Ecotypic variation among populations may become associated with widespread genomic differentiation, but theory predicts that this should happen only under particular conditions of gene flow, selection and population size. In closely related species, we might expect the strength of host‐associated genomic differentiation (HAD) to be correlated with the degree of phenotypic differentiation in host‐adaptive traits. Using microsatellite and Amplified Fragment Length Polymorphism (AFLP) markers, and controlling for isolation by distance between populations, we sought HAD in two congeneric species of butterflies with different degrees of host plant specialization. Prior work on Euphydryas editha had shown strong interpopulation differentiation in host‐adapted traits, resulting in incipient reproductive isolation among host‐associated ecotypes. We show here that Euphydryas aurinia had much weaker host‐associated phenotypic differentiation. Contrary to our expectations, we detected HAD in Euphydryas aurinia, but not in E. editha. Even within an E. aurinia population that fed on both hosts, we found weak but significant sympatric HAD that persisted in samples taken 9 years apart. The finding of significantly stronger HAD in the system with less phenotypic differentiation may seem paradoxical. Our findings can be explained by multiple factors, ranging from differences in dispersal or effective population size, to spatial variation in genomic or phenotypic traits and to structure induced by past histories of host‐adapted populations. Other infrequently measured factors, such as differences in recombination rates, may also play a role. A highly reproducible and robust cell-based high-throughput screening (HTS) assay was adapted for screening of small molecules for antiviral activity against influenza virus strain A/Vietnam/1203/2004 (H5N1). The NIH Molecular Libraries Small Molecule Repository (MLSMR) Molecular Libraries Screening Centers Network (MLSCN) 100,000-compound library was screened at 50 μM. The "hit" rate (>25% inhibition of the viral cytopathic effect) from the single-dose screen was 0.32%. The hits were evaluated for their antiviral activity, cell toxicity, and selectivity in dose-response experiments. The screen yielded 5 active compounds (SI value >3). One compound showed an SI(50) value of greater than 3, 3 compounds had SI values ranging from greater than 14 to 34, and the most active compound displayed an SI value of 94. The active compounds represent 2 different classes of molecules, benzoquinazolinones and thiazoloimidazoles, which have not been previously identified as having antiviral/anti-influenza activity. These molecules were also effective against influenza A/California/04/2009 virus (H1N1) and other H1N1 and H5N1 virus strains in vitro but not H3N2 strains. Real-time qRT-PCR results reveal that these chemotypes significantly reduced M1 RNA levels as compared to the no-drug influenza-infected Madin Darby canine kidney cells. 相似文献

The primary targets for SARS-CoV infection are the epithelial cells in the respiratory and intestinal tract. The angiotensin-converting enzyme 2 (ACE-2) has been identified as a functional receptor for SARS-CoV. ACE-2 has been shown to be expressed at the apical domain of polarized Calu-3 cells. In this report, interferon alfacon 1 was examined for inhibitory activities against SARS-CoV on human lung carcinoma epithelial Calu-3 cell line and the other three African green monkey kidney epithelial cell lines. Interferon alfacon 1 demonstrated significant antiviral activity in neutral red uptake assay and virus yield reduction assay. The data might provide an important insight into the mechanism of pathogenesis of SARS-CoV allowing further development of antiviral therapies for treating SARS infections. 相似文献

There are no specific approved drugs or vaccines for the treatment or prevention of infectious dengue virus and there are very few compounds known that inhibit the replication of this virus. This letter describes the concise synthesis of two uracil-based multifunctional compounds. One of these compounds (1) has strong activity against dengue virus. It also exhibits low activity against a few other RNA viruses, but is highly active against yellow fever virus, a related flavivirus. It is likely that the mechanism of action of the antiviral activity of this compound is through its inhibition of the enzyme, inosine monophosphate dehydrogenase (IMPDH). Molecular modeling studies reveal that the compound can have specific hydrogen bonding interactions with a number of amino acids in the active site of IMPDH, a stacking interaction with the bound natural substrate, IMP, and the ability to interfere with the binding of NAD+ with IMPDH, prior to the hydration step. 相似文献

Vanadium(III, IV, V)–chlorodipicolinate (dipic-Cl) complexes, including H[VIII(dipic-Cl)2] · 5H2O (V3dipic-Cl), VIVO(dipic-Cl)(H2O)2 (V4dipic-Cl) and K[VVO2(dipic-Cl)] (V5dipic-Cl), were prepared with the indicated oxidation states. Our aim was to evaluate the anti-diabetic effects of V3dipic-Cl, V4dipic-Cl and V5dipic-Cl in streptozotocin-induced diabetic rats. Vanadium complexes were orally administered to diabetic rats at concentrations
of 0.1–0.3 mg/ml in the drinking water. We found that vanadium–chlorodipicolinate (V–dipic-Cl) complexes at the concentration
of 0.1 mg/ml did not exhibit blood glucose-lowering effects when administered to diabetic rats for 20 days. However, the levels
of fasting blood glucose in diabetic rats were decreased after treatment with 0.3 mg/ml of V4dipic-Cl and V5dipic-Cl complexes for the following 20 days. Although administration of both V4dipic-Cl and V5dipic-Cl significantly lowered diabetic hyperglycemia, the vanadium intake from administration of V4dipic-Cl is nearly 1.5-fold greater compared to that of V5dipic-Cl. Treatment with the H2dipic-Cl ligand and all three V–dipic-Cl complexes significantly lowered serum cholesterol, while administration of the V5dipic-Cl complex lowered serum cholesterol significantly more than administration of the ligand alone. alone did not have an effect on serum triglyceride, while administration of the V4dipic-Cl and V5dipic-Cl significantly lowered the elevated serum triglyceride associated with diabetes. Oral administration of the ligand
and all V–dipic-Cl complexes did significantly lower diabetes elevated serum alkaline phosphatase. Treatment with H2dipic-Cl ligand and V4dipic-Cl and V5dipicCl significantly lowered diabetes elevated aspartate amino transferase. These results indicate that the health of the
treated animals did not seem to be further compromised compared to that of diabetic animals. In addition, oral administration
of H2dipic-Cl, V3dipic-Cl, V4dipic-Cl and V5dipic-Cl did not alter diabetic serum creatinine and blood urea nitrogen levels, suggesting no significant side effects of
vanadium treatment on renal functions at the dose of 0.3 mg/ml in diabetic rats. The results presented here suggest that the

Nitric oxide and prostaglandin E2 (PGE2play pivotal roles in both the pathogenesis of osteoarthritis and catabolic processes in articular cartilage. These mediators are influenced by both IL-1β and mechanical loading, and involve alterations in the inducible nitric oxide synthase (iNOS) and cyclo-oxygenase (COX)-2 enzymes. Nonrandom location of IS1 elements in the genomes of natural isolates of Escherichia coli 总被引:2,自引:0,他引:2
We have studied the spatial distribution of IS1 elements in the genomes of
natural isolates comprising the ECOR reference collection of Escherichia
coli. We find evidence for nonrandomness at three levels. Many pairs of IS1
elements are in much closer proximity (< 10 kb) than can be accounted
for by chance. IS1 elements in close proximity were identified by
long-range PCR amplification of the genomic sequence between them. Each
amplified region was sequenced and its map location determined by database
screening of DNA hybridization. Among the ECOR strains with at least two
IS1 elements, 54% had one or more pairs of elements separated by < 10
kb. We propose that this type of clustering is a result of "local hopping,"
in which we assume that a significant proportion of tranposition events
leads to the insertion of a daughter IS element in the vicinity of the
parental element. A second level of nonrandomness is found in strains with
a modest number of IS1 elements that are mapped through the use of inverse
PCR to amplify flanking genomic sequences: in these strains, the insertion
sites tend to be clustered over a smaller region of chromosome than would
be expected by chance. A third level of nonrandomness is observed in the
composite distribution of IS elements across strains: among 20 mapped IS1
elements, none were found in the region of 48-77 minutes, a significant
gap. One region of the E. coli chromosome, at 98 min, had a cluster of IS1
elements in seven ECOR strains of diverse phylogenetic origin. We deduce
from sequence analysis that this pattern of distribution is a result of
initial insertion in the most recent common ancestor of these strains and
therefore not a hot spot of insertion. Analysis using long- range PCR with
primers for IS2 and IS3 also yielded pairs of elements in close proximity,
suggesting that these elements may also occasionally transpose by local
hopping.

Fitness effects of amino acid replacements in the beta-galactosidase of Escherichia coli 总被引:7,自引:1,他引:6
Two genetic procedures were used to obtain amino acid replacements in the
lacZ-encoded beta-galactosidase in Escherichia coli. Amino acid
replacements could be obtained without regard to their effects on lactase
activity by selecting spontaneous mutations that relieved the strong
polarity of six nonsense mutations. When streaked on MacConkey- lactose
indicator plates, approximately 75% of these mutants gave strong red
lactose-fermenting colonies, and 25% gave white nonfermenting colonies.
Mutants from 11 other nonsense codons were isolated directly using
MacConkey-lactose indicator plates, on which positive color indication
requires only 0.5% of the wildtype lactase activity. Among the total of 17
codons, 25 variant beta-galactosidases were identified using
electrophoresis and thermal denaturation studies. The fitness effects of
these variant beta-galactosidases were determined using competition
experiments conducted with lactose as the sole nutrient limiting the growth
rate in chemostat cultures. Three of the replacements were deleterious, one
was selectively advantageous, and the selective effects of the remaining 21
were undetectable under conditions in which the smallest detectable
selection coefficient was approximately 0.4%/generation.
