The ability of beta2-microglobulin-deficient mice (B6.beta2micro(o)) mice to reject syngeneic and major histocompatability (MHC) class I-deficient tumor grafts was examined with a view to determining residual cytotoxic activities that exist in these mice. In particular, the cytotoxic activities of NK cells and CD8(+) cytotoxic T lymphocytes (CTL) reactive against self-MHC class I were assessed using a variety of gene-targeted mice. The creation of mice doubly deficient for perforin and beta2micro (B6.P(o).beta2micro(o)) enabled the determination that perforin was responsible for the cytotoxic activity of NK cells and CD8(+) CTL reactive against self-MHC class I. Dependence on perforin function was demonstrated for the cytotoxicity of these effectors in vitro and for the ability of these effectors to reject a variety of tumors in vivo.  相似文献

To evaluate breastfed infants' responses to scented objects, we videotaped the facial and bodily reactions of sixty-three infants as they explored, in succession, three toys that were identical in appearance but different in their characteristic odor. Two of the toys were scented with odorants previously shown to be transmitted to human milk, one with ethanol and the other with vanilla, whereas the third toy was unscented. Each videotape was subjected to frame-by-frame analysis to measure a variety of behaviors that are considered either to be exploratory in nature in that they lead to perceptual information about the object or to reflect the infants' hedonic reaction. Analyses of these behaviors revealed that the infants looked more and vocalized less in the presence of the vanilla-scented toy and spent less time manipulating the ethanol-scented toy when compared with the unscented toy. Moreover, differential exposure to the odors of ethanol and vanilla, as indicated by differential consumption of alcohol by a parent or use of vanilla-scented product by the mother, was related to differential responses to these odors. These findings suggest that human infants are able to detect and retain information about the chemical features of their environment.   相似文献

Rat thyroid contains thyrotropin-releasing hormone (TRH) and TRH-like peptides which react with TRH antisera. We have identified the TRH-like peptides in the thyroid and examined whether their levels are influenced by thyroid status. The peptides were extracted from the thyroid glands of five hyperthyroid rats and purified by ion-exchange chromatography on SP-Sephadex C25 and reversed-phase high performance liquid chromatography. The principal TRH-immunoreactive component exhibited the same retention on HPLC as synthetic pGlu-Glu-Pro amide and a secondary component corresponded to synthetic pGlu-Phe-Pro amide. In agreement with these assignments the main peptide was shown to be acidic when chromatographed on DEAE-Sephadex A25 and the second peptide neutral. The levels of TRH and TRH-like peptides in the thyroid were investigated in hyper-, hypo- and euthyroid rats. Hyperthyroidism was induced by chronic subcutaneous administration of triiodothyronine (T3) and hypothyroidism was produced by addition of propylthiouracil (PTU) to the drinking water. The amounts of the peptides were determined by radioimmunoassay with a TRH-antiserum, carried out after extraction from the tissues and purification by ion exchange chromatography. The mean concentration of TRH-like peptides in the thyroids of the hyperthyroid rats was 95.5+/-25.5 pmol/g, the mean concentration in the hypothyroid rats was 11.7+/-3.4 pmol/g, and in the euthyroid rats 17.6+/-3.2 pmol/g. The concentrations of TRH were less influenced by thyroid status: the values in hyper-, hypo- and euthyroid rats were 47.5+/-9.4, 42.1+/-6.3, and 17.2+/-1.6 pmol/g respectively. The results show that the levels of the TRH-like peptides in rat thyroid are highly sensitive to thyroid status, suggesting a possible involvement in thyroid regulation.  相似文献

Afforestation with short‐rotation coppice (SRC) willow plantations for the purpose of producing bioenergy feedstock was contemplated as one potential climate change mitigation option. The objectives of this study were to assess the magnitude of this mitigation potential by addressing: (i) the land area potentially available for SRC systems in the province of Saskatchewan, Canada; (ii) the potential biomass yields of SRC plantations; and (iii) the carbon implications from such a large‐scale afforestation program. Digital soils and land‐use data were used to identify, map, and group into clusters of similar polygons 2.12 million hectares (Mha) of agriculturally marginal land that was potentially suitable for willow in the Boreal Plains and Prairies ecozones in Saskatchewan. The Physiological Principles in Predicting Growth (3PG) model was calibrated with data from SRC experiments in Saskatchewan, to quantify potential willow biomass yields, and the Carbon Budget Model of the Canadian Forest Sector (CBM‐CFS3), was used to simulate stand and landscape‐level C fluxes and stocks. Short‐rotation willow plantations managed in 3 year rotations for seven consecutive harvests (21 years) after coppicing at Year 1 produced about 12 Mg ha?1 yr?1 biomass. The more significant contribution to the C cycle was the cumulative harvest. After 44 years, the potential average cumulative harvested biomass C in the Prairies was 244 Mg C ha?1 (5.5 Mg C ha?1 yr?1) about 20% higher than the average for the Boreal Plains, 203 Mg C ha?1 (4.6 Mg C ha?1 yr?1). This analysis did not consider afforestation costs, rate of establishment of willow plantations, and other constraints, such as drought and disease effects on biomass yield. The results must therefore be interpreted as a biophysical mitigation potential with the technical and economic potential being both lower than our estimates. Nevertheless, short‐rotation bioenergy plantations offer one potential mitigation option to reduce the rate of CO2 accumulation in the earth's atmosphere and further research is needed to operationalise such a mitigation effort.  相似文献
