aim To investigate the application of environmental modelling to reconstructive mapping of pre‐impact vegetation using historical survey records and remnant vegetation data. Location The higher elevation regions of the Fleurieu Peninsula region in South Australia were selected as a case study. The Fleurieu Peninsula is an area typical of many agricultural regions in temperate Australia that have undergone massive environmental transformation since European settlement. Around 9% of the present land cover is remnant vegetation and historical survey records from the ad 1880s exist. It is a region with strong gradients in climate and topography. Methods Records of pre‐impact vegetation distribution made in surveyors’ field notebooks were transcribed into a geographical information system and the spatial and classificatory accuracy of these records was assessed. Maps of remnant vegetation distribution were obtained. Analysis was undertaken to quantify the environmental domains of historical survey record and remnant vegetation data to selected meso‐scaled climatic parameters and topo‐scaled terrain‐related indices at a 20 m resolution. An exploratory analytical procedure was used to quantify the probability of occurrence of vegetation types in environmental domains. Probability models spatially extended to geographical space produce maps of the probability of occurrence of vegetation types. Individual probability maps were combined to produce a pre‐impact vegetation map of the region. Results Surveyors’ field notebook records provide reliable information that is accurately locatable to levels of resolution such that the vegetation data can be spatially correlated with environmental variables generated on 20 m resolution environmental data sets. Historical survey records of vegetation were weakly correlated with the topo‐scaled environmental variables but were correlated with meso‐scaled climate. Remnant vegetation records similarly not only correlated to climate but also displayed stronger relationships with the topo‐scaled environmental variables, particularly slope. Main conclusions A major conclusion of this study is that multiple sources of evidence are required to reconstruct past vegetation patterns in heavily transformed region. Neither the remnant vegetation data nor historical survey records provided adequate data sets on their own to reconstruct the pre‐impact vegetation of the Fleurieu Peninsula. Multiple sources of evidence provide the only means of assessing the environmental and historical representativeness of data sets. The spatial distribution of historical survey records was more environmentally representative than remnant vegetation data, which reflect biases due to land clearance. Historical survey records were also shown to be classificatory and spatially accurate, thus are suitable for quantitative spatial analyses. Analysis of different spatial vegetation data sets in an environmental modelling framework provided a rigorous means of assessing and comparing respective data sets as well as mapping their predicted distributions based on quantitative correlations. The method could be usefully applied to other regions where predictions of pre‐impact vegetation cover are required.  相似文献

So far, in all studies on the much-discussed hump-backed relationship between plant community productivity and species richness, productivity has been assessed through plant shoot biomass, i.e. it has been ignored that frequently most of the biomass is produced below ground. We revisited the 27 grassland and forest field-layer communities, studied earlier by Zobel and Liira, to sample root biomass, plant total biomass and root/shoot allocation, and learn how the incorporation of below-ground biomass data would affect the shape of the hump-backed relationship. In order to avoid scaling artefacts we estimated richness as the average count of species per 500 plant ramets (absolute richness). We also included relative richness measures. Relative richness was defined as richness per 500 ramets/size of the actual species pool (the set of species present in the community), relative pool size was defined as size of the actual species pool/size of the regional species pool (the set of species available in the region and capable of growing in the given community).
The biomass-absolute richness relationship was humped, irrespective of the biomass measure used, the hump being most obvious when plant total biomass was used as the independent variable. Evidently, the unimodal richness–productivity curve is not a sampling artefact, as suspected by Oksanen. However, relative richness was not related to community biomass (above-ground, below-ground or total). The hump-backed curve is shaped by the sizes of actual species pools in communities, implying that processes which are responsible for small-scale diversity pattern mainly operate on the community level.
Neither absolute nor relative richness were significantly related to root/shoot allocation. The presumably stronger (asymmetric) shoot competition at greater allocation to shoots appears not to suppress small-scale richness. However, there is a significant relationship between relative pool size and root/shoot allocation. Relatively more species from regional species pools are able to enter and persist in communities with more biomass allocated into roots.  相似文献

An emerging pattern is that function can be predicted for almost two-thirds of the 73,500 genes that were analyzed. The effect of solution composition and enzymic proteolysis on axonemes prepared from the sperm of sea urchins, Tripneustes gratilla, has been investigated. Aliquots of axonemes, prepared by treatment of sperm with Triton X-100 and differential centrifugation, were transferred to solutions of different composition with and without intervening tryptic proteolysis, and the particle conformations observed by dark-field and electron microscopy. In most solutions particles in partially digested preparations underwent conformational transformations to coiled or helix-like forms. Proteolysis was accompanied by an increase in the ATPase activity of the digest: by centrifuging down the insoluble digestion products it was shown that digestion resulted in the appearance of ATPase activity in the soluble phase with a concomitant decrease in ATPase activity in the pellet fraction. Gel electrophoresis showed this corresponded to the appearance of dynein in the supernatant and a decrease in dynein associated with the insoluble fraction. Supernatant dynein had a greater specific ATPase activity than dynein extracted from axonemes. Observations on specimens prepared for electron microscopy by thin sectioning allowed a rough correlation to be made between the dark-field observations, chemical analyses, and morphological alterations attendant with the proteolysis and solution conditions. It is concluded that in the intact axoneme the doublet tubules are under considerable tension and that proteolytic destruction of physical restraining elements allows spontaneous conformational alterations of the digestion products. In addition, proteolysis increases the specific ATPase activity of dynein and removes a portion of it from the axonemal structure.  相似文献
