Normal rat kidney cells infected with a cold-sensitive mutant of mouse sarcoma virus [NRK(MSV-lb)] morphologically transform when exposed to adenosine 3':5' cyclic monophosphate (cAMP) at the restrictive temperature. The cAMP-induced morphological changes occur rapidly and are reversible. Agents capable of elevating endogenous levels of cAMP [prostaglandin E1 (PGE1) and cholera toxin (CT)] induced morphological transformation of NRK(MSV-lb) cells at the restrictive temperature that was concentration dependent, potentiated by cAMP phosphodiesterase inhibitors, and not prevented by inhibitors of DNA, RNA, and protein synthesis. Prostaglandin E1 stimulated a transient increase in the intracellular level of cAMP with a concomitant morphological transformation and reversion of cells as cAMP levels decline. The maximum increase is reached by 10 min, followed by a decline to near basal level by 80 min. In contrast, incubation of cells with CT resulted in irreversible morphological transformation and increased levels of cAMP first detectable by 1 hr with maximum levels reached by 24 hr. Heated CT (100 degrees C, 20 min) was without effect. Addition of CT to reverted PGE1-treated cells resulted in morphological transformation suggesting the existence of discrete receptors in NRK(MSV-lb) cells.  相似文献

木本植物次生木质部轴向薄壁组织和射线薄壁组织在物质存储和转运等功能中起着重要的作用, 剖析次生木质部薄壁组织组成有助于深入探究其功能, 而小枝木质部薄壁组织组成特征还缺乏研究。该研究以天马国家级自然保护区内的18种木本植物幼树为研究对象, 测算了各物种小枝木质部薄壁组织的组成含量并检测了其谱系信号, 结合有关树干薄壁组织含量数据集, 初步探讨木本植物小枝木质部薄壁组织含量特征。研究结果表明: (1) 18种木本植物幼树小枝的薄壁组织总含量为9.96%-18.56%, 平均为14.80%; 其中射线薄壁组织含量为7.74%-15.45%, 高于轴向薄壁组织的含量(1.13%-7.49%); (2)小枝中薄壁组织总含量呈低于树干的趋势, 其中小枝射线薄壁组织含量低于树干, 而轴向薄壁组织含量高于树干, 这可能是由器官差异和生活史阶段差异造成的; (3)轴向薄壁组织的含量具有显著的谱系信号, 即物种亲缘关系越近其含量越相近。该研究初步验证了木本植物次生木质部薄壁组织的谱系信号, 同时暗示了器官和生活史阶段差异对薄壁组织含量具有重要影响。  相似文献

With the development of genetic maps and the identification of the most-likely positions of quantitative trait loci (QTLs) on these maps, molecular markers for lodging resistance can be identified. Consequently, marker-assisted selection (MAS) has the potential to improve the efficiency of selection for lodging resistance in a breeding program. This study was conducted to identify genetic loci associated with lodging resistance, plant height and reaction to mycosphaerella blight in pea. A population consisting of 88 recombinant inbred lines (RILs) was developed from a cross between Carneval and MP1401. The RILs were evaluated in 11 environments across the provinces of Manitoba, Saskatchewan and Alberta, Canada in 1998, 1999 and 2000. One hundred and ninety two amplified fragment length polymorphism (AFLP) markers, 13 random amplified polymorphic DNA (RAPD) markers and one sequence tagged site (STS) marker were assigned to ten linkage groups (LGs) that covered 1,274 centi Morgans (cM) of the pea genome. Six of these LGs were aligned with the previous pea map. Two QTLs were identified for lodging resistance that collectively explained 58% of the total phenotypic variation in the mean environment. Three QTLs were identified each for plant height and resistance to mycosphaerella blight, which accounted for 65% and 36% of the total phenotypic variation, respectively, in the mean environment. These QTLs were relatively consistent across environments. The AFLP marker that was associated with the major locus for lodging resistance was converted into the sequence-characterized amplified-region (SCAR) marker. The presence or absence of the SCAR marker corresponded well with the lodging reaction of 50 commercial pea varieties.Communicated by H. F. Linskens  相似文献

Although Curcuma L. (Zingiberaceae) is a conserved name, with C. longa L. as its conserved type, the type of C. longa is still uncertain. Numerous discussions about the identity of the taxon called C. longa by Linnaeus have been followed by various attempts to rename turmeric, suggestions as how to settle the type and proposals to conserve the name from a later author in order to stabilize the situation. Unfortunately, none of the previous proposals can be upheld for reasons which are discussed in this article. A lectotype is selected from extant material examined by Linnaeus and an epitype collected near the type locality is designated here. The identity of C. longa is discussed and a colour plate of the species is included. 通过对祁连山青海云杉林地土壤水分含量及主要影响因子的测定,结果表明,青海云杉林地土壤具有容重低、入渗率高、土壤水分含量垂向分布受降水影响明显等特征,灌木-青海云杉林、苔藓-青海云杉林和草地(对照)平均容重分别为0.522、0.641、0.874g·cm^-3.土壤初渗率分别为6.0、21.0、2.5mm·min^-1,稳渗率分别为3.98、11.2、0.5mm·min^-1.苔藓-青海云杉林地土壤可划分为4层:水分活跃层(0~30cm)、水分调节层(30~60cm)、水分传输层(60~80cm)和蓄水层(80ClTI以下),各层土壤贮水量分别为3.30、3.17、2.80和2.83mm·cm^-1.灌木-青海云杉林可分为3层:水分稳定层(0~30cm)、水分活跃层(30~60cm)和水分传输层(60cm以下),各层土壤贮水量分别为3.98、3.65、3.48mm·cm^-1.两种林地土壤水分含量的季节变化受降水影响较大,相比而言,苔藓.青海云杉林地入渗率更高,有较明显的蓄水层,水分传输能力更强,而灌木-青海云杉林地贮水能力更强.  相似文献

The nadD gene, encoding the enzyme nicotinic acid mononucleotide (NaMN) adenylyltransferase (AT), is essential for the synthesis of NAD and subsequent viability of the cell. The nadD gene in Bacillus subtilis (yqeJ) was identified by sequence homology with other bacterial nadD genes and by biochemical characterization of the gene product. NaMN AT catalyzes the reversible adenylation of both NaMN and the nicotinamide mononucleotide (NMN) but shows specificity for the nicotinate. In contrast to other known NMN ATs, biophysical characterizations reveal it to be a dimer. The NaMN AT crystal structure was determined for both the apo enzyme and product-bound form, to 2.1 and 3.2 A, respectively. The structures reveal a "functional" dimer conserved in both crystal forms and a monomer fold common to members of the nucleotidyl-transferase alpha/beta phosphodiesterase superfamily. A structural comparison with family members suggests a new conserved motif (SXXXX(R/K)) at the N terminus of an alpha-helix, which is not part of the shared fold. Interactions of the nicotinic acid with backbone atoms indicate the structural basis for specificity.  相似文献

We evaluated the effect of temperature regimes (six constant and four alternating temperatures), light qualities (five red : far red ratios) and water potentials (ΨW; seven NaCl and polyethylene glycol 6000 [PEG] solutions) on the percentage and germination rate, as well as the post‐seminal development morphology, that allow Pilosocereus arrabidae seeds to germinate in a hot semiarid climate on the south‐eastern Brazilian coast. The results showed that seeds germinated similarly between constant and alternating temperatures, with an optimal germination at 25/20°C and 20°C. Pilosocereus arrabidae seeds were photoblastic positive and the final germination percentage was inhibited at low red : far red ratios. Maximum germination was obtained in distilled water (0 MPa) and decreases of ΨW in the solutions reduced the germination, which was lower in NaCl than in iso‐osmotic PEG solutions. Germination inhibition appears to be osmotic because the recovery response was high when non‐germinated seeds from both iso‐osmotic solutions were transferred to water. Seeds of P. arrabidae are small and germination is phaneroepigeal. Despite the slow growth typically seen in seedlings and adults of Cactaceae, germination in this species depends on the ability of the seeds to appropriately sense and react to environmental cues that correlate with times and places under low‐risk growth conditions.  相似文献

Hop chlorotic disease was first described in England in 1930, but it has since been seldom seen and its etiology has remained unknown. In 1983 a patch of plants with the disease occurred in a large area of hops (Humulus lupulus) cv. Bramling Cross planted at Yalding, Kent in 1967. All plants in a rectangular area enclosing the disease outbreak were infected with hop mosaic, hop latent and prunus necrotic ringspot viruses; the diseased plants were additionally infected with arabis mosaic virus (AMV). The disease was also associated with seed-transmitted AMV, and was induced in hop seedlings inoculated with partially purified preparations of AMV originating from chlorotic disease-affected hops prepared from Chenopodium quinoa. The disease appears to be caused by AMV, but AMV isolates from hops with chlorotic disease were serologically indistinguishable from AMV isolates from hops with symptoms of bare-bine and/or nettlehead and showed similar pathogenicity in diagnostic hosts. The basis of the difference between isolates in their pathogenicity in hop remains unknown.  相似文献
