Mineralocorticoid activity of glycyrrhetinic acid (GR) was studied in vivo (electrical potential difference in rat rectum) and in vitro (brush border Mg2+-HCO3- ATPase in rat small intestine, kidney cytosol binding of GR with and without RU-28362, anti-glucocorticoid compound) in order to clarify the mechanism of mineralocorticoid-like activity of GR. Scatchard analysis of [3H]aldosterone showed that Kd of higher affinity site (type I) 6.0 X 10(-9) M, Bmax 1.0 X 10(-14) mol/mg protein, and Kd of lower affinity site (type II) 1.6 X 10(-7) M, Bmax 7.5 X 10(-14) mol/mg protein. GR competed for [3H]aldosterone binding sites in kidney cytosol at the concentration of 10(4) times as that of unlabeled aldosterone. RU-28362 displaced aldosterone binding curve, whereas GR binding kinetic was not affected by this compound. Adrenalectomy caused a significant fall in brush border Mg2+-HCO3- ATPase activity (75% reduction compared with the initial level) which was not restored by GR administration. Forty cases of lymphoma were categorized as Burkitt-type lymphoma in a study of fine needle aspiration (FNA) smears. These constituted 14.3% of all cases of non-Hodgkin's lymphoma diagnosed between 1974 and 1982. The median age was 22 years in these cases, 81.8% of which had extranodal tumors. The majority of the cells in the smears (59.8% +/- 8.32%) were in the 11 micron to 15 micron size range and 60.3% +/- 10.3% had noncleaved nuclei. An average 71% of the cells contained cytoplasmic and/or nuclear vacuolizations. Nonneoplastic macrophages were present in the smears in 87.5% of the cases. A study of paraffin-embedded sections in 17 cases revealed the characteristic "starry-sky" appearance in 11; in 5 it was not clearly appreciated and in 1 the nonneoplastic macrophages were absent. FNA cytology was found to be quite reliable for arriving at a diagnosis of Burkitt-type lymphoma. More than 50% of the cases were managed without resort to subsequent surgical biopsy. Exploratory laparotomy was avoided in 69% of the cases having abdominal tumors. 相似文献

The effect of Ep on radioactive glucose and methyl-alpha-D-glucoside transport by rat erythrocytes and bone marrow cells were studied. There is initial linearity followed by saturation kinetics of [14C]glucose transport by the erythrocytes of starved and starved plus Ep-treated rats at different concentrations of glucose. Starvation caused slight inhibition of glucose transport which increased markedly on Ep administration to starved rats. Normal animals failed to show any significant change in glucose transport after Ep treatment. Methyl-alpha-D-glucoside inhibited the Ep-stimulated glucose transport significantly. Ep also stimulated the transport of radioactive methyl-alpha-D-glucoside which was competitively inhibited in presence of D-glucose. Glucose transport in erythrocytes was found to be sensitive to metabolic inhibitors like azide and DNP. A sulfhydryl reagent and ouabain also inhibited the transport process. Ep stimulated glucose and methyl-alpha-D-glucoside transport in the bone marrow cells of starved rats. The sugar analog competitively inhibited the glucose transport in bone marrow cells and vice versa. 相似文献

Summary An in vitro culture system has been developed for barley (Hordeum vulgare), which yields high frequencies of high quality microspore-derived embryos without an intervening callus phase. The embryos are very similar to zygotic embryos with regard to their morphology and germination capacity. These embryos were encapsulated in sodium alginate to produce individual beads containing one embryo each. In accordance with the literature, these beads are denoted artificial seeds. The artificial seeds germinated well and with a root system superior to that of non-encapsulated embryos. The artificial seeds also maintained their germination capacity for at least 6 months, whereas non-encapsulated embryos did not survive more than 2 weeks in storage. Artificial seeds, thus, probably provide a simple and universal delivery system of in vitro plantlets to greenhouse or field. 相似文献

A monomeric glycoprotein (SGP) of Mr 32,000 was isolated to purity from scorpion (Heterometrus bengalensis) haemolymph by (NH4)2SO4 fractionation, chromatofocusing and h.p.l.c. The homogeneity of SGP is confirmed by polyacrylamide-gel electrophoresis. SGP is soluble in 100%-satd. (NH4)2SO4 solution. Needle-shaped crystals of SGP were obtained in an aqueous environment. The glycan part of the molecule contains arabinose, which does not commonly occur in animal glycoproteins. Amino acid analysis demonstrated a preponderance of glycine, tyrosine and glutamic acid. SGP enhances phenol oxidase (EC 1.14.18.1) activity. 相似文献

Mesophyll protoplasts from leaves of aseptically grown shoot tips of Diplotaxis muralis were isolated (6.2–7.1×105 protoplasts/g fresh weight of tissue) using one step enzyme digestion. The protoplasts (71% viability) underwent divisions (4.2+0.1%) on plating in M8PS2 medium and ultimately formed calli with 0.45+0.03% plating efficiency. Plant regeneration could be achieved both through embryogenesis and organogenesis. The efficiency of plant regeneration through organogenesis was 9 times higher than embryogenesis. Forty eight out of 52 plants regenerated so far from 3 independent experiments were normal with respect to fertility and meiotic chromosomal behavior.Abbreviations BAP

Vicia faba lectin contained 40-57% beta-conformation, 4-23% alpha-conformation along with random coil at pH 7.2 depending upon the analytical methods used. The percentage of beta-conformation increased with the addition of N-acetyl-D-glucosamine or methyl alpha-D-mannopyranoside. The structural transitions of V. faba lectin were affected by alkali at pH 9.6 and 10.6. Binding constants and free energy changes for the interaction between V. faba lectin and N-acetyl-D-glucosamine and methyl alpha-D-mannopyranoside were estimated at pH 7.2 using the c.d. and fluorescence methods. 相似文献
