Molecular ecology methods based on 18S rRNA amplification and sequencing have revealed an astounding diversity of microbial eukaryotes in every environment sampled so far. This is certainly true of new species and genera, as essentially every new survey discovers a wealth of novel diversity at this level. This is almost certain for taxa that are higher in taxonomic hierarchy, as many molecular surveys reported novel clades within established protistan phyla, with some of these clades repeatedly confirmed by subsequent studies. It may also be that the molecular approaches discovered several lineages of the highest taxonomic order, but this claim has not been vigorously verified as yet. Overall, the field of protistan diversity remains in its infancy. The true scale of this diversity is unknown, and so are the distribution of this diversity, its patterns, spatial and temporal dynamics, and ecological role. The sampled diversity appears to be just the tip of the iceberg, and this offers outstanding opportunities for microbial discovery for the purposes of both basic and applied research. A lateral flow immunoassay for the rapid detection of Clavibacter michiganensis subsp. sepedonicus bacteria causing potato ring rot was developed. Multimembrane composites (test strips) containing polyclonal antibodies against the bacteria and gold nanoparticle-antibody conjugates were used for the analysis. The test strips are suitable for the analysis of potato tuber and leaf extracts within 10 min; the detection limit of bacteria is 4 × 105 cells/mL. No cross-reactivity with strains of Clavibacter michiganensis subsp. michiganensis, Pectobacterium carotovorum subsp. carotovorum and saprophytes of healthy potato plants was detected. The results of analysis of 26 potato samples by the developed tests were compared with those obtained by the PCR method and using the commercial enzyme immunoassay kits. The results of lateral flow immunoassay were confirmed in 96.2% of cases, which supports the high correlation with other analytical approaches. The developed immunoassay may be considered as a promising means of phytosanitary control.  相似文献

The aim of this study was to determine the reference interval of serum cobalt-binding capacity (CoBC), and to estimate the effect of factors unrelated to oxidative modification of serum albumin on this diagnostic marker. Healthy volunteers (n = 194), patients with autoimmune diseases (n = 44) and patients with type 2 diabetes mellitus (n = 50) were enrolled in the study. The regional reference interval was found to be 0,462–0,744 mmol Co2+/L of serum. The study of serum CoBC in patients with autoimmune pathology and type 2 diabetes mellitus has shown that the CoBC level strongly depends on the serum protein profile. Therefore, this test may be used to diagnose the ischemia, although other pathologies associated with changes in the ratio of blood protein fractions can also influence the serum CoBC level.  相似文献

Moderate heating of collagenous tissues such as cartilage and cornea by infrared laser irradiation can produce biologically nondestructive structural rearrangements and relaxation of internal stresses resulting in the tissue reshaping. The reshaping results and eventual changes in optical and biological properties of the tissue strongly depend on the laser‐irradiation regime. Here, a speckle‐contrast technique based on monochromatic illumination of the tissue in combination with strain mapping by means of optical coherence elastography (OCE) is applied to reveal the interplay between the temperature and thermal stress fields producing tissue modifications. The speckle‐based technique ensured en face visualization of cross correlation and contrast of speckle images, with evolving proportions between contributions of temperature increase and thermal‐stresses determined by temperature gradients. The speckle‐technique findings are corroborated by quantitative OCE‐based depth‐resolved imaging of irradiation‐induced strain‐evolution. The revealed relationships can be used for real‐time control of the reshaping procedures (e.g., for laser shaping of cartilaginous implants in otolaryngology and maxillofacial surgery) and optimization of the laser‐irradiation regimes to ensure the desired reshaping using lower and biologically safer temperatures. The figure of waterfall OCE‐image demonstrates how the strain‐rate maximum arising in the heating‐beam center gradually splits and drifts towards the zones of maximal thermal stresses located at the temperature‐profile slopes.  相似文献

There is growing interest in the development of novel single-chain bispecific antibodies for retargeting of immune effector T cells to tumor cells. Until today, functional fusion constructs consisting of a single-chain bispecific antibody and a fluorescent protein were not reported. Such molecules could be useful for an in vivo visualization of this retargeting process. Recently, we established two novel single-chain bispecific antibodies. One is capable of retargeting T cells to CD33, and the other is capable of retargeting T cells to the prostate stem cell antigen (PSCA). CD33 is an attractive immunotarget on the surface of tumor cells from patients with acute myeloid leukemia (AML). The PSCA is a potential target on prostate cancer cells. Flanking the reading frame encoding the green fluorescent protein (GFP) with a recently described novel helical linker element allowed us to establish novel single-chain bispecific fusion antibodies. These fluorescent fusion antibodies were useful to efficiently retarget T cells to the respective tumor cells and visualize the formation of immune synapses between effector and target cells.  相似文献

Successful socialization requires the ability of understanding of others’ mental states. This ability called as mentalization (Theory of Mind) may become deficient and contribute to everyday life difficulties in multiple sclerosis. We aimed to explore the impact of brain pathology on mentalization performance in multiple sclerosis. Mentalization performance of 49 patients with multiple sclerosis was compared to 24 age- and gender matched healthy controls. T1- and T2-weighted three-dimensional brain MRI images were acquired at 3Tesla from patients with multiple sclerosis and 18 gender- and age matched healthy controls. We assessed overall brain cortical thickness in patients with multiple sclerosis and the scanned healthy controls, and measured the total and regional T1 and T2 white matter lesion volumes in patients with multiple sclerosis. Performances in tests of recognition of mental states and emotions from facial expressions and eye gazes correlated with both total T1-lesion load and regional T1-lesion load of association fiber tracts interconnecting cortical regions related to visual and emotion processing (genu and splenium of corpus callosum, right inferior longitudinal fasciculus, right inferior fronto-occipital fasciculus, uncinate fasciculus). Both of these tests showed correlations with specific cortical areas involved in emotion recognition from facial expressions (right and left fusiform face area, frontal eye filed), processing of emotions (right entorhinal cortex) and socially relevant information (left temporal pole). Thus, both disconnection mechanism due to white matter lesions and cortical thinning of specific brain areas may result in cognitive deficit in multiple sclerosis affecting emotion and mental state processing from facial expressions and contributing to everyday and social life difficulties of these patients.  相似文献

The effect of supercompensation and the conditions of its origination have been analyzed using a special analytical solution of the equation of the overdamped Duffing oscillator. The threshold values of the functional shift after the exposure of a living organism to intensive external load have been found. If these values are exceeded, the restoration process goes through the stages of enhanced and lowered working capacity. Expressions for the times of transition to these phases after the onset of recovery and for the time of occurrence of the maxima of these phases have been obtained and analyzed.  相似文献
