Although soil contains only traces of soluble carbohydrates, plant roots take up glucose and sucrose efficiently when supplied in artificial media. Soluble carbohydrates and other small metabolites found in soil are in part products from exudation from plant roots. The molecular nature of the transporters for uptake and exudation is unknown. Here, fluorescence resonance energy transfer (FRET) glucose and sucrose sensors were used to characterize accumulation and elimination of glucose and sucrose in Arabidopsis roots tips. Using an improved image acquisition set-up, FRET responses to perfusion with carbohydrates were detectable in roots within less than 10 sec and over a wide concentration range. Accumulation was fully reversible within 10-180 sec after glucose or sucrose had been withdrawn; elimination may be caused by metabolism and/or efflux. The rate of elimination was unaffected by pre-incubation with high concentrations of glucose, suggesting that elimination is not due to accumulation in a short-term buffer such as the vacuole. Glucose and sucrose accumulation was insensitive to protonophores, was comparable in media differing in potassium levels, and was similar at pH 5.8, 6.8 and 7.8, suggesting that both influx and efflux may be mediated by proton-independent transport systems. High-resolution expression mapping in root tips showed that only a few proton-dependent transport of the STP (Sugar Transport Protein) and SUT/SUC (Sucrose Transporter/Carrier) families are expressed in the external cell layers of root tips. The root expression maps may help to pinpoint candidate genes for uptake and release of carbohydrates from roots.  相似文献

The cell surface adhesion molecule LFA-1 coordinates leukocyte trafficking and is a costimulatory molecule for T cell activation. We developed a panel of mAbs that recognize activation epitopes on the CD18 subunit, and show that stimulation of T lymphocytes appears to be accompanied by a conformational change in a subpopulation of LFA-1 that does not require ligand binding. Activation epitope up-regulation requires divalent cations, is sensitive to cellular signal transduction events, and correlates with cell adhesion. In addition, the stimulated appearance of these activation epitopes is absent in cell lines from patients with leukocyte adhesion deficiency-1/variant that has previously been shown to be defective in LFA-1 activation. Thus, these activation epitope Abs can be used to dissect signal transmission to CD18. Evidence suggests that these CD18 activation epitopes are induced early in cellular activation and are independent of actin rearrangement necessary for avid adhesion. We have also determined that function-blocking CD18 Abs inhibit the induction of activation epitopes. One activation epitope Ab binds to a site on CD18 distinct from that of the blocking Abs, indicating that the blocking Abs suppress a conformational change in LFA-1. We also find that these neoepitopes are present on rLFA-1 with high affinity for ICAM-1 and their binding is modulated in parallel with the affinity of LFA-1 for ICAM-1. Collectively, these neoepitope Abs identify a subpopulation of LFA-1 most likely with high affinity for ICAM-1 and necessary for LFA-1 function.  相似文献

Heterotrophic and autotrophic culture in agar and in polyurethane foam, the latter used as an alternative tissue support to agar, resulted in potato microplants with different in vitro morphologies. The microplants were visually characterised in terms of their relative developmental maturity, by comparing the respective leaf shapes in vitro with ontogenetic differences in leaf shape in glasshouse-grown potato plants. Cytosine methylation in the DNA of microplants of the different morphologies was determined using a method based on the AFLP technique but employing methylation-sensitive restriction enzymes (MSAP analysis) to test the hypothesis that DNA methylation could be used to characterise differences in microplant development in vitro. In three of the four treatments there was a good correlation between the visual assessment of relative morphological maturity and DNA base methylation levels. In these microplants there was increased DNA methylation in the leaves with mature leaf morphology represented by a decreased number of restriction fragments. The fourth in vitro morphology had the most juvenile leaf shape but did not have the predicted level of DNA methylation, having a relatively low number of restriction fragments. Subtraction analysis was used to discriminate the fragments that were unique to the juvenile and mature in vivo leaf morphologies. Comparison of the fragment patterns from the microplants with the latter reference profiles, confirmed the relationship with the total DNA methylation as detected by MSAP analysis, that is, the number of common fragments with the juvenile or mature in vivo leaf profiles, respectively. However, none of the fragment profiles, while sharing some common bands at random, was identical to any other; or to that of either the juvenile or mature in vivo leaf. The anomalous relationship of the microplants with most juvenile leaf shape and highest DNA methylation was confirmed. The measurement of DNA methylation in in vitro plants is discussed in the context of the development of a method to assess the quality of microplants produced by different in vitro protocols.  相似文献

Surficial soil development was studied in four wetland basins created on the floodplain of the Des Plaines River near Chicago, Illinois, USA. These studies determined changes in the spatial distribution of plant-available nutrients as a result of establishing two different wetland hydrologic regimes. Three wetland basins had mineral soils and one an organic soil. A geostatistical analysis including kriging of collected data indicated that all soil parameters showed significant changes in their spatial structure as a result of the water inputs and unidirectional flows. The degree of spatial variability as indicated by autocorrelation in the soil data (i.e., points closer to one another are more similar than points further apart due to the influence of landscape processes) declined for all parameters except Mg+2. Temporal changes in the spatial patterns of extractable phosphorus (P) and percent organic carbon (OC) tended to be inverse; P declined in areas where OC increased and vice versa. The spatial pattern of these changes was dissimilar in the mineral soils as compared to the organic soil and was related to patterns of primary productivity. Zones of P uptake and OC accumulation were also related to wetland hydrology and primary productivity. Changes in the distribution of nutrients, particularly P, may be viewed as a result of nutrient spirals within the wetlands. By comparison, the reorganization in the concentrations of K+ and Ca+2 appear to have been mediated by cation exchange processes. The formation of new concentration gradients was strongly related to both flow pathways and the different water inflow rates. The formation of concentration gradients in exchangeable cations was not reflected in the average concentrations within each basin. Mean values changed significantly in only a few instances. Reducing data in this way missed important biogeochemical changes occurring within the experimental wetland basins. The specific activity of adenosine diphosphatase (ADPase) was determined in rat tissue homogenates using a specific radioassay. 14 different tissues were sampled and activity was found in most homogenates examined. The levels ranged from very little activity in the pancreas to high levels in the duodenum and ileum. High levels of ADPase activity found in the gastrointestinal tract could be partially attributed to non-specific alkaline phosphatase. It is concluded that ADPase activity is widely distributed in rat tissues.  相似文献

ABSTRACT

Introduction: Two of the most ubiquitous fatigue countermeasures used by shift-working nurses are napping and caffeine. This mixed-methods case study investigated the ways nurses and midwives utilised napping and caffeine countermeasures to cope with shift work, and associated sleep, physical health and psychological health outcomes.

Materials and Methods: = 130 Australian shift-working nurses and midwives (mean age = 44 years, range = 21–67, 115F, 15M) completed the Standard Shiftwork Index. A sub-set of 22 nurses and midwives completed an in-depth interview.

Results: Nearly 70% of participants reported napping. Those who napped during night shifts had significantly less total sleep time before (F2,75 = 5.5, < 0.01) and between days off (F2,82 = 3.9, < 0.05). By the end of the night shift, average hours of time awake were significantly less for prophylactic and on-shift nappers compared to non-nappers (F2,85 = 97.2, p < 0.001). Since starting shift work, the percentage of high caffeine consumers (>400 mg/day) increased from 15% to 33% of the sample and an average of 4 (SD = 2) caffeinated beverages per day was reported. Increased caffeine consumption was associated with greater sleep disturbance (= 0.26, < 0.05), psychological distress (= 0.37, < 0.001), abdomen pain (= 0.27, < 0.05) and weight gain since starting shift work (= 0.25, < 0.05). Interviews confirmed these relationships and revealed that caffeine consumption on night shift was common, whereas napping on night shift was dependent on a number of factors including ability to sleep during the day.

Conclusion: This study identified reasons shift workers chose to engage in or abstain from napping and consuming caffeine, and how these strategies related to poor sleep and health outcomes. Further research is required to help develop recommendations for shift workers regarding napping and caffeine consumption as fatigue countermeasures, whilst taking into account the associated hazards of each strategy.  相似文献

Uptake by roots from contaminated soil is one of the key steps in the entry of radiocaesium into the food chain. We have measured the uptake by roots of radiocaesium and its transfer to shoots of a heathland grass, sheep fescue (Festuca ovina L.) from two contrasting agricultural soils, a sandy podzol and a clayey calcareous soil. A culture device which keeps the roots separate from the soil was used thus allowing rhizosphere soil to be obtained easily and enhancing the effect of root action. Biomass production and 137Cs in shoots and roots were recorded. Cs adsorption was studied on both the initial, nonrhizosphere soil and on rhizosphere soil in dilute soil suspension. Cs desorption was measured by resuspending subsamples of contaminated soil in solutions containing various concentrations of stable Cs. The proportion of Cs fixed, i.e. not readily desorbable, was calculated by comparison of the adsorption and desorption isotherms. Uptake by roots varied considerably between soils and did not appear to be diffusion limited. Root-to-shoot transfer did not differ for the two soils studied. Root action considerably enhanced Cs adsorption on the soils, particularly the in sandy podzol with a low Cs affinity. The value of Kd was increased by up to an order of magnitude. A large proportion of adsorbed Cs was found to be fixed, the Kd was up to seven times greater on desorption than adsorption, indicating that up to 80% of adsorbed Cs was not readily exchangeable.
