Trichomonas vaginalis is a parasitic protist of the Excavata group. It contains an anaerobic form of mitochondria called hydrogenosomes, which produce hydrogen and ATP; the majority of mitochondrial pathways and the organellar genome were lost during the mitochondrion-to-hydrogenosome transition. Consequently, all hydrogenosomal proteins are encoded in the nucleus and imported into the organelles. However, little is known about the membrane machineries required for biogenesis of the organelle and metabolite exchange. Using a combination of mass spectrometry, immunofluorescence microscopy, in vitro import assays and reverse genetics, we characterized the membrane proteins of the hydrogenosome. We identified components of the outer membrane (TOM) and inner membrane (TIM) protein translocases include multiple paralogs of the core Tom40-type porins and Tim17/22/23 channel proteins, respectively, and uniquely modified small Tim chaperones. The inner membrane proteins TvTim17/22/23-1 and Pam18 were shown to possess conserved information for targeting to mitochondrial inner membranes, but too divergent in sequence to support the growth of yeast strains lacking Tim17, Tim22, Tim23 or Pam18. Full complementation was seen only when the J-domain of hydrogenosomal Pam18 was fused with N-terminal region and transmembrane segment of the yeast homolog. Candidates for metabolite exchange across the outer membrane were identified including multiple isoforms of the β-barrel proteins, Hmp35 and Hmp36; inner membrane MCF-type metabolite carriers were limited to five homologs of the ATP/ADP carrier, Hmp31. Lastly, hydrogenosomes possess a pathway for the assembly of C-tail-anchored proteins into their outer membrane with several new tail-anchored proteins being identified. These results show that hydrogenosomes and mitochondria share common core membrane components required for protein import and metabolite exchange; however, they also reveal remarkable differences that reflect the functional adaptation of hydrogenosomes to anaerobic conditions and the peculiar evolutionary history of the Excavata group. 相似文献

Discrimination of foreign eggs is one of the most studied aspects of host defences against avian brood parasites. Although many factors affecting host egg‐recognition processes have already been evaluated, only a few attempts have been made to test the importance of light conditions in microhabitats of host nests. Here, we examined whether the objectively measured nest light environment affects great reed warbler (Acrocephalus arundinaceus) responses towards real common cuckoo (Cuculus canorus) eggs. More specifically, we predicted that parasitic eggs will be rejected with a lower frequency from nests placed in darker conditions than those in lighter conditions. However, we found no effect of the ambient light on egg‐rejection behaviour alone, but the photosynthetically active radiation exhibited a positive interactive effect with chromatic contrast between cuckoo and host eggs. Most rejection events were accomplished when cuckoo eggs of poor mimicry were laid in well‐lit nests. Our study suggests that this phenomenon may have important implications for the evolution of egg mimicry and host egg discrimination. We encourage further testing of the light environment hypothesis in other host species breeding in variable nest microhabitats and light conditions. 相似文献

Traits related to seed dispersal, clonality and bud bank affect the success or failure of plant species. Using data from 13 successional seres in various human-made habitats the spectra of traits associated with dispersal and persistence were compared to determine the traits that can be used to predict the occurrence of particular plant species at each stage in a succession and how the importance of these traits changes over time. Differences in the traits of species associated with primary and secondary successions were also studied. Species with seeds that are dispersed by water (hydrochory) decreased in abundance during the course of succession. Species with a splitting main root, monocyclic and dicyclic shoots also decreased in abundance. Species capable of forming a potential below-ground bud bank, hypogeogenous rhizome and retaining a long-term connection with clonal offspring increased in abundance. The results indicate that seed dispersal is more important in determining the species composition in the early stages of succession whereas bud banks and clonal traits are more important in the later stages and for colonizing a locality. Primary and secondary seres did not remarkably differ in the trait spectra of the species present indicating that these trends occur in both types of succession. 相似文献

Ecological communities and their response to environmental gradients are increasingly being described by various measures of trait composition. Aggregated trait averages (i.e. averages of trait values of constituent species, weighted by species proportions) are popular indices reflecting the functional characteristics of locally dominant species. Because the variation of these indices along environmental gradients can be caused by both species turnover and intraspecific trait variability, it is necessary to disentangle the role of both components to community variability. For quantitative traits, trait averages can be calculated from ‘fixed’ trait values (i.e. a single mean trait value for individual species used for all habitats where the species is found) or trait values for individual species specific to each plot, or habitat, where the species is found. Changes in fixed averages across environments reflect species turnover, while changes in specific traits reflect both species turnover and within‐species variability in traits. Here we suggest a practical method (accompanied by a set of R functions) that, by combining ‘fixed’ and ‘specific averages’, disentangles the effect of species turnover, intraspecific trait variability, and their covariation. These effects can be further decomposed into parts ascribed to individual explanatory variables (i.e. treatments or environmental gradients considered). The method is illustrated with a case study from a factorial mowing and fertilization experiment in a meadow in South Bohemia. Results show that the variability decomposition differs markedly among traits studied (height, Specific Leaf Area, Leaf N, P, C concentrations, leaf and stem dry matter content), both according to the relative importance of species turnover and intraspecific variability, and also according to their response to experimental factors. Both the effect of intraspecific trait variability and species turnover must be taken into account when assessing the functional role of community trait structure. Neglecting intraspecific trait variability across habitats often results in underestimating the response of communities to environmental changes. 相似文献

PurposeTo evaluate the opportunities of single photon emission tomography/computerized tomography (SPECT-CT) for localization of axillary sentinel lymph nodes (ASLNs) and subsequent radiotherapy planning in women with early breast cancer.Material and methodsIndividual topography of ASLN was determined in 151 women with clinical T1-2N0M0 breast cancer. SPECT-CT visualization of ASLNs was initiated 120 min after intra-peritumoral injection of 99mTc-radiocolloids. Doses absorbed by virtual ASLNs after the whole breast irradiation with standard and extended tangential fields were calculated on a treatment planning station.ResultsSPECT-CT demonstrated a large variability of ASLN localization. They were detected in the central subgroup in 94 (61%) patients, in pectoral – in 77 (51%), and in interpectoral – in 4 (3%) patients. Sentinel lymph nodes “lying on the chest” were revealed in 35 (23%) cases.We found that with standard tangential fields coverage of ASLNs was obtained only in 20% of evaluated women. Extended tangential fields can effectively irradiate ASLNs localized in all axillary sub-regions with the exception of ASLNs “lying on the chest”.ConclusionSPECT-CT mapping of ASLNs in women with cT1-2N0M0 breast cancer reveals their variable localization.
