Protein import to the nucleus is a signal-mediated process that exhibits saturation kinetics. We investigated whether signal bearing proteins compete with U2 and U6 snRNPs during import. When injected into Xenopus oocytes, saturating concentrations of P(Lys)-BSA, a protein bearing multiple nuclear localization signals from SV40 large T-antigen, reduce the rate of [125I]P(Lys)-BSA and of [125I]nucleoplasmin import, consistent with their competing for and sharing the same limiting component of the import apparatus. In contrast, saturating concentrations of P(Lys)-BSA do not reduce the rate of HeLa [32P]U2 snRNP assembly or import. The import of U6 snRNP is also competed by P(Lys)-BSA. We conclude that U2 snRNP is imported into oocyte nuclei by a kinetic pathway that is distinct from the one followed by P(Lys)-BSA, nucleoplasmin, and U6 snRNP. 相似文献

Biological soil crusts (BSCs) cover non-vegetated areas in most arid and semiarid ecosystems. BSCs play a crucial role in the redistribution of water and sediments and, ultimately, in the maintenance of ecosystem function. The effects of BSCs on water infiltration are complex. BSCs increase porosity and micro-topography, thus enhancing infiltration, but, at the same time, they can increase runoff by the secretion of hydrophobic compounds and clogging of soil pores upon wetting. BSCs confer stability on soil surfaces, reducing soil detachment locally; however, they can also increase runoff, which may increase sediment yield. Although the key role of BSCs in controlling infiltration–runoff and erosion is commonly accepted, conflicting evidence has been reported concerning the influence of BSCs on runoff generation. Very little is known about the relative importance of different BSC features such as cover, composition, roughness, or water repellency, and the interactions of these attributes in runoff and erosion. Because BSC characteristics can affect water flows and erosion both directly and indirectly, we examined the direct and indirect effects of different BSC features on runoff and erosion in a semiarid ecosystem under conditions of natural rainfall. We built structural equation models to determine the relative importance of BSC cover and type and their derived surface attributes controlling runoff and soil erosion. Our results show that the hydrological response of BSCs varies depending on rainfall properties, which, in turn, determine the process governing overland flow generation. During intense rainfalls, runoff is controlled not only by rainfall intensity but also by BSC cover, which exerts a strong direct and indirect influence on infiltration and surface hydrophobicity. Surface hydrophobicity was especially high for lichen BSCs, thus masking the positive effect of lichen crust on infiltration, and explaining the lower infiltration rates recorded on lichen than on cyanobacterial BSCs. Under low intensity, rainfall volume exerts a stronger effect than rainfall intensity, and BSC features play a secondary role in runoff generation, reducing runoff through their effect on surface micro-topography. Under these conditions, lichen BSCs presented higher infiltration rates than cyanobacterial BSCs. Our results highlight the significant protective effect against erosion exerted by BSCs at the plot scale, enhancing surface stability and reducing sediment yield in both high- and low-magnitude rainfall events. 相似文献

The morphological changes in the gill chloride cells of the armored catfish, Hypostomus tietensis , were investigated after 15 days' exposure to either distilled or hard water. The thickness of the water–blood barrier in the lamellae increased significantly in fish kept in distilled water due to the high proliferation of chloride cells. The apical surface of about 68% of chloride cells was sharply reduced by the development of an apical crypt with a sponge-like surface, although no change in the chloride cell fractional area was found. In contrast, H. tietensis kept in Na+ , Cl− and Ca2+ rich water displayed no significant changes in the number of chloride cells or in their apical surface morphology compared with the control fish. Chloride cell response to ion challenge in H. tietensis suggested the involvement of different strategies to maintain homeostasis in ion-poor water, which may be related to the life history of species. 相似文献

Resistance to endoparasitoids in aphids involves complex interactions between insect and microbial players. It is now generally accepted that the facultative bacterial symbiont Hamiltonella defensa of the pea aphid Acyrthosiphon pisum is implicated in its resistance to the parasitoid Aphidius ervi. It has also been shown that heat negatively affects pea aphid resistance, suggesting the thermosensitivity of its defensive symbiosis. Here we examined the effects of heat and UV-B on the resistance of A. pisum to A. ervi and we relate its stability under heat stress to different facultative bacterial symbionts hosted by the aphid. For six A. pisum clones harboring four different facultative symbiont associations, the impact of heat and UV-B was measured on their ability to resist A. ervi parasitism under controlled conditions. The results revealed that temperature strongly affected resistance, while UV-B did not. As previously shown, highly resistant A. pisum clones singly infected with H. defensa became more susceptible to parasitism after exposure to heat. Interestingly, clones that were superinfected with H. defensa in association with a newly discovered facultative symbiont, referred to as PAXS (pea aphid X-type symbiont), not only remained highly resistant under heat stress, but also expressed previously unknown, very precocious resistance to A. ervi compared to clones with H. defensa alone. The prevalence of dual symbiosis involving PAXS and H. defensa in local aphid populations suggests its importance in protecting aphid immunity to parasitoids under abiotic stress. 相似文献

Three dermaseptins, DS 01, DD K, and DD L, were compared with respect to their structural features and interactions with liposomes. Circular dichroic spectra at alcohols of different chain lengths revealed that DS 01 has the higher helicogenic potential in hydrophobic media. Binding of DS 01, DD K, and DD L to liposomes induced significant blue shifts of the emission spectra of the single tryptophan located at position 3 of all sequences indicating association of the peptides with bilayers. Kinetics evaluation of atomic force microscopy images evidenced the strong fusogenic activity of DS 01 whereas DD K and DD L showed increased lytic activities. 相似文献

We employed a prime-boost regimen in combination with the expression library immunization protocol to improve the protective effectiveness of a genomic library used as immunogen. To demonstrate the feasibility of this novel strategy, we used as a prime a serogroup B Neisseria meningitidis random genomic library constructed in a eukaryotic expression vector. Mice immunized with different fractions of this library and boosted with a single dose of meningococcal outer membrane vesicles elicited higher bactericidal antibody titers compared with mice primed with the empty vector. After the boost, passive administration of sera from mice primed with two of these fractions significantly reduced the number of viable bacteria in the blood of infant rats challenged with live N. meningitidis. The method proposed could be applied to the identification of subimmunogenic antigens during vaccine candidate screening by employing expression library immunization. 相似文献

Four strains of endophytic microorganisms isolated from carrot root were able to carry out the reduction of the carbonyl group with diverse degree of enantio-, and diasteroselectivity. Furthermore, biotransformation in the presence of bacterial inhibitor affects the stereochemical outcome of the reaction, and the concomitant addition of a yeast inhibitor results in a large decrease in the conversion percentage. These results indicate that endophytic microorganisms might be involved in the enantioselective reduction of ketones and ketoesters with fresh carrot root pieces. 相似文献
