Lignin confers recalcitrance to plant biomass used as feedstocks in agro‐processing industries or as source of renewable sugars for the production of bioproducts. The metabolic steps for the synthesis of lignin building blocks belong to the shikimate and phenylpropanoid pathways. Genetic engineering efforts to reduce lignin content typically employ gene knockout or gene silencing techniques to constitutively repress one of these metabolic pathways. Recently, new strategies have emerged offering better spatiotemporal control of lignin deposition, including the expression of enzymes that interfere with the normal process for cell wall lignification. In this study, we report that expression of a 3‐dehydroshikimate dehydratase (QsuB from Corynebacterium glutamicum) reduces lignin deposition in Arabidopsis cell walls. QsuB was targeted to the plastids to convert 3‐dehydroshikimate – an intermediate of the shikimate pathway – into protocatechuate. Compared to wild‐type plants, lines expressing QsuB contain higher amounts of protocatechuate, p‐coumarate, p‐coumaraldehyde and p‐coumaryl alcohol, and lower amounts of coniferaldehyde, coniferyl alcohol, sinapaldehyde and sinapyl alcohol. 2D‐NMR spectroscopy and pyrolysis‐gas chromatography/mass spectrometry (pyro‐GC/MS) reveal an increase of p‐hydroxyphenyl units and a reduction of guaiacyl units in the lignin of QsuB lines. Size‐exclusion chromatography indicates a lower degree of lignin polymerization in the transgenic lines. Therefore, our data show that the expression of QsuB primarily affects the lignin biosynthetic pathway. Finally, biomass from these lines exhibits more than a twofold improvement in saccharification efficiency. We conclude that the expression of QsuB in plants, in combination with specific promoters, is a promising gain‐of‐function strategy for spatiotemporal reduction of lignin in plant biomass.  相似文献

Bombesin, substance P and several structurally related peptides cause excessive grooming behavior after intracerebroventricular injection in mice. The present study describes the behavioral characteristics of these effects after acute administration. Substance P caused an elevation of grooming behavior which was short-lasting (less than 15 minutes), while bombesin induced both grooming and scratching behavior with a duration of action of about 2.5 hours. After repeated injections of high doses of either bombesin or a metabolically stable substance P analog, no tolerance-formation to these peptide-induced effects could be observed. Morphine partially antagonized bombesin-induced behaviors at a dose of 7.5 mg/kg subcutaneously while the same dose did not attentuate substance P-induced grooming. These results suggest that the behavioral changes induced by substance P and bombesin are mediated by distinct mechanisms. The lack of tolerance formation, together with the partial antagonism by morphine, suggests that the bombesin-induced behaviors may be related to a stimulation of nociceptive mechanisms.  相似文献

We evaluated the effects of the exotic tree Fraxinus uhdei on decomposition dynamics and nutrient turnover in a montane Hawaiian rainforest. We used reciprocal transplants of litterbags between forests dominated by Fraxinus and by the native Metrosideros polymorpha to distinguish between endogenous (litter quality) and exogenous (for example, microclimate, nutrient availability, microbial and invertebrate communities) effects of Fraxinus on mass loss and nutrient dynamics of decomposing litter. Fraxinus produced greater quantities of litter that was thinner, had higher N and P concentrations, and lower concentrations of lignin and soluble polyphenols. Microbes decomposing Fraxinus litter produced fewer enzymes involved in N and P acquisition and more of those involved in cellulose degradation. Differences in litter quality and microbial activity resulted in a strong effect of litter type on rates of mass loss, whereby Fraxinus litter decomposed and released nutrients at nearly twice the rate of Metrosideros litter (k=0.82 versus 0.48), regardless of site of decomposition. Although site of decomposition had no effect on rates of litter mass loss, Fraxinus litter decomposed under a Fraxinus canopy mineralized approximately 20% less P after one year than Fraxinus litter decomposed under a Metrosideros canopy. Furthermore, Fraxinus litter decomposed under a Fraxinus canopy immobilized greater amounts of N and P in the early stages of decay, suggesting that the large amounts of N and P in Fraxinus litterfall have raised nutrient availability to decomposers in the forest floor. Greater immobilization of N and P under a Fraxinus canopy may act as a governor on rates of nutrient cycling, limiting the degree to which Fraxinus invasion accelerates N and P cycling in this system.  相似文献

Lomatium is the largest North American genus ofApiaceae (=Umbelliferae). Relations amongLomatium and other genera have not been well understood. TheApiaceae-Araliaceae lineage and genera of North AmericanApiaceae were analyzed phenetically and cladistically in turn, and seven phenetic alliances of genera are identified. Data on frequencies of character-state combinations were assembled and usingSporne's notions of maximal and minimal co-variation of states, two character-states were identified that are derived among Umbellifer genera. Using cladistic analysis of the alliance consisting ofLomatium and six other genera we identified three sister groups toLomatium:Polytaenia alone;Polytaenia withPseudocymopterus, and a group of four genera. Not all genera in the group of seven withLomatium are supported by autapomorphies impeding phylogenetic reconstructions. Both phenetics and cladistics have helped to suggest and evaluate relationships; to call attention to groups requiring further study, and in particular to identify derived character-states.  相似文献

Micropipette aspiration (MA) has been widely used to measure the biomechanical properties of cells and biomaterials. To estimate material parameters from MA experimental data, analytical half-space models and inverse finite element (FE) analyses are typically used. The half-space model is easy to implement but cannot account for nonlinear material properties and complex geometrical boundary conditions that are inherent to MA. Inverse FE approaches can account for geometrical and material nonlinearities, but their implementation is resource-intensive and not widely available. Here, by making analogy between an analytical uniaxial tension model and a FE model of MA, we proposed an easily implementable and accurate method to estimate the material parameters of tissues tested by MA. We first adopted a strain invariant-based isotropic exponential constitutive model and implemented it in both the analytical uniaxial tension model and the FE model. The two models were fit to experimental data generated by MA of porcine aortic valve tissue (45 spots on four leaflets) to estimate material parameters. We found no significant differences between the effective moduli estimated by the two models ( $p > 0.39$ ), with the effective moduli estimated by the uniaxial tension model correlating significantly with those estimated by the FE model ( $p < 0.001; R^{2}= 0.96$ ) with a linear regression slope that was not different than unity ( $p = 0.38$ ). Thus, the analytical uniaxial tension model, which avoids solving resource-intensive numerical problems, is as accurate as the FE model in estimating the effective modulus of valve tissue tested by MA.  相似文献

It is generally accepted that postcopulatory sexual selection drives rapid divergence of genital morphology among isolated populations. The mode of selection operating upon genitalia can be explored by comparing patterns of population divergence in genetic and genitalic traits. We collected Antichiropus variabilis millipedes from eight localities across the species range. Levels of among-population genetic divergence, at microsatellite loci, and the mitochondrial COI gene were very high. Following geometric morphometric analyses, genital morphology was also found to be highly divergent among the populations surveyed, whereas head morphology had not diverged as markedly. However, pairwise comparisons of F(ST) and P(ST) showed that among-population divergence in both genital and head shape was significantly lower than that experienced by neutral genetic markers. Our results suggest that the genitalia of A. variabilis are currently experiencing a period of stabilizing selection, the mode of selection expected for genitalia that function in species recognition via a "lock-and-key" mechanism. Our results demonstrate that although genital morphology can clearly diverge among genetically isolated populations, divergence is not necessarily as rapid as commonly argued, and continuous directional sexual selection may not always underpin the evolutionary divergence of male genitalia.  相似文献

A defense-inducible maize gene was discovered through global mRNA profiling analysis. Its mRNA expression is induced by pathogens and defense-related conditions in various tissues involving both resistant and susceptible interactions. These include Cochliobolus heterostrophus and Cochliobolus carbonum infection, ultraviolet light treatment, the Les9 disease lesion mimic background, and plant tissues engineered to express flavonoids or the avirulence gene avrRxv. The gene was named Zm-mfs1 after it was found to encode a protein related to the major facilitator superfamily (MFS) of intregral membrane permeases. It is most closely related to the bacterial multidrug efflux protein family, typified by the Escherichia coli TetA, which are proton motive force antiporters that export antimicrobial drugs and other compounds, but which can be also involved in potassium export/proton import or potassium re-uptake. Other related plant gene sequences in maize, rice, and Arabidopsis were identified, three of which are introduced here. Among this new plant MFS subfamily, the characteristic MFS motif in cytoplasmic TM2-TM3 loop, and the antiporter family motif in transmembrane domain TM5 are both conserved, however the TM7 and the cytoplasmic TM8-TM9 loop are divergent from those of the bacterial multidrug transporters. We hypothesize that Zm-Mfs1 is a prototype of a new class of plant defense-related proteins that could be involved in either of three nonexclusive roles: (1) export of antimicrobial compounds produced by plant pathogens; (2) export of plant-generated antimicrobial compounds; and (3) potassium export and/or re-uptake, as can occur in plant defense reactions.  相似文献

1. Behavioral experiments with jittering echoes examined acoustic images of sonar targets in the echolocating bat, Eptesicus fuscus, along the echo delay or target range axis. Echo phase, amplitude, bandwidth, and signal-to-noise ratio were manipulated to assess the underlying auditory processes for image formation. 2. Fine delay acuity is about 10 ns. Calibration and control procedures indicate that this represents temporal acuity rather than spectral discrimination. Jitter discrimination curves change in phase when the phase of one jittering echo is shifted by 180 degrees relative to the other, showing that echo phase is involved in delay estimation. At an echo detectability index of about 36 dB, fine acuity is 40 ns, which is approximately as predicted for the delay accuracy of an ideal receiver. 3. Compound performance curves for 0 degrees and 180 degrees phase conditions match the crosscorrelation function of the echoes. The locations of both 0 degrees and 180 degrees phase peaks in the performance curves shift along the time axis by an amount that matches neural amplitude-latency trading in Eptesicus, confirming a temporal basis for jitter discrimination.  相似文献
