We report a new cellular mechanism of rod photoreceptor adaptation in vivo, which is triggered by daylight levels of illumination. The mechanism involves a massive light-dependent translocation of the photoreceptor-specific G protein, transducin, between the functional compartments of rods. To characterize the mechanism, we developed a novel technique that combines serial tangential cryodissection of the rat retina with Western blot analysis of protein distribution in the sections. Up to 90% of transducin translocates from rod outer segments to other cellular compartments on the time scale of tens of minutes. The reduction in the transducin content of the rod outer segments is accompanied by a corresponding reduction in the amplification of the rod photoresponse, allowing rods to operate in illumination up to 10-fold higher than would otherwise be possible. 相似文献

The biological role of exogenous carbon dioxide during substrate assimilation with a various degree of reductivity is evaluated. The investigation of metabolic pathways of carbon dioxide incorporation into the metabolic processes of methaneoxidizing bacteria shows that the HCO3- ion assimilation is catalyzed by phosphoenolpyruvate carboxylase and in certain strains also by the key enzyme of autotrophic pathway of the carbon dioxide assimilation, ribulose-1,5-diphosphate carboxylase. The theoretical calculations and experimental studies indicate that exogenous carbon dioxide is a necessary participant of the metabolic processes of methane or methanol assimilation. It is also an acceptor of the excess electrons of these compounds. It is the degree of reductivity of the substrate metabolized that determines the activity of the exogenous carbon dioxide fixation by microorganisms. The carbon dioxide fixation by heterotrophic microorganisms must be considered, therefore, as a process which is mostly due to the elementary composition of the source of carbon under conversion. 相似文献

Summary The electric properties of the bilayer lecithin membranes have been studied in the presence of the antibiotic nigericin. When the antibiotic concentration is about 10–6
m the conductivity of the BLM is increased up to 10–7 ohm–1 cm–2. The potassium ion concentration gradient gives rise to a transmembrane potential of the order of 40 mV per 10-fold concentration gradient with the side of the higher potassium concentration negative. The transmembrane potential produced by the hydrogen ion concentration gradient is a function of the potassium ion concentration which is equal on both sides of the membrane. For low potassium ion concentrations the hydrogen potential has the expected polarity with the solution having higher concentration of protons negative. For potassium ion concentrations exceeding 0.03m the hydrogen potential has the reverse polarity. This unexpected result cannot be accounted for in terms of the available simple hypotheses about the charge transport mechanism for nigericin in BLM. In order to account for the experimental results obtained, a theoretical approach has been developed based on the assumption that charge is transported across the membrane by nigericin dimers. The theoretical predictions are in satisfactory agreement with the experimental results. The model also yields some predictions which may be verified in future experiments. 相似文献

In the Ross Sea and Amundsen Sea, four representatives of the genus Paralepidapedon—Paralepidapedon cf. dubium Prudhoe et Bray 1973 sensu Sokolov et Gordeev 2013, P. lepidum (Gaevskaya et Rodyuk 1988), Paralepidapedon sp., and P. variabile sp. n.—were found in demersal fishes Muraenolepis marmorata and Macrourus whitsoni. Paralepidapedon variabile sp. n. is described from Muraenolepis marmorata in the Amundsen Sea. Paralepidapedon variabile sp. n. differs from other species of the genus Paralepidapedon by the position of the anterior border of the vitellarium at the level of the anterior edge of the ventral sucker or genital pore and by the highly variable shape of the testes: from roundish with a smooth edge to sinuate–lobate. Due to the lack of complete understanding of metabolic networks and reaction pathways, establishing a universal mechanistic model for mammalian cell culture processes remains a challenge. Contrarily, data-driven approaches for modeling these processes lack extrapolation capabilities. Hybrid modeling is a technique that exploits the synergy between the two modeling methods. Although mammalian cell cultures are among the most relevant processes in biotechnology and indeed looks ideal for hybrid modeling, their application has only been proposed but never developed in the literature. This study provides a quantitative assessment of the improvement brought by hybrid models with respect to the state-of-the-art statistical predictive models in the context of therapeutic protein production. This is illustrated using a dataset obtained from a 3.5 L fed-batch experiment. With the goal to robustly define the process design space, hybrid models reveal a superior capability to predict the time evolution of different process variables using only the initial and process conditions in comparison to the statistical models. Hybrid models not only feature more accurate prediction results but also demonstrate better robustness and extrapolation capabilities. For the future application, this study highlights the added value of hybrid modeling for model-based process optimization and design of experiments. 相似文献
