The preferred conformations of the glycerol region of a phospholipid have been explored using replica exchange molecular dynamics
(MD) simulations and compared with the results of standard MD approaches and with experiment. We found that due to isomerization
rates in key torsions that are slow on the timescale of atomistic MD simulations, standard MD is not able to produce accurate
equilibrium conformer distributions from reasonable trajectory lengths (e.g., on the 100 ns) timescale. Replica exchange MD,
however, results in quite efficient sampling due to the rapid increase in isomerization rate with temperature. The equilibrium
distributions obtained from replica exchange MD have been compared with the results of experimental nuclear magnetic resonance
observations. This comparison suggests that the sampling approach demonstrated here is a valuable tool that can be used in
evaluating force fields for molecular simulation of lipids. 相似文献

The effectiveness of hybrid vigor, which can counteract deleterious effects of inbreeding in flightless Harmonia axyridis Pallas (Coleoptera: Coccinellidae), was investigated. First, we performed the reciprocal cross between two isofemale lines
and compared survival and reproductive characteristics between isofemale and hybrid lines under laboratory conditions. The
survival of one of the hybrid lines was significantly higher than that of the two isofemale lines. Early fecundity tended
to be higher in the two hybrid lines than in the two isofemale lines. Second, we compared the effectiveness of control of
Aphis gossypii Glover (Homoptera: Aphididae) between isofemale and hybrid lines by release experiments in greenhouses. The number of A. gossypii was suppressed in treatments in which two hybrid lines were released compared with those in which two isofemale lines were

Seasonal resource availability may act as a constraint on plant phenology and thereby influence the range of growth responses
observed among populations of annual species, especially those occupying a wide range of environments. We compared a mesic
and a xeric population of the non-native, annual grass, Bromus tectorum, to examine phenology in response to interspecific competition and water availability. Using a target-neighborhood approach,
we assessed how phenological patterns of the two populations affected morphological and growth responses to enhanced resource
availability represented by late-season soil moisture. The xeric population exhibited a highly constrained phenology and was
unable to extend the growing season despite available soil resources. Because of the low phenotypic variation, allocation
to reproduction was similar across resource conditions. In contrast, the mesic population flowered later and showed a more
opportunistic phenology in response to late-season water availability. The mesic population was not able to maintain consistent
reproductive allocation at low resource levels. The responses of the two populations to late-season water availability were
not affected by the density of neighboring plants. We suggest that post-introduction selection pressure on B. tectorum in the xeric habitat has resulted in a more fixed phenology which limits opportunistic response to unpredictable, particularly
late-season resource availability. Opportunistic and fixed responses represent contrasting strategies for optimizing fitness
in temporally varying environments and, while both play important roles for ensuring reproductive success, these results suggest
that local adaptation to temporal resource variation may reflect a balance between flexible and inflexible phenology. 相似文献

Contamination of Cronobacter spp. (Enterobacter sakazakii) in infant formulas and other food products is a severe problem. Here a loop-mediated isothermal amplification (LAMP) assay
was developed for rapidly detecting Cronobacter spp. in powdered infant formula. Sequences of 16S/23S rDNA internal intergenic spacer of Cronobacter spp. were used as the target template to design LAMP primers. The detection outcome can be evaluated by the white precipitate
or the fluorescence intensity under ultraviolet irradiation, both visible to naked eyes. The sensitivity and specificity of
the LAMP assay was further analyzed in comparison with that of regular PCR and real time quantitative PCR. The results showed
that all of Cronobacter spp. strains display positive reaction to the detections while all of the non-Cronobacter spp. strains were negative, and that the LAMP assay exhibits a high sensitivity of 9.1 fg/μL (The sensitivity of regular
PCR and real time quantitative PCR is 91 and 9.1 pg/μL, respectively.). The amplified reaction could be accomplished in about
1 h, with the results visible to naked eyes. Hence, the LAMP assay developed by this study can provide a rapid and simple

BACKGROUND: Optogenetic techniques using light-driven ion channels or ion pumps for controlling excitable cells have greatly facilitated the investigation of nervous systems in vivo. A model organism, C. elegans, with its small transparent body and well-characterized neural circuits, is especially suitable for optogenetic analyses. METHODOLOGY/PRINCIPAL FINDINGS: We describe the application of archaerhodopsin-3 (Arch), a recently reported optical neuronal silencer, to C. elegans. Arch::GFP expressed either in all neurons or body wall muscles of the entire body by means of transgenes were localized, at least partially, to the cell membrane without adverse effects, and caused locomotory paralysis of worms when illuminated by green light (550 nm). Pan-neuronal expression of Arch endowed worms with quick and sustained responsiveness to such light. Worms reliably responded to repeated periods of illumination and non-illumination, and remained paralyzed under continuous illumination for 30 seconds. Worms expressing Arch in different subsets of motor neurons exhibited distinct defects in the locomotory behavior under green light: selective silencing of A-type motor neurons affected backward movement while silencing of B-type motor neurons affected forward movement more severely. Our experiments using a heat-shock-mediated induction system also indicate that Arch becomes fully functional only 12 hours after induction and remains functional for more than 24 hour. CONCLUSIONS/SGNIFICANCE: Arch can be used for silencing neurons and muscles, and may be a useful alternative to currently widely used halorhodopsin (NpHR) in optogenetic studies of C. elegans. 相似文献

Enchytraeus japonensis is a small oligochaete that reproduces mainly asexually by fragmentation (autotomy) and regeneration. As sexual reproduction can also be induced, it is a good animal model for the study of both somatic and germline stem cells. To clarify the features of stem cells in regeneration, we investigated the proliferation and lineage of stem cells in E. japonensis. Neoblasts, which have the morphological characteristics of undifferentiated cells, were found to firmly adhere to the posterior surface of septa in each trunk segment. Also, smaller neoblast‐like cells, which are designated as N‐cells in this study, were located dorsal to the neoblasts on the septa. By conducting 5‐bromo‐2′‐deoxyuridine (BrdU)‐labeling‐experiments, we have shown that neoblasts are slow‐cycling (or quiescent) in intact growing worms, but proliferate rapidly in response to fragmentation. N‐cells proliferate more actively than do neoblasts in intact worms. The results of pulse‐chase experiments indicated that neoblast and N‐cell lineage mesodermal cells that incorporated BrdU early in regeneration migrated toward the autotomized site to form the mesodermal region of the blastema, while the epidermal and intestinal cells also contributed to the blastema locally near the autotomized site. We have also shown that neoblasts have stem cell characteristics by expressing Ej‐vlg2 and by the activity of telomerase during regeneration. Telomerase activity was high in the early stage of regeneration and correlated with the proliferation activity in the neoblast lineage of mesodermal stem cells. Taken together, our results indicate that neoblasts are mesodermal stem cells involved in the regeneration of E. japonensis. 相似文献

We investigated relationships between the flowering phenology of Prunus yedoensis "Somei-yoshino" (cherry blossom) and the local temperatures in Japan. Our observations were carried out across the Okayama Plain, which included Okayama City (about 700,000 inhabitants), from the winter of 2008 to the spring of 2009. Local air temperature (AT) and the globe temperature (GT) were recorded at the tree height. The flowering dates (FDs) of P. yedoensis were earliest in the central commercial area (located at the center of the plain), followed by the north residential area (further inland), and finally the south residential area (seaward). The recorded FDs were related to the period-averaged daily maximum/minimum AT and GT, and the phenologically effective AT and GT defined in this study. Of these parameters, the phenologically effective GTs correlated most with the FDs. Since the GT is determined by AT, solar and infrared radiations, and wind speed, our previous result suggests that a combination of these three components surrounding the tree is more important for budding and flowering than is AT alone. The supposition is supported by the flowering of P. yedoensis being the latest at the coastal region of the Okayama Plain where the AT were higher than at the inland region, excluding the urban area; it is probably caused by stronger winds there than at the other sites. 相似文献

Cytoplasmic axin expression frequently produces punctuate structures in cells, but the nature of axin puncta has not been fully elucidated. In an effort to analyze cytoplasmic axin puncta, we established HeLa cells expressing axin in a doxycycline-inducible manner (HeLa-Axin). We observed that axin accumulated in an aggregate-like pattern in perinuclear areas and appeared to be associated with mitochondria, Golgi apparatus, and endoplasmic reticulum (ER), but not lysosomes. Further biochemical analysis suggested that some part of the cytoplasmic axin pool was associated with mitochondria. In addition, mitochondrial proteins [i.e., cytochrome oxidase IV (CoxIV) and cytochrome c] were slightly higher in HeLa-Axin cells than in HeLa-EV cells, suggesting altered mitochondrial degradation. HeLa-Axin cells were then treated with staurosporine (STS) to determine if the mitochondria-induced apoptosis pathway was altered. Compared to STS-treated control cells (HeLa-EV), HeLa-Axin cells had less STS-induced cytotoxicity and reduced caspase-3 activation and PARP cleavage. Given that mitochondria outer membrane potential was unchanged, HeLa-Axin cells might be relatively resistant to STS-mediated mitochondrial damage. Mitochondria associated with axin aggregates were resistant to detergent-mediated permeabilization. These results suggest that axin forms aggregate-like structures in association with mitochondria, which render mitochondria resistant to STS-induced membrane damage and cytotoxicity. 相似文献

The transport of co-encapsulated solutes through the melittin-induced pores in the membrane of giant phospholipid vesicles
was studied, and the characteristics of the pore formation process were modeled. Molecules of two different sizes (dextran
and the smaller, fluorescent marker Alexa Fluor) were encapsulated inside the vesicles. The chosen individual vesicles were
then transferred by micromanipulation from the stock suspension to the environment with the melittin (MLT). The vesicles were
observed optically with a phase-contrast microscope and by monitoring the fluorescence signal. Such an experimental setup
enabled an analysis of a single vesicle’s response to the MLT on the basis of simultaneous, separate measurements of the outflow
of both types of encapsulated molecules through the MLT-induced pores in the membrane. The mechanisms of the MLT’s action
were suggested in a model for MLT pore formation, with oligomeric pores continuously assembling and dissociating in the membrane.
Based on the model, the results of the experiments were explained as a consequence of the membrane’s permeability dynamics,
with a continuously changing distribution of pores in the membrane with regard to their size and number. The relatively stable
“average MLT pore” characteristics can be deduced from the proposed model. 相似文献
