Summary The corticotropin-releasing factor (CRF)-containing neurons were investigated in the brain of the domestic fowl by means of the peroxidase-antiperoxidase technique at the light-microscopic level. The detection of CRF-immunoreactivity was facilitated by silver intensification. CRF-containing perikarya were found in the paraventricular, preoptic and mammillary nuclei of the hypothalamus and in some extrahypothalamic areas (nuclei dorsomedialis and dorsolateralis thalami, nucleus accumbens septi, lobus parolfactorius, periaqueductal gray of the mesencephalon, nucleus oculomotorius ventralis). Immunoreactive nerve fibers and terminals were demonstrated in the external zone of the median eminence and the organum vasculosum of the lamina terminalis. These results indicate that an immunologically demonstrable CRF-neurosecretory system also exists in the avian central nervous system.  相似文献

The stable intermediates formed in the reaction of cytosine, cytidine and adenosine with chloracetaldehyde were isolated. The -CH2CH/OH/- bridge between the exo and endo nitrogen atoms of the parent base was found in these compounds by means of PMR spectroscopy. Their acid-induced dehydration resulted in formation of appropriate ethenoderivatives. The rate constants of the intermediate formation and its dehydration were found to be 38x10(-4) and 47x10(-4) /min-1/ for adenosine, and 33x10(-4) and 10x10(-4) /min-1/ for cytidine. The PH range of 4.5--5.0 was found to be optimum for both adenosine and cytidine reactions. The quantitative modification of these two nucleosides in the presence of guanosine may be achieved with high selectivity only at a low pH of 3.0--4.0 N6-methyladenosine and N4-methylcytidine react quantitatively with chloroacetaldehyde and the reaction rate is higher than in the case of the parent nucleosides. The structure of the reaction products was assigned on the basis of PMR spectroscopy.  相似文献

In order to study the characteristic signs of an induced estrus in beef cattle maintained in the tropics, 31 animals, 16 pure Charolais and 15 Charolais/Brahman cross, were continuously observed during the first 100 hours following prostaglandin injection. The animals were pastured in a field of approximately 3 hectares. A highly significant relationship (P < 0.01) seemed to exist between behavioural tendencies and the interval following the onset of estrus. Behaviour included butting, attempts to mount, smelling of genitalia (active) and allowing to be mounted and have genitalia smelled (passive). No consistant pattern of sexual activity could be established since some animals were more active than others. Mounts were observed to occur between 6 and 9 in the morning (33%), 9 and 12 at night (16%) and midnight and three in the morning (13%). Differences existed between the Charolais and the Brahman cross with respect to the number of mounts (P < 0.05) and in the response to an induced estrus (P < 0.05). No correlation was found between grazing habits and sexual activity, the former expressed as the number of animals standing up during each hour.  相似文献

The fertility-inhibiting effects of long-term (8 weeks) consumption of maize infected with a fungus producing F2 toxin (zearalenone) was studied in adult male and female albino rats. The fertility rate was further decreased by 25-30% if the animals were kept on contaminated diet up to 14 weeks. The gonadal weight was decreased, follicular maturation and spermatogenesis were disturbed. The toxic diet consumed by mothers during pregnancy and lactation induced permanent changes in reproductive organs, disorders in vaginal cyclicity and disturbed fertility in the offspring. Neonatal administration of purified F2 toxin provoked similar changes. It is suggested that this fungal toxin may cause sterility syndrome in the offspring, similar to that produced by androgen or estrogen administration.  相似文献

The paper describes the ultrastructural changes of the sympathetic neurons following guanethidine administration in mice. The main characteristics changes, after prolonged administration (1-10 weeks) of guanethidine (Ismelin, Ciba-Geigy, Basel, Switzerland) at doses of 30-50 mg/kg/day i.p. and i.m. were: (1) Mitochondrial damages: swelling and deformation of the mitochondria, with disruption and dispersion of the cristae and condensation or dissolution of the matrix. Membranolysis of the outer and inner mitochondrial membranes. (2) Neural processes: swelling and disorganization of the internal architecture in the postganglionic axonal and dendritic synaptic varicosities. (3) Adrenergic vesicles: disappearance of the granular vesicles in the early stage of the experiment, increase of the coated vesicles in the later stage of treatment. The effect of guanethidine is specific for the postganglionic adrenergic neurons, but the observed destruction is not caused solely by guanethidine; other chemical substances induce the same or similar degeneration in the sympathetic ganglion.  相似文献

Triglyceride-rich lipoproteins from adult rat plasma were labelled in vivo with 3H in the esterified fatty acids and 14C in the labelled glyceride glycerol of neutral lipids by injecting i.v. sodium 9-10 (n)-[3H] palmitate and [U-14C] glycerol, after which the prelabelled lipoproteins were purified by ultracentrifugation and dialysis. The lipoproteins were incubated in vitro, in the presence or not of heparin, with pieces of epididymal fat pads or isolated adipocytes from fed rats. The disappearance of both [3H]- and [14C] lipids from the media was greater when incubations were performed with adipocytes than with fat-pad pieces and it increased with heparin in both preparations. More 3H-label than 14C was found in the tissue lipids, a higher percentage being present in adipocytes than in fat-pad pieces, and the amount of label in tissue lipids was always enhanced by heparin. Some 14C-label appeared as esterified fatty acids in both tissue preparations and it also was enhanced by the presence of heparin. These findings are in agreement with the recognized influence of heparin on the release of lipoprotein lipase and show the direct relationship between heparin action and tissue ability to take up products of lipoprotein triglyceride breakdown. They also demonstrate the ability of adipose tissue to metabolize glycerol coming from the hydrolysis of lipoprotein glycerides.  相似文献

Hoyle and Wickramasinghe have recently suggested that life may have originated in cometary nuclei rather than directly on Earth. Even though comets are known to contain substantial amounts of organic compounds which may have contributed to the formation of biochemical molecules on the primitive Earth, it is doubtful that the process of chemical evolution has proceeded in comets beyond the stage that has occurred in carbonaceous chondrites. Some of the arguments which do not favor the occurrence of biopoesis in comets are:
  1. A large layer of cometary ices is ablated from the nucleus' surface each time the comet passes through perihelion, so that essentially most of the organic products on the surface would be sublimed, blown off or polymerized.
  2. Because of the low temperatures of the cometary ices, polymers formed on one perihelion passage would not migrate deep enough into the nucleus to be preserved before they would be ablated away by the next perihelion passage.
  3. In the absence of atmosphere, and discrete liquid and solid surfaces, it is difficult to visualize the synthesis of key life molecules, such as oligopeptides, oligonucleotides and phospholipids by condensation and dehydration reactions as is presumed to have occurred in the evaporating ponds of the primitive Earth.
  4. Observations suggest that cometary nuclei have a rather weak structure. Hence, the low central pressures in comets combined with the high vapor pressures of cometary ices at the melting point of water ice, suggest that a liquid core is not a tenable structure. Yet, even if a cometary nucleus is compact enough to hold a liquid core and a transient liquid water environment was provided by the decay of26Al, the continuous irradiation in water of most of the biologically relevant polymers would have hydrolyzed and degraded them.
  5. Needless to say that the effects of radiation on self-replicating systems would also have caused the demise of any life forms which may have appeared under any circumstances.
  6. Concerning viruses, the high specificity of host-parasite relationships and their coevolutionary lines of descent, rule out a cometary origin for them.
In summary, the view that life originated in comets is untenable in the light of all the available evidence.  相似文献
