Broadening prospects of using single-walled carbon nanotubes (SWNTs) in medicine and biotechnology raise the concerns about both their toxicity and the mechanisms of biodegradation and elimination from the body. SWNTs biodegradation as a result of catalytic activity of myeloperoxidase (MPO) was shown in the isolated MPO system as well as in the suspension of neutrophils [Kagan V.E. et al., 2010]. In the present study we analyzed the ability of different MPO-produced oxidants to oxidize and to degrade SWNTs. The comparison of the ability of various peroxidases to degrade SWNTs in vitro revealed that myeloperoxidase, due to its ability to produce hypochlorite, and lactoperoxidase, due to its ability to produce hypobromite, are extremely efficient in the degrading of carbon nanotubes. The biodegradation of SWNTs in the model system can also be induced by free radicals generated as a result of heme degradation and, to a lesser extent, by active oxoferryl intermediates of peroxidases. Our experiments showed that in the presence of blood plasma, peroxidase intermediates or free radical products of heme degradation were unable to initiate biodegradation of carbon nanotubes, only the generation of hypochlorite by MPO can cause the biodegradation of carbon nanotubes in vivo. At high concentrations, hypochlorite caused decrease in optical absorbance of plasma-containing SWNTs suspension, which is indicative of the nanotube degradation. Our results unambiguously suggest that hypochlorite can serve as a main oxidizing agent to modify and degrade nanotubes at the sites of inflammation and in phagosomes.  相似文献

In this work, the diameters of protein complexes formed upon interaction of ceruloplasmin (CP) with lactoferrin (LF) and myeloperoxidase (MPO) were determined. Gage dependence of the diameter of protein particles (myoglobin, albumin, LF, CP, MPO, aldolase, ferritin) on their molecular mass logarithm was calculated. The diameter of a complex formed upon mixing CP and LF was 8.4 nm, which is in line with the radius of gyration obtained previously when the 1CP-1LF complex was studied by small-angle X-ray scattering. The diameter of a complex formed upon interaction of CP with MPO is 9.8 nm, corresponding to the stoichiometry 2CP: 1MPO. The diameter of a complex formed when LF is added to the 2CP-1MPO complex is 10.7 nm. The latter is consistent with the notion of a pentameric structure 2LF-2CP-1MPO with molecular mass of about 585 kDa.  相似文献

Systemic investigation of the mechanisms that control locomotions in the man provides elaboration of some problems concerning properties and construction of the locomotory apparatus, especially morphometric characteristics of the muscles--length and shoulder of the thrust force. Various methods for determining length and shoulder of the thrust force are considered. The first method is based on substituting the muscles for mathematical or mechanical model. The second method makes it possible to define the degree of the muscle elongation by means of measuring the distance between the ends of the sectioned tendon; it includes regression equations, connecting a relative elongation of the muscles with the angle change in the joints. As a result of perfection of the previously known methods, mathematical models are made, owing to them it is possible to calculate morphometric characteristics of 8 muscles in the lower extremities using computer facilities. As an example, results of calculations, by means of various methods, morphometric characteristics of the gastrocnemius muscle are presented. They are obtained at registration of the kinematics of the run with a maximal speed. Satisfactive coincidence of the results is obtained.  相似文献

The synthesis of (+)-usnic acid derivatives is described. The derivatives contain one or two cyano groups, connected to the acetophenone fragment of dibenzofuran core by linkers of different length and character, or some other modifications. The influence of these compounds on the activity of recombinant human tyrosyl-DNA phosphodiesterase 1 and MCF-7 tumor cells’ viability has been estimated. The data indicate a distinct dependence of functional characteristics of the compounds on their structure.  相似文献
