Eleocharis subarticulata is recorded as the third species of Cyperaceae with a reduced chromosome number ( n  = 3), following reports on Rhynchospora tenuis ( n  = 2) and Fimbristylis umbellaris ( n  = 3). For Eleocharis, the numbers recorded to date vary from 2 n  = 10 to 2 n  = c. 196, with x  = 5 as the possible basic number. The karyotype of E. subarticulata was studied using conventional staining (mitosis and meiosis), C-CMA3/DAPI banding, and FISH with 45S rDNA and telomere probes. The chromosomes showed no primary constrictions, as expected in the holocentric chromosomes of Cyperaceae. The meiotic behaviour was abnormal, with a single multivalent ring of six chromosomes at metaphase I, resulting from multiple translocations. At anaphase I six chromatids migrated to each pole, evidencing the inverted meiosis, and these groups were also visible at metaphase II. The C-CMA3/DAPI banding technique showed only four terminal GC-rich blocks. FISH with 45S rDNA probes revealed four terminal signals, probably associated with GC-rich blocks. The telomeric probe located terminal signals in all the chromosomes, besides a hybridization site in the middle of the large pair. The occurrence of ectopic telomeric sites has not been described previously for plants with holokinetic karyotypes and with reduced chromosome numbers. Shiga toxin-producing Escherichia coli isolates from two 2006 outbreaks were compared to other O157:H7 isolates for virulence genotype, biofilm formation, and stress responses. Spinach- and lettuce-related-outbreak strains had similar pulsed-field gel electrophoresis patterns, and all carried both stx2 and stx2c variant genes. Cooperative biofilm formation involving an E. coli O157:H7 strain and a non-O157:H7 strain was also demonstrated.  相似文献

Self-illuminating quantum dots, also known as QD-BRET conjugates, are a new class of quantum dot bioconjugates which do not need external light for excitation. Instead, light emission relies on the bioluminescence resonance energy transfer from the attached Renilla luciferase enzyme, which emits light upon the oxidation of its substrate. QD-BRET combines the advantages of the QDs (such as superior brightness and photostability, tunable emission, multiplexing) as well as the high sensitivity of bioluminescence imaging, thus holding the promise for improved deep tissue in vivo imaging. Although studies have demonstrated the superior sensitivity and deep tissue imaging potential, the stability of the QD-BRET conjugates in biological environment needs to be improved for long-term imaging studies such as in vivo cell tracking. In this study, we seek to improve the stability of QD-BRET probes through polymeric encapsulation with a polyacrylamide gel. Results show that encapsulation caused some activity loss, but significantly improved both the in vitro serum stability and in vivo stability when subcutaneously injected into the animal. Stable QD-BRET probes should further facilitate their applications for both in vitro testing as well as in vivo cell tracking studies.  相似文献

Chironomids (Diptera) typically comprise the most abundant group of macroinvertebrates collected in water quality surveys. Species in the genus Cricotopus display a wide range of tolerance for manmade pollutants, making them excellent bioindicators. Unfortunately, the usefulness of Cricotopus is overshadowed by the difficulty of accurately identifying larvae using current morphological keys. Molecular approaches are now being used for identification and taxonomic resolution in many animal taxa. In this study, a sequence-based approach for the mitochondrial gene, cytochrome oxidase I (COI), was developed to facilitate identification of Cricotopus species collected from Baltimore area streams. Using unique COI sequence variations, we developed profiles for seven described Cricotopus sp., four described Orthocladius sp., one described Paratrichocladius sp. and one putative species of Cricotopus. In addition to providing an accurate method for identification of Cricotopus, this method will make a useful contribution to the development of keys for Nearctic Cricotopus.  相似文献

Conservation actions could be more efficient if there is congruence among taxa in the distribution of species. Patterns in the geographical distribution of five taxa were used to identify nationally important regions for conservation in Canada. Two measures of surrogacy were significantly and positively correlated among taxa, and conservation areas selected for one taxon represented other taxa significantly better than random selections. However, few large protected areas exist in the sites of highest conservation value in southern Canada; these regions are therefore a priority for future conservation regard. By focusing this effort on threatened and endangered species, which are a national priority in Canada, most other species could also benefit.  相似文献

Stable isotope (δ15N and δ13C) values of individual tooth annuli of female Steller sea lions ( n = 120) collected from the 1960s through the 1980s were used for retrospective analyses of temporal changes in food webs in the Gulf of Alaska and North Pacific Ocean. We also examined isotopically contour feathers of tufted puffins ( n = 135) and crested auklets ( n = 37) through this period to test for broader isotopic patterns indicative of whole food web changes. Steller sea lions decreased slightly in δ13C and increased in δ15N values, suggesting an increasing trophic level and change in foraging location or oceanographic isotopic signature. Steller sea lion first and second tooth annuli were enriched in 15N and depleted in 13C compared with subsequent annuli, indicating the effects of maternal influence through weaning. The general pattern of increasing δ15N values among Steller sea lions supports previous conclusions regarding a reduction or redistribution of forage fishes and an increase of demersal and semi-demersal species in the North Pacific ecosystem. There were no significant changes in δ15N values for either bird species. However, δ13C values in both bird species again suggested changes in foraging location or a shift in oceanographic currents.  相似文献

A novel caspase-like activity, which is directly regulated with cell proliferation is a candidate to regulate the abundance of the cyclin-dependent kinase inhibitor, p27(KIP1), in human lymphoid cells. This activity, which we term KIPase activity, can also cleave a subset of caspase substrates. Here we demonstrate that KIPase is a novel enzyme distinct from any of the previously characterized human caspases. We show that KIPase is active in a variety of cell lineages, its activity is associated with the proliferation of the human T-cell line, Jurkat, and is not inhibited by the broad spectrum caspase inhibitor z-VAD-fmk. Gel filtration analysis revealed that KIPase has a native molecular mass of approximately 100-200 kDa. Furthermore, the activity of KIPase does not change during apoptosis induced by either ligation of FAS or exposure of cells to etoposide. The uniqueness of KIPase is demonstrated by the fact that none of the human caspases tested (1-10) are able to cleave a specific KIPase substrate (Ac-DPSD-AMC) and that an aldehyde modified derivative of the DPSD tetra peptide is unable to inhibit caspases, but is a good inhibitor of KIPase activity. This supports a hypothesis whereby KIPase is a currently unidentified caspase-like enzyme which regulates the abundance of p27(KIP1) in a proliferation-dependent manner.  相似文献

A 3D modeling method to calculate the surface areas of coral branches   总被引:1,自引:1,他引:0  
The quantification of physiological and biochemical parameters in coral branches require normalization to a stable factor, such as the tissue biomass or surface area. Three dimensional (3D) animation software (Gmax®) was evaluated for estimating the surface area of simple coral branches. The software was highly predictive of the known surface areas of small (20–60 mm long) plastic rods and cones (r 2 > 0.99), and of small (30–60 mm) Acropora millepora branches (r 2 = 0.98) whose surface area had been obtained using the traditional wax-weight method. Two normalization parameters, 3D modeled surface area and tissue biomass (measured as protein), were then compared for A. millepora branches collected in summer and winter. In winter, protein and surface area were correlated (r 2 > 0.61), but not in summer, indicating that choice of normalizing parameter will influence the outcome of experimental analyses.  相似文献

A small molecule that safely mimics the ability of dietary restriction (DR) to delay age-related diseases in laboratory animals is greatly sought after. We and others have shown that resveratrol mimics effects of DR in lower organisms. In mice, we find that resveratrol induces gene expression patterns in multiple tissues that parallel those induced by DR and every-other-day feeding. Moreover, resveratrol-fed elderly mice show a marked reduction in signs of aging, including reduced albuminuria, decreased inflammation, and apoptosis in the vascular endothelium, increased aortic elasticity, greater motor coordination, reduced cataract formation, and preserved bone mineral density. However, mice fed a standard diet did not live longer when treated with resveratrol beginning at 12 months of age. Our findings indicate that resveratrol treatment has a range of beneficial effects in mice but does not increase the longevity of ad libitum-fed animals when started midlife.  相似文献
