The effect of intravenous infusion of neurotensin (NT) and NT-fragments on pentagastrin stimulated gastric acid secretion was investigated in healthy subjects. Neurotensin was infused in three doses (72, 144 and 288 pmol/kg per h). An N-terminal fragment (NT 1-8), a C-terminal fragment (NT 8-13) and an NT-analogue, substituted at the C-terminal tyrosine residue (Phe11-NT) were infused in two doses (72 and 144 pmol/kg per h). Concentrations of the infused peptides were measured in peripheral venous blood by radioimmunoassay. Plasma levels of NT 1-13, NT 1-8 and Phe11-NT increased in a dose-dependent manner; NT 1-13 to 50 (34-69), 78 (54-113) and 143 (112-242) pmol/l (medians and range) at 72, 144 and 288 pmol/kg per h, NT 1-8 to 405 (340-465) and 1215 (915-1300) pmol/l, and Phe11-NT to 200 (110-245) and 390 (250-410) pmol/l at 72 and 144 pmol/kg per h, respectively. Increases in plasma levels of NT 8-13 could not be detected during the infusion, suggesting that the fragment is rapidly metabolized in man. Neurotensin 1-13 inhibited gastric acid secretion in a dose-dependent manner and the decrease in gastric acid secretion was linearly related to plasma levels of NT 1-13. Neurotensin 1-8 and NT 8-13 inhibited gastric acid secretion only at 144 pmol/kg per h, while the analogue Phe11-NT had no effect. The results showed that the inhibition of gastric acid secretion produced by NT was dose-dependent and linearly related to circulating levels of NT, and that under physiological conditions this effect presumably is elicited by the C-terminal part of the peptide.  相似文献

Urinary epidermal growth factor (EGF) has been demonstrated recently to originate from the kidneys. The present study was undertaken to investigate the adrenergic and cholinergic influence on secretion of renal EGF. beta-Adrenergic agonists increased the level of urinary EGF, while propranolol, a beta-adrenergic blocking agent, decreased basal and beta-adrenergic stimulated total output of urinary EGF. Acetylcholine and the anticholinergic agent atropine had no effect on the output of EGF in urine. Also chemical sympathectomy induced by 6-hydroxydopamine reduced the urinary output of EGF. None of the experimental groups had a median serum concentration above the detection limit of the assay. The present study shows that secretion of renal EGF is under the influence of the sympathetic nervous system and release of EGF is stimulated by activation of beta-adrenergic receptors in the kidneys.  相似文献

The radiosensitizer 2-amino-5-nitrothiazole (ANT) can react with platinum to form many products because of the availability of two potential nitrogen donors as ligands for metals. Two of these complexes, both with two ANT molecules in the trans configuration, differ because of their linkages. In the first, Pt is bound to ANT via the amine group (A) and in the second via the thiazole ring nitrogen (R). Isomer R is a better radiosensitizer than A in hypoxic Chinese hamster ovary cells, giving enhancement ratios of 1.6 versus 1.15 with 100 mumol dm-3 complex. The ring-bound isomer also exhibits higher toxicity than the amine bound in air and under conditions of hypoxia. Sensitization by isomer R is much higher under conditions of hypoxia than in air. In an assay to assess DNA binding, isomer R inhibited restriction enzyme cleavage of DNA but isomer A did not (up to 6 h at 300 mumol dm-3). It appears from this study on two very similar complexes that the complex which exhibits stronger binding may be targetting the radiosensitizer to the DNA, resulting in greatly improved sensitization.  相似文献

Summary Epidermal growth factor (EGF) is a peptide which stimulates cell mitotic activity and differentiation, has a cytoprotective effect on the gastroduodenal mucosa, and inhibits gastric acid secretion.The immunohistochemical localization of EGF in the Brunner's glands and the submandibular glands is well documented. The localization of EGF in other tissues is still unclarified.In the present study, the immunohistochemical localization of EGF in tissues from rat, man and a 20 week human fetus were investigated. In man and rat, immunoreaction was found in the submandibular glands, the serous glands of the nasal cavity, Brunner's glands of the duodenum, the Paneth cells of the small intestine, and the tubular cells of the kidney. In the fetus EGF was found in the kidney and in the intestinal Paneth cells.Antisera raised against rat submandibular EGF did not recognize EGF in human tissues, whereas antisera against human urinary EGF worked in rat as well as man. EGF was found only in cells with an exocrine function.  相似文献

Aims: To investigate the transmission of Salmonella spp. between production animals (pigs and cattle) and wildlife on production animal farms in Denmark. Methods and Results: In the winter and summer of 2001 and 2002, 3622 samples were collected from Salmonella-infected and noninfected herds of pigs and cattle and surrounding wildlife. Salmonella was detected in wildlife on farms carrying Salmonella-positive production animals and only during the periods when Salmonella was detected in the production animals. The presence of Salmonella Typhimurium in wild birds significantly correlated to their migration pattern and food preference. Conclusions: Salmonella was transmitted from infected herds of production animals (cattle and pigs) to wildlife that lived amongst or in close proximity to them. Significance and Impact of the Study: Salmonella in animal food products is associated with the occurrence of Salmonella in primary animal production. Strategies to control the introduction and spread of infection should include wildlife management, as the nearby wildlife may act as reservoirs for Salmonella spp. and/or may be passive carriers of the bacteria.  相似文献

The aim of this study is to evaluate the possible consequences of climate change on a representative sample of forest herbs in Europe. A fuzzy climatic envelope was used to predict the location of suitable climatic conditions under two climatic change scenarios. Expected consequences in terms of lost and gained range size and shift in distribution for 26 forest herbs were estimated. These results were combined in an Index of Predicted Range Change for each species. Finally, the effects of habitat fragmentation for potential dispersal routes were evaluated and options for management on a European scale are discussed. Generally, a good agreement of the estimated suitability under the present climate and the observed current distribution was observed. However, species vary a lot in the degree to which they occupy the presently climatically suitable areas in Europe. Many species are absent from large areas with suitable climate and thus could be said to have poor range‐filling capacity. A general change in location (range centroid) of the total suitable area was observed: The total suitable area will on average move strongly northwards and moderately eastwards under the relatively mild B1 scenario and more strongly so under the A2 scenario. The required average minimum migration rate per year to track the potential range shift is 2.1 km under the B1 scenario and 3.9 km under the A2 scenario. Moderate losses in the total suitable area in Europe are predicted for most species under both scenarios. However, the predicted changes are very variable, with one species (Actaea erythrocarpa) experiencing total range elimination in Europe (A2 scenario) while the total suitable area is predicted to show large increases for other species. The species that are predicted to experience the greatest proportional losses in their climatically suitable area within their presently realised range tend to have northern or eastern range centroids. The Index of Predicted Range Change roughly divides the species studied in four groups: One species face a high risk of extinction; eight species are expected to experience moderate to severe threat of extinction; 11 species are not considered at risk and, finally, six species may actually benefit from global warming. An analysis of potential migration routes shows the importance of maintaining and, if possible, improving the network of forest throughout Europe to make migration possible. It is also suggested to closely monitor the status of boreal and subalpine species that are most threatened by global warming. Finally it is recommended that special concern should be given to increased protection and restoration of forest habitats in southern montane areas for their crucial long‐term importance for the maintenance of European plant diversity.  相似文献
