The effect of specific restriction endonuclease on the simian adenovirus SV20 DNA was studied. It was shown that endonucleases SalI, XbaI, EcoRI, BamHI, HindIII cleaved the viral DNA into 3, 4, 5, 5, 8 specific fragments respectively. The sequence of fragments (physical map) was determined and found to be B-C-A for enzyme SalI, C-D-B-A--for enzyme Xbal, E-A-C-D-B--for enzyme EcoRI, B-E-C-A-D--for enzyme BamHI and B-E-A-C-(GH)-D-F--for enzyme HindIII. The G-C content of specific fragments was studied. The "right"-"left" orientation of the physical map of the simian adenovirus 20 DNA based on the G-C content was made in respect with the nomenclature of human adenoviruses.  相似文献

Calnexin and ERp57 act cooperatively to ensure a proper folding of proteins in the endoplasmic reticulum (ER). Calnexin contains two domains: a lectin domain and an extended arm termed the P-domain. ERp57 is a protein disulfide isomerase composed of four thioredoxin-like repeats and a short basic C-terminal tail. Here we show direct interactions between the tip of the calnexin P-domain and the ERp57 basic C-terminus by using NMR and a novel membrane yeast two-hybrid system (MYTHS) for mapping protein interactions of ER proteins. Our results prove that a small peptide derived from the P-domain is active in binding ERp57, and we determine the structure of the bound conformation of the P-domain peptide. The experimental strategy of using the MYTHS two-hybrid system to map interaction sites between ER proteins, together with NMR, provides a powerful new strategy for establishing the function of ER complexes.  相似文献

Genotypic variability at six allozyme loci and six morphological shell characters was examined in 450 individuals (four samples) of mollusk Nucella heysana from the Vrangel' Bight (Nakhodka Bay) and the Vostok Bay (Peter the Great Gulf, Sea of Japan). An analysis of variation in allele frequencies showed that each of the two localities (Vostok and Nakhodka bays) in the analyzed region is inhabited by a single, albeit genetically heterogeneous, population. A canonical analysis and an analysis of variance of individual heterozygosity (Ho) and morphological variation indicate an association between Ho and morphological variation depending on habitat (interaction) in settlements in the Vrangel' Bight and, to a lesser extent, in the Vostok Bay. These results indicate that, in extreme environmental conditions, allozyme phenotypes may be selected either themselves or as markers of genes linked with them.  相似文献

For the first time, the authors assessed and mapped the chromosome QTLs (Quantitative Trait Loci) for the manifestation of morpho-physiological and agronomic indices of plant water status and related quantitative traits, such as plant height, weight, and dry matter content in spring bread wheat (Triticum aestivum L.). Following the study of ten agronomic traits, 13 QTLs were mapped on linkage groups 1A, 1B, 2B, 2D, 4A, 5A, 5B, 5D, 6A, and 6D. Some of the identified QTLs concurrently determined several traits. The physiological components of water status were shown to correlate with quantitative traits in wheat plants, such as plant height, weight, and dry matter content, and the correlation coefficients were calculated for all traits under study. Water retention capacity after 3 h correlated with water retention capacity after 24 h (r xy = 0.47). The correlations were also established between water retention capacity after 3 h and plant height at booting stage (r xy = 0.29) and between water retention capacity after 3 h and plant dry weight (r xy = 0.33). Statistical calculations supported generally observed negative correlation (up to ?1) between leaf water and dry matter contents, as well as between the root indices of variance in the mapping population of wheat lines. The results obtained in the present study will promote future efforts to fine-map the genes residing within the identified QTLs, to eventually clone these genes in order to establish the physiological mechanisms for maintaining water homeostasis in higher plant cells and to accomplish the practical implementation of marker-assisted assessment of water status in wheat plants studied on the basis of morpho-physiological and economical indices.  相似文献

Simian adenovirus 20 DNA was specifically cleavered by restriction endonucleases EcoRI, BamHI, XbaI and HindIII. The transformation activity of the DNA digest was investigated. BamHI, XbaI, and HindII DNA digests were able to transform the primary rat kidney cell culture (Wistar) as well as the native SV20 DNA. The transforming activity was revealed in a specific fragment of the viral DNA, obtained after the treatment of the DNA with BamHI (fragment B), with molecular weight 5.4 x 10(6) dalton. This fragment is located in the left end of the viral genome. The lack of cell transformation by the EcoRI-hydrolysate of viral DNA may serve a proof of the extremely left position of the oncogene in the viral genome, since of EcoRI-fragment chips off a fragment with molecular weight 3 x 10(5) dalton fr om the left side of DNA molecule.  相似文献

Mice of C57BL/6J line with higher exploratory and motor activities and with lower emotionality and pain threshold recorded in standard tests, as compared with the animals of CBA/Lac line, have also been found to manifest a more expressed aggression in their intermale contacts in reaction to a syngeneous partner. It is suggested that the studied physiological and behavioural parameters may determine in considerable degree the character of aggressive and submissive mice behaviour in an agonistic interaction.  相似文献

The parameters of erythrocyte ghost protein's fluorophores by nitrate's anions were studied in patients with various hereditary myodystrophy. In all the groups under examination the share of fluorophores accessible to a quencher was close to 1. In erythrocyte membranes of healthy donors the relevant constant quenchering was about 17.3 +/- 1.9 M-1 while those of patients were decreased by 3.1 (Duchenne's myodystrophy) and by about 2.0 (other forms of primary and secondary progressive muscular dystrophies). The most probable reason for the decreasing constant of quenchering is the increase of negative charges on the erythrocyte membrane proteins.  相似文献
