(+)-Borneol suppresses conditioned fear recall and anxiety-like behaviors in mice

Fear- and anxiety-related psychiatric disorders have been one of the major chronic diseases afflicting patients for decades, and new compounds for treating such disorders remain to be developed. (+)-Borneol, a bicyclic monoterpene found in several species of Artemisia and Dipterocarpaceae, is widely used for anxiety, pain and anesthesia in Chinese medicine. Meanwhile, it can potentiate GABA (γ-aminobutyric acid) activity directly in recombinant GABAA receptors. The present study was to investigate the effects of (+)-Borneol on both contextual and cued fear recall. Interestingly, microinjection of (+)-Borneol into the dorsal hippocampus inhibited 24 h and 7 d contextual fear, whereas its infusion into ventral hippocampus only reduced 24 h cued fear responses. Moreover, microinjection of (+)-Borneol into dorsal but not ventral hippocampus suppressed anxiety-like behaviors in the open field test, light/dark exploration and the elevated plus maze test. As selective GABA_A receptor antagonist bicuculline reversed the effect of (+)-Borneol on contextual fear paradigm and the drug potentiated GABA-evoked currents in acute hippocampus slices, modulation of the GABAergic neurotransmission may explain the effects of (+)-Borneol. Our findings suggest that (+)-Borneol can serve as a new therapeutic in fear- and anxiety-related disorders.     

Kakeromamide A,a new cyclic pentapeptide inducing astrocyte differentiation isolated from the marine cyanobacterium Moorea bouillonii

Kakeromamide A (1), a new cyclic pentapeptide encompassing a thiazole ring moiety and a β-amino acid, was isolated from the marine cyanobacterium Moorea bouillonii. Its structure was elucidated by the spectral analysis and the modified Marfey’s method. Compound 1 induced differentiation of neural stem cells into astrocytes at the concentration of 10?µM.     

Adapting Strategy of Biennial Endangered <Emphasis Type="Italic">Halenia corniculata</Emphasis> (L.) Cornaz to Environmental Stress and Its Implications for Conservation Plan

Under intensive environmental stress, reported that plants have complementary and positive interactions instead of competition among species. In order to examine whether there as a possible correlation between the performance of Halenia corniculata and the coverage/height of other plants with the same distribution, a total of 30 plots were established at two representative distributional areas. The height, total number of chasmogamous flowers and cleistogamous flowers, their overall plots coverage, and observed plants species on each plot were determined. Our results revealed that the number of chasmogamous and cleistogamous flowers of Halenia corniculata was increased when its height was increased. The performance of Halenia corniculata was found to have a significant correlation with the overall coverage of observed plants species in the plot as well as the height of observed plants in established plots. Our results suggest that the biennial plant Halenia corniculata might be able to choose a site inhabited by other perennial plants in the high-altitude region under environmental stress for appropriate growth and reproduction. One main reason that Halenia corniculata seedlings could be established in regions where other plants are inhabited might be due to the degree of light requirement by the seedling stage is different from that at the adult stage. The size of Halenia corniculata population in South Korea was found to be proportional to that of available sites that could support its life cycle, suggesting that available sites need to be secure in order to maintain its life cycle.     

A method for expansion of T cells from cynomolgus monkey (<Emphasis Type="Italic">Macaca fascicularis</Emphasis>)

T cells have been successfully applied to cancer immunotherapy. However, a challenge is an expansion of T cells from cynomolgus monkey, which is a valuable non-human primate model for T cell therapy transferring to the clinic. Here, we compared several strategies to expand cynomolgus monkey T cell and developed an appropriate method. Our study demonstrated that 100 ng/ml CD3 mAb?+?1% PHA+ 1000 U/ml IL2 therapy significantly expanded peripheral blood CD3+ T cells without compromising T cell phenotype in vitro. The results of this study could be used for T cell remodeling, which has significant therapeutic potential in Chimeric Antigen Receptor-T (CAR-T) cell immunotherapy.     

Discovery of 4′-OH-flurbiprofen Mannich base derivatives as potential Alzheimer’s disease treatment with multiple inhibitory activities

A series of 4′-OH flurbiprofen Mannich base derivatives were designed, synthesized and evaluated as potential multifunctional agents for the treatment of Alzheimer’s disease. The biological screening results indicated that most of these derivatives exhibited good multifunctional activities. Among them, compound 8n demonstrated the best inhibitory effects on self-induced Aβ_1-42 aggregation (65.03% at 25.0?μM). Moreover, this representative compound also exhibited good antioxidant activity, biometal chelating ability and anti-neuroinflammatory activity in vitro. Furthermore, compound 8n displayed appropriate blood-brain barrier permeability. These multifunctional properties highlight compound 8n as promising candidate for further development of multi-functional drugs against AD.
