The genus and the species have been described in detail, measurements of body parts along with illustrations have also been provided for future references. Wingless信号传导是果蝇胚胎和幼虫发育过程中的一个关键性的信号传导通路。已鉴定出来许多参与Wg Wnt信号传导的Wg或其脊椎动物同源基因Wnt下游传导通路的基因。Wg下游的Wg信号传导是由核TCF LEF 1通过Armadillo (Arm) β catenin介导的。pygopus (pygo)是一个最近发现的Wg Wnt信号传导通路新成员。通过细胞定位实验发现pygo专一性的表达在细胞核中。运用反义mRNA作探针的原位杂交技术 ,观察到了pygo基因在果蝇胚胎中的表达特性。虽然pygo普遍表达于果蝇胚胎发生全过程 ,但pygo在前囊胚层 (pre blastoderm)中的表达水平相对较高 ,这说明胚胎发生过程中来自母方的贡献较高。在幼虫组织 (包括翅成虫盘 ,眼成虫盘和腿成虫盘 )的发育过程中 ,pygo的表达水平总的来说比较低。然而 ,比较翅成虫盘 ,眼成虫盘和腿成虫盘的pygo表达水平 ,则在翅成虫盘和腿成虫盘pygo的表达水平相对较高。  相似文献

The gene encoding catalase from the psychrophilic marine bacterium Vibrio salmonicida LFI1238 was identified, cloned and expressed in the catalase-deficient Escherichia coli UM2. Recombinant catalase from V. salmonicida (VSC) was purified to apparent homogeneity as a tetramer with a molecular mass of 235 kDa. VSC contained 67% heme b and 25% protoporphyrin IX. VSC was able to bind NADPH, react with cyanide and form compounds I and II as other monofunctional small subunit heme catalases. Amino acid sequence alignment of VSC and catalase from the mesophilic Proteus mirabilis (PMC) revealed 71% identity. As for cold adapted enzymes in general, VSC possessed a lower temperature optimum and higher catalytic efficiency (k cat/K m) compared to PMC. VSC have higher affinity for hydrogen peroxide (apparent K m) at all temperatures. For VSC the turnover rate (k cat) is slightly lower while the catalytic efficiency is slightly higher compared to PMC over the temperature range measured, except at 4°C. Moreover, the catalytic efficiency of VSC and PMC is almost temperature independent, except at 4°C where PMC has a twofold lower efficiency compared to VSC. This may indicate that VSC has evolved to maintain a high efficiency at low temperatures.  相似文献

Ketonic bile acids have been found to be quantitatively important in urine of healthy infants during the neonatal period. In order to determine their structures, the bile acids in urine from 11 healthy infants were analyzed by gas-liquid chromatography-mass spectrometry (GLC-MS) and three samples with particularly high levels of ketonic bile acids were selected for detailed studies by ion exchange chromatography, fast atom bombardment mass spectrometry, microchemical reactions, and GLC-MS. The major ketonic bile acid was identified as 7 alpha, 12 alpha-dihydroxy-3-oxo-5 beta-chol-1-enoic acid, not previously described as a naturally occurring bile acid. The positional isomer 7 alpha, 12 alpha-dihydroxy-3-oxo-4-cholenoic acid, recently described as a major urinary bile acid in infants with severe liver diseases, was also excreted by most infants. Three acids related to cholic acid were identified: 7 alpha, 12 alpha-dihydroxy-3-oxo-, 3 alpha, 12 alpha-dihydroxy-7-oxo-, and 3 alpha, 7 alpha-dihydroxy-12-oxo-5 beta-cholanoic acids. Five bile acids having one oxo and three hydroxy groups were also present. Based on mass spectra and biological considerations two of these were tentatively given the structures 1 beta, 7 alpha, 12 alpha-trihydroxy-3-oxo- and 1 beta, 3 alpha, 12 alpha-trihydroxy-7-oxo-5 beta-cholanoic acids. Some of the others had a hydroxy group at C-4 or C-2. The levels of ketonic bile acids were higher on the third than on the first day of life, and lower after 1 month. The formation and excretion especially of 3-oxo bile acids is proposed to result from changes of the redox state in the liver in connection with birth.  相似文献
