The concentrations of 16 alpha-hydroxydehydroepi-androsterone sulfate (16 alpha-OHDHAS) and androst-5-ene-3 beta, 16 alpha-, 17 beta-triol-3-sulfate (A-TriolS) were measured in the plasma and breast cyst fluid (BCF) of women with gross cystic disease of the breast. In the 19 BCF samples analyzed, the 16 alpha-OHDHAS and A-TriolS concentrations ranged from 15 to 1130 ng/mL, and 12 to 871 ng/mL, respectively. However the concentrations of these steroids in the sera of these women were lower (15-179 ng/mL, 8-80 ng/mL, respectively). Estriol-3-sulfate (E3-3S) concentrations in the BCF samples ranged from barely detectable (0.2 ng/mL) to 3 micrograms/mL. In BCF or serum a positive linear correlation was observed in the concentration of 16 alpha-OHDHAS and A-TriolS (p less than 0.001 and 0.05, respectively). However, in the same patients no statistical significance was observed in the BCF vs serum concentrations of these two steroids. When the specimens from this and previous studies were combined, positive correlation was found between potassium ion concentration and E3-3S or 16 alpha-OHDHAS. The origin of the high concentration of E3-3S is still obscure. Although no linear correlation between 16 alpha-OHDHAS and E3-3S was observed, the possibility of a precursor-product relationship between the two is not elimnated.  相似文献

Eight rhesus monkeys were used to study responses of radial artery blood flow velocity (RABFV) and heart rate (HR) to low (0 to -20 mmHg) and high (0 to -60 mmHg) ramp exposures during supine lower body negative pressure (LBNP). These levels were chosen to separate peripheral vascular responses associated with stimulation of low- and high-pressure baroreceptors. Four monkeys had efferent and afferent cardiac denervation by use of the Randall procedure with pharmacological (phenylephrine and atropine) verification. Animals were studied 3 wk after surgery to avoid reinnervation. Findings were compared with those of four identically treated intact animals. Denervated animals showed no change in RABFV or HR during low-level LBNP; however, HR increased significantly (P less than 0.05) when LBNP reached -50 mmHg and blood flow velocity also fell (P less than 0.05) starting at -30 mmHg pressure. In contrast, intact animals showed steady decreases in RABFV during both high- and low-pressure protocols, with HR showing a 6-beat/min increase (P less than 0.05) starting at -20 mmHg pressure. As with denervated animals, intact animals showed a more pronounced increase in HR after reaching a level of -60 mmHg suction. Cardiac output (electromagnetic flowmeter, ascending aorta) fell significantly in both groups starting at -30 mmHg pressure. Left ventricular pressure (Konigsberg pressure cell) in three intact animals showed a progressive fall in systolic pressure starting at -10 mmHg suction, which became significant at -55 mmHg pressure. These results demonstrate that cardiac denervation by use of the Randall technique significantly affects RABFV and HR responses to LBNP in rhesus monkeys. The lack of RABFV change during LBNP in denervated animals suggests that these changes coupled with HR response can be used as an effective method to verify the completeness of denervation of low-pressure baroreceptors in animals that have undergone intrapericardial denervation.  相似文献

Poaching is the most immediate threat to African elephants (Loxodonta africana). Several continental-wide surges in poaching have occurred since the latter half of the twentieth century, and the latest surge occurred from 2007 to 2012. The behavioral responses of elephants to poaching risk has not been studied widely because of a lack of high-resolution movement data collected simultaneously with verified causes of mortality. We managed to collate 2 such datasets from 2004 to 2013. We studied the spatial-temporal changes in movement behavior of 11 elephants in their core areas. Past studies have focused on elephant movement along corridors. We tested for the effect of poaching risk on their path straightness (i.e., tortuosity) while controlling for other environmental and human activities in the landscape using a set of generalized linear mixed models. To test for temporal variation of tortuosity, we used a time-series linear model. Elephants turned less frequently while they were in poaching locations and at times with a high level of poaching activity, even though their speed did not change. The variation of tortuosity is a good indicator of differences in poaching risk as perceived by the elephants, which could complement patrol-based anti-poaching efforts by wildlife managers, especially in remote, inaccessible landscapes. © 2019 The Authors. This review summarizes the basic reproductive physiology of dromedary and Bactrian camels. Camels are seasonal breeders with a relatively short breeding season during the cooler months. The onset of the breeding season can be influenced by local environmental factors such as temperature and pasture availability although decreased libido of the male as the environmental temperature increases is also a factor. Oestrous behaviour is highly variable in duration and intensity and is therefore unreliable for the detection of oestrus and difficult to relate to follicular activity in the ovaries. Camels are induced ovulators and thus normally only ovulate in response to mating. In the absence of mating, ovarian follicles tend to regress after a period of growth and maturity, whereas if male and females are kept together the female gets mated when the dominant follicle measures between 1.3 and 1.7 cm in diameter and the corpus luteum that develops has a lifespan of only 10-12 days. Peripheral concentrations of oestradiol increase with increasing follicle diameter until the follicle reaches 1.7 cm in diameter at which time they start to decrease even if the follicle continues to grow. The concentrations of progesterone remains low in non-mated animals but in mated camels it increases 3-4 days after ovulation (day of ovulation=Day 0) to reach maximum concentrations on Days 8-9 before decreasing rapidly on Days 10-11 in the non-pregnant animal. Ovulation can also be reliably induced using either Gonadotrophin Releasing Hormone (GnRH) or human Chorionic Gonadotrophin (hCG) but only when the follicle measures between 1.0 and 1.9 cm in diameter. Ovulation does not typically occur from follicles that grow beyond 2.0 cm in diameter but these follicles typically develop echogenic strands of fibrin as the follicle degenerates. The gestation period of camels is 13 months but the time of resumption of follicular activity following parturition is highly variable and influenced by nutritional status and lactation. Females that lose their offspring or have offspring which are weaned have a mature follicle develop within 10-12 days, whilst in well-fed lactating females mature follicles do not develop until 30-60 days postpartum.  相似文献

1. Elucidation of the mechanism determining the spatial scale of patch selection by herbivores has been complicated by the way in which resource availability at a specific scale is measured and by vigilance behaviour of the herbivores themselves. To reduce these complications, we studied patch selection by an animal with negligible predation risk, the African elephant. 2. We introduce the concept of nutrient load as the product of patch size, number of patches and local patch nutrient concentration. Nutrient load provides a novel spatially explicit expression of the total available nutrients a herbivore can select from. 3. We hypothesized that elephant would select nutrient-rich patches, based on the nutrient load per 2500 m(2) down to the individual plant scale, and that this selection will depend on the nitrogen and phosphorous contents of plants. 4. We predicted that elephant would cause more adverse impact to trees of lower value to them in order to reach plant parts with higher nutrient concentrations such as bark and root. However, elephant should maintain nutrient-rich trees by inducing coppicing of trees through re-utilization of leaves. 5. Elephant patch selection was measured in a homogenous tree species stand by manipulating the spatial distribution of soil nutrients in a large field experiment using NPK fertilizer. 6. Elephant were able to select nutrient-rich patches and utilized Colophospermum mopane trees inside these patches more than outside, at scales ranging from 2500 down to 100 m(2) . 7. Although both nitrogen and phosphorus contents of leaves from C. mopane trees were higher in fertilized and selected patches, patch choice correlated most strongly with nitrogen content. As predicted, stripping of leaves occurred more in nutrient-rich patches, while adverse impact such as uprooting of trees occurred more in nutrient-poor areas. 8. Our results emphasize the necessity of including scale-dependent selectivity in foraging studies and how elephant foraging behaviour can be used as indicators of change in the availability of nutrients.  相似文献

Quantifying the clearance of extractables and leachables (E/L) throughout ultrafiltration/diafiltration (UFDF) operations allows for greater flexibility in the implementation of single‐use technologies in steps upstream of the UFDF process. A proof‐of‐concept study was completed in which the clearance of 7 E/L from single‐use technologies (trimethylsilanol, hexanoic acid, butyrolactone, t‐butyl alcohol, caprolactam, acetonitrile, and benzyl alcohol) in four representative proteins were measured and monitored during the UFDF process using quantitative NMR. This study demonstrated that the defined E/L spiked into a variety of protein solutions can be cleared to <1 ppm by 9 diavolumes from a maximum initial load concentration of 1,000 ppm. However, in some cases a rebound effect was observed in the recovered pool to >1 ppm, which is explained in detail. The overall clearance trend observed for both buffer control and protein‐containing solutions resembled the ideal clearance trend where no apparent interactions were observed between E/L with the protein, UFDF system, or with other defined E/L which may be present in the system. Additionally, the UFDF system is capable of clearing these potential E/L from single‐use technologies below 1 ppm irrespective of initial concentrations in the load (1,000 or 100 ppm), independently from the type of protein. In general, mass recoveries were within ±15% of each spiked compound in protein solutions and their respective buffer controls, suggesting spiked E/L do not interact strongly with protein. By demonstrating the product independent clearance trends of the spiked E/L across UFDF, these results will contribute to the simplification of the E/L toxicology assessment and allow modular manufacturing approach for single‐use technologies in biopharmaceutical manufacturing. © 2016 American Institute of Chemical Engineers Biotechnol.
