The addition of nocturnal, Hoplias malabaricus, and diurnal, Crenicichla alta, predatory fishes downstream of barrier waterfalls increases predation threat for a killifish, Rivulus hartii, in Trinidadian streams. We hypothesized that the diel patterning of predation risk would affect prey movement rates, and tested this hypothesis by comparing movement in river sites/zones containing both the nocturnal and diurnal predator with movement in river sites/zones containing only the nocturnal taxon. We evaluated this prediction in the framework of an intermediate threat hypothesis (ITH) that holds that movement will be highest at some intermediate level of threat. We marked prey fish in study sites in two watersheds of a river, each with waterfalls that divided the river into three zones: a predator absent zone (P0), a zone with one nocturnal predator (P1), and a zone with one nocturnal and one diurnal predator (P2), and tested the ITH prediction that movement will be ordered as P0<P1>P2. The single predator promoted longitudinal movement by Rivulus (P0<P1), while zones with the two predators retarded movement for small Rivulus (P1>P2) as predicted by the ITH. However, movement by larger, less vulnerable Rivulus remained elevated (P1=P2 or P2>P1). A displacement experiment in each zone found that threat tended to reduce the probability of a displaced fish reaching home, but the two predator zones did not differ from one another in their effect on this probability. Hence, the prediction that predator activity over the full 24 h diel cycle would retard movement, P2<P1, was not supported with respect to homing. Because habitat and predator communities change predictably from headwater streams to larger rivers in many lotic ecosystems, we present a conceptual model for predicting fish movement behavior along this continuum. The model posits an important role for predation threat, and the size and spacing of refuge patches, suggesting that human alterations of these factors will affect the natural movement of fish in streams. 相似文献

We sought to test the hypothesis that stomatal development determines the timing of gas exchange competency, which then influences leaf temperature through transpirationally driven leaf cooling. To test this idea, daily patterns of gas exchange and leaflet temperature were obtained from leaves of two distinctively different developmental stages of smooth sumac (Rhus glabra) grown in its native habitat. Juvenile and mature leaves were also sampled for ultrastructural studies of stomatal development. When plants were sampled in May-June, the hypothesis was supported: juvenile leaflets were (for part of the day) from 1.4 to 6.0 degrees C warmer than mature leaflets and as much as 2.0 degrees C above ambient air temperature with lower stomatal conductance and photosynthetic rates than mature leaflets. When measurements were taken from July to October, no significant differences were observed, although mature leaflet gas exchange rates declined to the levels of the juvenile leaves. The gas exchange data were supported by the observations that juvenile leaves had approximately half the number of functional stomata on a leaf surface area basis as did mature leaves. It was concluded that leaf temperature and stage of leaf development in sumac are strongly linked with the higher surface temperatures observed in juvenile leaflets in the early spring possibly being involved in promoting photosynthesis and leaf expansion when air temperatures are cooler. 相似文献

Background
The use of manipulation has long been advocated in the treatment of chronic obstructive pulmonary disease (COPD), but few randomized controlled clinical trials have measured the effect of manipulation on pulmonary function. In addition, the effects of individual manipulative techniques on the pulmonary system are poorly understood. Therefore, the purpose of this study was to determine the immediate effects of four osteopathic techniques on pulmonary function measures in persons with COPD relative to a minimal-touch control protocol.Methods
Persons with COPD aged 50 and over were recruited for the study. Subjects received five, single-technique treatment sessions: minimal-touch control, thoracic lymphatic pump (TLP) with activation, TLP without activation, rib raising, and myofascial release. There was a 4-week washout period between sessions. Protocols were given in random order until all five techniques had been administered. Pulmonary function measures were obtained at baseline and 30-minutes posttreatment. For the actual pulmonary function measures and percent predicted values, Wilcoxon signed rank tests were used to test within-technique changes from baseline. For the percent change from baseline, Friedman tests were used to test for between-technique differences.Results
Twenty-five subjects were enrolled in the study. All four tested osteopathic techniques were associated with adverse posttreatment changes in pulmonary function measures; however, different techniques changed different measures. TLP with activation increased posttreatment residual volume compared to baseline, while TLP without activation did not. Side effects were mild, mostly posttreatment chest wall soreness. Surprisingly, the majority of subjects believed they could breathe better after receiving osteopathic manipulation.Conclusion
In persons with COPD, TLP with activation, TLP without activation, rib raising, and myofascial release mildly worsened pulmonary function measures immediately posttreatment relative to baseline measurements. The activation component of the TLP technique appears to increase posttreatment residual volume. through interactions with muscarinic receptors. Changes in muscarinic acetylcholine receptor (mAChR) have been implicated
in the pathophysiology of many major diseases of the central nervous system (CNS). Previous reports from our laboratory on
streptozotocin (STZ) induced diabetic rats showed down regulation of muscarinic M1 receptors in the brainstem, hypothalamus,
cerebral cortex and pancreatic islets. In this study, we have investigated the changes of acetylcholine esterase (AChE) enzyme
activity, total muscarinic and muscarinic M1 receptor binding and gene expression in the corpus striatum of STZ – diabetic
rats and the insulin treated diabetic rats. The striatum, a neuronal nucleus intimately involved in motor behaviour, is one
of the brain regions with the highest acetylcholine content. ACh has complex and clinically important actions in the striatum
that are mediated predominantly by muscarinic receptors. We observed that insulin treatment brought back the decreased maximal
velocity (Vmax) of acetylcholine esterase in the corpus striatum during diabetes to near control state. In diabetic rats there was a decrease
in maximal number (Bmax) and affinity (Kd) of total muscarinic receptors whereas muscarinic M1 receptors were increased with decrease in affinity in diabetic rats.
We observed that, in all cases, the binding parameters were reversed to near control by the treatment of diabetic rats with
insulin. Real-time PCR experiment confirmed the increase in muscarinic M1 receptor gene expression and a similar reversal
with insulin treatment. These results suggest the diabetes-induced changes of the cholinergic activity in the corpus striatum

Metastatic renal cell carcinoma (RCC) is highly resistant to conventional systemic treatments, including chemotherapy, radiotherapy and hormonal therapies. Previous studies have shown over-expression of EGFR is associated with high grade tumors and a worse prognosis. Recent studies suggest anticancer therapies targeting the EGFR pathway have shown promising results in clinical trials of RCC patients. Therefore, characterization of the level and localization of EGFR expression in RCC is important for target-dependent therapy. In this study, we investigated the clinical significance of cellular localization of EGFR in human normal renal cortex and RCC. RCC and adjacent normal kidney tissues of 63 patients were obtained for characterization of EGFR expression. EGFR protein expression was assessed by immunohistochemistry on a scale from 0 to 300 (percentage of positive cells × staining intensity) and Western blotting. EGFR membranous staining was significantly stronger in RCC tumors than in normal tissues (P < 0.001). In contrast, EGFR cytoplasmic staining was significantly higher in normal than in tumor tissues (P < 0.001). The levels of membranous or cytoplasmic EGFR expression in RCC tissues were not correlated with sex, tumor grade, TNM stage or overall survival (P > 0.05). These results showed abundant expression of membranous EGFR in RCC, and abundant expression of cytoplasmic EGFR in normal tissues. EGFR expression in RCC was mostly located in the cell membrane, whereas the EGFR expression in normal renal tissues was chiefly seen in cytoplasm. Polarized radial glia are crucial to the formation of the cerebral cortex. They serve as neural progenitors and as guides for neuronal placement in the developing cerebral cortex. The maintenance of polarized morphology is essential for radial glial functions, but the extent to which the polarized radial glial scaffold is static or dynamic during corticogenesis remains an open question. The developmental dynamics of radial glial morphology, inter-radial glial interactions during corticogenesis, and the role of the cell polarity complexes in these activities remain undefined. Here, using real-time imaging of cohorts of mouse radial glia cells, we show that the radial glial scaffold, upon which the cortex is constructed, is highly dynamic. Radial glial cells within the scaffold constantly interact with one another. These interactions are mediated by growth cone-like endfeet and filopodia-like protrusions. Polarized expression of the cell polarity regulator Cdc42 in radial glia regulates glial endfeet activities and inter-radial glial interactions. Furthermore, appropriate regulation of Gsk3 activity is required to maintain the overall polarity of the radial glia scaffold. These findings reveal dynamism and interactions among radial glia that appear to be crucial contributors to the formation of the cerebral cortex. Related cell polarity determinants (Cdc42, Gsk3) differentially influence radial glial activities within the evolving radial glia scaffold to coordinate the formation of cerebral cortex. 相似文献

Glucose homeostasis in humans is an important factor for the functioning of nervous system. Hypoglycemia and hyperglycemia is found to be associated with central and peripheral nerve system dysfunction. Changes in acetylcholine receptors have been implicated in the pathophysiology of many major diseases of the central nervous system (CNS). In the present study we showed the effects of insulin induced hypoglycemia and streptozotocin induced diabetes on the cerebellar cholinergic receptors, GLUT3 and muscle cholinergic activity. Results showed enhanced binding parameters and gene expression of Muscarinic M1, M3 receptor subtypes in cerebellum of diabetic (D) and hypoglycemic group (D + IIH and C + IIH). α7nAchR gene expression showed a significant upregulation in diabetic group and showed further upregulated expression in both D + IIH and C + IIH group. AchE expression significantly upregulated in hypoglycemic and diabetic group. ChAT showed downregulation and GLUT3 expression showed a significant upregulation in D + IIH and C + IIH and diabetic group. AchE activity enhanced in the muscle of hypoglycemic and diabetic rats. Our studies demonstrated a functional disturbance in the neuronal glucose transporter GLUT3 in the cerebellum during insulin induced hypoglycemia in diabetic rats. Altered expression of muscarinic M1, M3 and α7nAchR and increased muscle AchE activity in hypoglycemic rats in cerebellum is suggested to cause cognitive and motor dysfunction. Hypoglycemia induced changes in ChAT and AchE gene expression is suggested to cause impaired acetycholine metabolism in the cerebellum. Cerebellar dysfunction is associated with seizure generation, motor deficits and memory impairment. The results shows that cerebellar cholinergic neurotransmission is impaired during hyperglycemia and hypoglycemia and the hypoglycemia is causing more prominent imbalance in cholinergic neurotransmission which is suggested to be a cause of cerebellar dysfunction associated with hypoglycemia. 相似文献
