Japanese red pine (Pinus densiflora) is widely present in China, Japan, and Korea. Its green pine leaves have traditionally been used as a food as well as a coloring agent. After being shed, pine leaves change their color from green to brown within two years, and although the brown pine leaves are abundantly available, their value has not been closely assessed. In this study, we investigated the potential anti-photoaging properties of brown pine leaves for skin. Brown pine leaf extract (BPLE) inhibited UVB-induced matrix metalloproteinase-1 (MMP-1) expression to a greater extent than pine leaf extract (PLE) in human keratinocytes and a human skin equivalent model. HPLC analysis revealed that the quantity of trans-communic acid (TCA) and dehydroabietic acid (DAA) significantly increases when the pine leaf color changes from green to brown. BPLE and TCA elicited reductions in UVB-induced MMP-1 mRNA expression and activator protein-1 (AP-1) transactivation by reducing DNA binding activity of phospho-c-Jun, c-fos and Fra-1. BPLE and TCA also inhibited UVB-induced Akt phosphorylation, but not mitogen activated protein kinase (MAPK), known regulators of AP-1 transactivation. We additionally found that BPLE and TCA inhibited phosphoinositide 3-kinase (PI3K), the upstream kinase of Akt, in vitro. In summary, both BPLE and its active component TCA exhibit protective effects against UVB-induced skin aging. Taken together, these findings underline the potential for BPLE and TCA to be utilized as anti-wrinkling agents and cosmetic ingredients, as they suppress UVB-induced MMP-1 expression.  相似文献

There are about 15 species of Gracilaria reported in Vietnam. Of these, Gracilaria asiatica Zhang et Xia is being cultivated on a large scale in Northern Vietnam, which has a subtropical climate. During the rainy season, from May to October, the growth of G. asiatica is drastically reduced or even ceases due to very low salinity and high temperature. Therefore, it is important to improve the tolerance of G. asiatica to a wide range of salinity and temperatures. This paper presents the results of research on strain improvement of G. asiatica using irradiation and selection media. Three irradiation doses of 20, 60, and 100 krad were tested against the control (with no irradiation). Afterward, the seaweed biomass was cultivated on a selected medium, ESS-1, containing NaCl in concentrations of 23‰ (C1) and 0‰ (C2). The results showed that a higher survival rate of G. asiatica was observed with the 20- and 60-krad doses. The protein content and composition of selected seaweeds were analyzed and compared with the control. SDS-PAGE showed no remarkable difference in the protein composition between the control and irradiated samples. However, the 67-kDa protein band of seaweed treated with 20 and 60 krad, then grown on ESS-1 medium with 23% NaCl, had a higher density than other samples. This protein was reported to play an important role in G. asiatica, by enhancing its tolerance to variable salinity and temperature. Although the organic and inorganic content of all samples remained almost the same, the content and quality of agar extracted from irradiated seaweeds were higher than those of the controls. Due to the high amount of 3.6 anhydro-α-L-galactose combined with low amounts of sulfate found in irradiated seaweeds, the freezing and melting points of extracted agar were lower. Eventually, this resulted in higher condensation and better quality of agar, such as in its gel-forming ability. The quality of fluid agar extracted from selected seaweeds improved as shown in the remarkable decrease in Ca2+, Mg2+, and total Fe ion content, thus lowering its melting point compared with the control. The major constituents from the fruits of Maclura pomifera are the prenylated isoflavones, osajin (1) and pomiferin (2). Their structures were elucidated using NMR spectroscopic techniques and mass spectrometric analysis. Compound 2 showed potential inhibitory activity in histone deacetylase (HDAC) enzyme assay. It also exhibited growth inhibitory activity on five human tumor cell lines and more sensitive inhibitory activity on the HCT-15 colon tumor cell line. Further structure-activity relationships of position 3 on ring B from aromatic ring will be reported in due course.  相似文献

Three tyrosyl gallate derivatives (1-3) with variable hydroxyl substituent at the aromatic ring of tyrosol were synthesized and evaluated as potent inhibitors on tyrosinase activity and melanin formation in melan-a cells. Among three tyrosyl gallate derivatives, 4-hydroxyphenethyl 3,4,5-trihydroxybenote (1) (IC(50)=4.93 microM), 3-hydroxyphenethyl 3,4,5-trihydroxybenote (2) (IC(50)=15.21 microM), and 2-hydroxyphenethyl 3,4,5-trihydroxybenote (3) (IC(50)=14.50 microM) exhibited significant inhibitory effect on tyrosinase activity. Compound 1 was the most active compound, though it did not show the inhibitory effect on melanin formation in melan-a cells. However, compounds 2 (IC(50)=8.94 microM) and 3 (IC(50)=13.67 microM) significantly suppressed the cellular melanin formation without cytotoxicity. This study shows that the position of hydroxyl substituent at the aromatic ring of tyrosol plays an important role in the intracellular regulation of melanin formation in cell-based assay system.  相似文献

In this study, dioscin was isolated from Dioscoreae Rhizoma (DR), which is the rhizome of Dioscorea batatas D(ECNE). that inhabits broad areas of Korea and Japan. To determine whether dioscin induced growth hormone (GH) release, we evaluated its induction effects on GH release both in vitro and in vivo. The 70% methanol extract of DR, and its n-hexane and n-BuOH fractions, induced rat GH (rGH) release in rat pituitary cells 10-fold, 8-fold, and 5- fold higher than the control (0.36 +/- 0.02 nM), respectively (p < 0.05 each). The dioscin-induced rGH release of the cells was concentration-dependent and its ED(50) was 1.14 x 10(-5) M. Within 90 minutes after intravenous administration of 10 microg/kg (p < 0.05 at t(max)), dioscin caused the greatest increase in rGH concentration (C(max)) in the rat plasma (34.16 +/- 14.10 ng/ml) (n = 4), which was twice as high as the control group (12.88 +/- 3.29 ng/ml) (n = 27).  相似文献

To identify and assess the distribution patterns of coastal dune vegetation along the eastern, southern, and western coasts of South Korea, we investigated the plant communities and soil factors at 30 sites. In all, 12 communities on CCA (canonical correspondence analysis) Axis 1 and 2 could be arranged into 3 groups: 1 ) 2 communities ofElymus mollis andIschaemum anthephoroides, with medium sand contents; 2) 6 communities ofCarex pumila, Carex kobomugi, Ixeris repens, Zoysia mac-rostachya, Calystegia soldanella, andVitex rotundifolia, with coarse sand contents; and 3) 4 communities ofLathyrus japonicus,Glehnia littoralis, Messerschmidia sibirica, andRosa rugosa, with very coarse sand contents. As identified via CCA ordination, the distribution of these groups was positively correlated with soil particle sizes.  相似文献
