The chemical composition and the antimicrobial activity of the essential oil isolated from the aerial parts of Hypericum maculatum Crantz were determined. In total, 109 compounds were identified, with germacrene D (21.5%), nonane (6.5%), (E)‐β‐farnesene (5.3%), δ‐cadinene (4.5%), and ledol (4.4%) as the main constituents. The chemical compositions of this oil and of four previously studied H. maculatum oils were compared using multivariate statistical analyses, viz., agglomerative hierarchical cluster and principal component analyses. Based on the results, the interrelationship among the hitherto studied H. maculatum oil samples, including the oil characterized here, was discussed. The study of the antimicrobial potential of the oil against five bacterial and two fungal strains showed that the oil had mainly moderate antimicrobial effects. 相似文献

A simple and efficient protocol for direct somatic embryogenesis and plant regeneration of kohlrabi (Brassica oleracea var. gongylodes) was developed. Somatic embryos were induced from immature zygotic embryos at different developmental stages cultured on Murashige and Skoog medium supplemented with 0, 0.5, 1.0, or 1.5 mg/l 2,4-dichlorophenoxyacetic acid. Zygotic embryos at the early cotyledonary stage, which were cultured for 4 wk on plant growth regulator-free (PGR-free) medium, displayed the highest percentage of somatic embryogenesis (80.7%). Embryogenic tissue could be subcultured on the same medium for over 1 yr. Embryogenic lines derived from early cotyledonary stage zygotic embryos displayed the highest intensity of secondary embryogenesis (highest mean number of new somatic embryos per responsive somatic embryo explant). Histological analyses confirmed the direct origin of the secondary somatic embryos. Prolonged culturing of embryogenic tissue on PGR-free medium led to somatic embryo development into plantlets that were successfully acclimated in the greenhouse with a survival rate of 72.5%. Flow cytometry analysis showed no ploidy variation in 96.7% of the acclimated plants. 相似文献

Repeat-associated small interfering RNAs (rasiRNAs) are derived from various genomic repetitive elements and ensure genomic stability by silencing endogenous transposable elements. Here we describe a novel subset of 46 rasiRNAs named LNCR rasiRNAs due to their homology with one long non-coding RNA (LNCR) of Spodoptera frugiperda. LNCR operates as the intermediate of an unclassified transposable element (TE-LNCR). TE-LNCR is a very invasive transposable element, present in high copy numbers in the S. frugiperda genome. LNCR rasiRNAs are single-stranded RNAs without a prominent nucleotide motif, which are organized in two distinct, strand-specific clusters. The expression of LNCR and LNCR rasiRNAs is developmentally regulated. Formation of heterochromatin in the genomic region where three copies of the TE-LNCR are embedded was followed by chromatin immunoprecipitation (ChIP) and we observed this chromatin undergo dynamic changes during development. In summary, increased LNCR expression in certain developmental stages is followed by the appearance of a variety of LNCR rasiRNAs which appears to correlate with subsequent accumulation of a heterochromatic histone mark and silencing of the genomic region with TE-LNCR. These results support the notion that a repeat-associated small interfering RNA pathway is linked to heterochromatin formation and/or maintenance during development to establish repression of the TE-LNCR transposable element. This study provides insights into the rasiRNA silencing pathway and its role in the formation of fluctuating heterochromatin during the development of one holocentric organism. 相似文献

This paper brings out the results of the study on the levels of selected trace elements (Cu, Fe, Mn, Zn and Cr) in aerial
parts of Thymus pannonicus All. (Lamiaceae) and rhizosphere soil from twelve locations in Serbia. Prior to assays by flame and flameless atomic absorption
spectrometry, samples were subjected to microwave-assisted acid digestion. Real and potential acidity of soil samples were
also measured. Obtained results for soil samples, although slightly higher for some elements (Cu: 12.38–45.18 mg/kg; Fe: 22102–46193
mg/kg; Mn: 776.95–4901.27 mg/kg; Zn: 62.27–214.02 mg/kg; Cr: 48.86–69.13 mg/kg), were found to fit into biogeochemical background.
Element contents in plant samples differed depending on collecting site (Cu: 5.26–14.07 mg/kg; Fe: 25.92–1454.07 mg/kg; Mn:
89.29–278.25 mg/kg; Zn: 1.81–10.64 mg/kg; Cr: 1.11–3.51 mg/kg), which can be partly explainable by different nutrient availability
influenced by soil acidity. Zinc levels in T. pannonicus were below expected and seem to be strongly influenced by plant physiological properties. 相似文献

The discovery of an homolog of the human centromeric protein B, CENP-B, in an EST database of the holocentric insect species Spodoptera frugiperda prompted us to further characterize that gene because i) CENP-B has not been described in invertebrates yet ii) it should be a milestone in the molecular characterization of the holocentric centromere of Lepidoptera.Like its human counterpart, the Sf CENP-B protein is related to the transposase of the pogo transposable element (TE) of D. melanogaster. In this paper, we show evidences that the lepidopteran cenpB gene has evolved from domestication of a transposase. Furthermore, the Sf CENP-B nuclear location and its ability to bind to a retrotransposon derived sequence in vivo argue in favor of a functional homology to CENP-B proteins. 相似文献

The frequency and incidence of Pyrenochaeta terrestris and symptom type on the roots of each internode of four maize hybrids of different maturity groups were studied 70 days after sowing. The fungus developed in the roots of all developed internodes (from the primary to the sixth or seventh internodes of all tested hybrids). The average frequency and incidence of P. terrestris in the roots of late and medium early maturity hybrids ranged from 29.5 to 55.2% and from 11.8 to 22.7%, respectively. The highest frequency of the fungus was at the 2nd root internode (93.3%), and its greatest incidence was detected in the mesocotyl of the medium early hybrid H‐1 (56.9%). Necrosis predominated in the roots of the medium early (i.e. medium late maturity hybrids, 44.5% and 44.3%, respectively), whereas reddish pink symptoms were recorded in the roots of the late hybrids (51% and 42.5%). Because the fungus always produces a distinctive red pigment on carnation leaf agar (CLA) in a light regime, these conditions can be recommended for the reliable identification of P. terrestris, even if pycnidia are not formed. These are the first reports of the successive distribution of the fungus in each maize root internode of different hybrids, as well as the use of CLA medium in the identification of the P. terrestris. 相似文献

The graft-versus-myeloma (GVM) effect represents a powerful form of immune attack exerted by alloreactive T cells against multiple myeloma cells, which leads to clinical responses in multiple myeloma transplant recipients. Whether myeloma cells are themselves able to induce alloreactive T cells capable of the GVM effect is not defined. Using adoptive transfer of T naive cells into myeloma-bearing mice (established by transplantation of human RPMI8226-TGL myeloma cells into CD122(+) cell-depleted NOD/SCID hosts), we found that myeloma cells induced alloreactive T cells that suppressed myeloma growth and prolonged survival of T cell recipients. Myeloma-induced alloreactive T cells arising in the myeloma-infiltrated bones exerted cytotoxic activity against resident myeloma cells, but limited activity against control myeloma cells obtained from myeloma-bearing mice that did not receive T naive cells. These myeloma-induced alloreactive T cells were derived through multiple CD8(+) T cell divisions and enriched in double-positive (DP) T cells coexpressing the CD8αα and CD4 coreceptors. MHC class I expression on myeloma cells and contact with T cells were required for CD8(+) T cell divisions and DP-T cell development. DP-T cells present in myeloma-infiltrated bones contained a higher proportion of cells expressing cytotoxic mediators IFN-γ and/or perforin compared with single-positive CD8(+) T cells, acquired the capacity to degranulate as measured by CD107 expression, and contributed to an elevated perforin level seen in the myeloma-infiltrated bones. These observations suggest that myeloma-induced alloreactive T cells arising in myeloma-infiltrated bones are enriched with DP-T cells equipped with cytotoxic effector functions that are likely to be involved in the GVM effect. 相似文献

Phytodecta fornicata Brüggemann is a serious pest of alfalfa (Medicago
sativa L.) that causes significant crop loss in the Balkan peninsula of Europe. We introduced a wound-inducible oryzacystatin II
(OCII) gene to alfalfa to evaluate its effect on survival of P. fornicata larvae. Feeding bioassays with second, third and fourth instars were carried out using transgenic plants that were shown
to express OCII at 24 and 48 h after wounding. Second and third instars were the most sensitive to the ingestion of OCII,
whereas no effects were observed with fourth instars. About 80% of the second and third instars died after 2 days of feeding
on the transgenic plants as compared to 0–40% on the controls. This is the first report that demonstrates significant increase
in mortality of P. fornicata on transgenic plants that express a cysteine proteinase inhibitor gene, and this knowledge should lead to the development
of effective management strategies for this devastating pest of alfalfa. 相似文献
