Meiosis creates genetic diversity by recombination and segregation of chromosomes. The synaptonemal complex assembles during meiotic prophase I and assists faithful exchanges between homologous chromosomes, but how its assembly/disassembly is regulated remains to be understood. Here, we report how two major posttranslational modifications, phosphorylation and ubiquitination, cooperate to promote synaptonemal complex assembly. We found that the ubiquitin ligase complex SCF is important for assembly and maintenance of the synaptonemal complex in Drosophila female meiosis. This function of SCF is mediated by two substrate-recognizing F-box proteins, Slmb/βTrcp and Fbxo42. SCF-Fbxo42 down-regulates the phosphatase subunit PP2A-B56, which is important for synaptonemal complex assembly and maintenance. 相似文献

TiO2 nanoparticles hazard is associated to their photocatalytic activity causing release of DNA damaging ROS (Reactive Oxygen Species), lipid peroxidation and skin damage. Various coatings have been proposed to minimize photocatalysis, while keeping the potential to block UV radiations. Uncoated and variously coated commercial nano-titania have been classified on the basis of UVB-induced lipoperoxidation of linoleic acid. A selection of the most and the least protective specimens was then examined by ESR (Electron Spin Resonance) to evidence the presence of surface paramagnetic centres and the release of ROS in aqueous suspensions (spin trapping). Paramagnetic centres and ROS were correlated with the extent of lipid peroxidation. When tested on porcine skin (mimicking the human one), titania acted as on linoleic acid. The combined use of lipid peroxidation of simple fatty acids with ESR analysis is here proposed as a possible screening tool for the evaluation of the potential toxicity of nano-titania in sunscreen preparations. 相似文献

In the present study we tested the ability of different antioxidant agents, used alone or in combination, to reduce the reactive oxygen species (ROS) levels and to increase the glutathione peroxidase (GPx) activity. Moreover, we tested the ability of such antioxidant agents to reduce the serum levels of proinflammatory cytokines IL-6 and TNF f . Fifty-six advanced stage cancer patients with tumors at different sites were included in the study: they were mainly stage III (12.5%) and stage IV (82.1%). The study was divided into two phases. In the 1 st phase 28 patients were divided into five groups and a single different antioxidant agent was administered to each group. The selected antioxidant agents were: alpha lipoic acid or carboxycysteine-lysine salt, amifostine, reduced glutathione, vitamin A plus vitamin E plus Vitamin C. In the 2 nd phase of the study 28 patients were divided into five groups and a combination of two different antioxidant agents was administered to each group. The antioxidant treatment was administered for 10 consecutive days. The patients were studied at baseline and after antioxidant treatment. Our results show that all single antioxidants tested were effective in reducing the ROS levels and three of them in increasing GPx activity, too. Among the combinations of antioxidant agents, three were effective in reducing ROS, while three were effective in increasing GPx activity (arm 4 was effective in both instances). Comprehensively, the "antioxidant treatment" was found to be effective both on ROS levels and GPx activity. Moreover, the antioxidant treatment was able to reduce serum levels of IL-6 and TNF f . Furthermore, a correlation was shown between the Eastern Cooperative Oncology Group Performance Status of patients and blood levels of ROS, GPx activity, serum levels of proinflammatory cytokines. 相似文献

Summary Bupleurum dianthifolium Guss., an endemic species of the small island of Marettimo just West of Sicily, is one of the few constantly hermaphroditic Umbelliferae. It is markedly protandrous. Its chromosome number is n = 8. The flower develops acropetally from the petals to the stamens, and the carpels; the development of the sepals stops very soon. The microspores individualise simultaneously and the reproductive nucleus divides while the pollen grain is still in the anther; the tapetum is of the secretory type. The two carpels fuse at their tips and the ovary is divided into two chambers by the carpophore which develops from the bottom central part of the cavity. Two ovules start developing in each chamber; the upper ones cease their development while the lower ones grow into pendulous, anatropous ovules with only one integument. The ovule is tenuinucellate. The archesporium is unicellular. No parietal cell is formed. A group of cells trophic in function is present at the basis of the embryo sac, and disappears after fertilisation has occurred. The female gametophyte is of the normal type, monosporic, 8-nucleated, with the antipodal cells sunken in the chalazal nutritive apparatus and disappearing before the polar nuclei fuse. The albumen is nuclear. A fibrillary body has been observed in the cytoplasm of the megaspore mother-cell. This body after the meiosis remains as an exclusive property of the chalazal megaspore, i. e. the spore that will give the embryo sac. This body can still be seen in the binucleate gametophyte, and disappears during the 4-nucleated gametophyte stage. 相似文献
