Analysis of Dysgenesis-Induced Lethal Mutations on the X Chromosome of a Q Strain of DROSOPHILA MELANOGASTER 总被引:4,自引:1,他引:3 下载免费PDF全文
The Q strain known as v6 was tested for its ability to induce X-linked lethal mutations in male and female hybrids from crosses with M strains in the P-M system of hybrid dysgenesis. All measurements of the mutation rate were made on the X chromosome derived from the v6 strain. The lethal rate for young hybrid males from the cross M female X v6 male was 1.11% per chromosome. For older males, it was only 0.44%, suggesting that there is less mutational or more repair activity in the germ cells of the older males or that mutant cells are selectively eliminated as the hybrid males age. The lethal rate for hybrid females from comparable crosses was approximately the same for both ages that were tested. However, it was substantially less than the rate for the hybrid males--only 0.26% per chromosome. Genetically identical hybrid females from reciprocal crosses also showed a low mutation rate, 0.13% per chromosome. Again, there was no difference between young and old flies. Mapping experiments established that most of the lethal mutations that were recovered from the male and female hybrids were located in two regions on the X chromosome, one between bands 14B13 and 15A9 , the other between bands 19A1 and 20A , which encompasses the maroonlike locus. More refined mapping of the lethals in the maroonlike region demonstrated that the vast majority of these affected a single gene located in band 19C4 . Cytological analysis of the lethal chromosomes revealed that several carried rearrangements, including inversions, duplications and deficiencies. Chromosome breakage occurred primarily in bands 14D1 -3 and 18F- 20A , and most of the breaks in the latter segment were located in 19C . However, rearrangements involving 19C and mutations of the gene in 19C4 were mutually exclusive events. In situ hybridization of a P element probe to the chromosomes of v6 demonstrated that P elements reside at a minimum of five sites on the X chromosome. These P element sites correspond to the mutational and breakage hot spots on that chromosome. The combined genetic and cytological data imply that most of the X-linked lethal mutations that occur in M X v6 hybrids are due to local P element action. Consideration of these and other data suggest that v6 is a weak P strain in the P-M system of hybrid dysgenesis and that other Q strains might also be regarded in this way. 相似文献

Frequency selectivity of hearing was measured in the green treefrog, Hyla cinerea. A psychophysical technique based on reflex modification was used to obtain masked threshold estimates for pure tones (300-5,400 Hz) presented against two levels of broadband masking noise. A pure tone (S-1) presented 200 ms prior to a reflex-eliciting stimulus (S-2) inhibited the motor reflex response to S-2. The magnitude of this reflex modification effect varied systematically with the sound pressure level (SPL) of S-1, and threshold was defined as the SPL of S-1 at which the reflex modification effect disappeared. Masked thresholds were used to calculate critical ratios, an index of the auditory system's frequency selectivity. The frequency selectivity of the treefrog's hearing is greatest and critical ratios are lowest (22-24 dB) at about 900 and 3,000 Hz, the two spectral regions dominant in the male treefrog's species-specific advertisement call. These results suggest that the treefrog's auditory system may be specialized to reject noise at biologically-relevant frequencies. As in other vertebrates, critical ratios remain constant when background noise level is varied; however, the shape of the treefrog's critical ratio function across frequencies differs from the typical vertebrate function that increases with increasing frequency at a slope of about 3 dB/octave. Instead, the treefrog's critical ratio function resembles its pure tone audiogram. Although the shape of the treefrog's critical ratio function is atypical, the critical ratio values themselves are comparable to those of many other vertebrates in the same frequency range. The kinetics of three physical parameters: cell density, relative cytoplasmic viscosity and DNA stability to denaturation have been measured during the period preceding cell death induced by hyperthermia, methylprednisolone and a series of cancer chemotherapeutic agents. This series of measurements employed cultured human lymphoblastoid cells as an experimental system to establish the changes that can be observed in the early stages of cell death, prior to applying such measurements to tissue biopsies from solid human tumours. Cell death, induced by hyperthermia up to 43°C, methylprednisolone, vincristine, 5-fluorouracil, BCNU and melphalan, showed essentially identical and reproducible changes corresponding to those which characterize programmed cell death (apoptosis). Such changes could also be observed following hyperthermia above 43°C, but reproducibility was poor and increasing damage to the cell membranes was evident. In cells treated with adriamycin or methotrexate, cell sub-populations showing an increase in cell density were not detected. Measurements of DNA stability were readily performed by flow cytofluorometry thus allowing rapid quantitation of the fraction of cells in the early stages of cell death. Modified flow cytometric instrumentation would further allow measurement of cytoplastic viscosity as an additional parameter to indicate entry into programmed cell death. This suggests that these measurements could readily be applied to cell suspensions derived from tumour tissue biopsies for a more accurate assessment of tumour growth rate, and to allow monitoring of response to therapy in sequential tumour biopsies. 相似文献

Twenty ward stock bottles of aqueous 1% Printol were examined and 17 were found to be contaminated with Alcaligenes faecalis. Organisms were present in dead space behind the plastic liners of the bottle caps, where they could have survived washing. A. faecalis was also isolated from 31 out of 34 1% Printol solutions in use in the hospital. Pseudomonas aeruginosa was grown from two samples of 1% Printol in one ward, but not from stock bottles.The minimal bactericidal concentration (M.B.C.) in aqueous solution of Hycolin, Sudol, and Stericol for A. faecalis and Ps. aeruginosa was 1 in 320. The M.B.C. of Printol for both organisms was 1 in 80. The activity of all four disinfectants was reduced in the presence of large amounts of organic matter. Sudol was the least affected. Polyethylene, of which stock bottles were made, did not reduce the activity of the disinfectants. It is suggested that, ideally, stock bottles of disinfectant diluted ready for use should be autoclaved before they are refilled. 相似文献

Insect sperm flagella, however, usually contain in addition to the typical 9+2 arrangement of tubules, an additional row of nine singlet tubules which are called accessory tubules. Each of these accessory tubules in the Colorado potato beetle (Leptinotarsa decemlineata (Say)) contains a single central prototubule surrounded by an array of 15 or 16 peripheral prototubules. 1. A series of oligosaccharides was isolated from Salmonella milwaukee lipopolysaccharide by partial acid hydrolysis. 2. Structural studies on these oligosaccharides indicated that the O-specific side chain of this lipopolysaccharide has a repeating pentasaccharide unit that is probably alpha-d-galactosyl-(1-->3)-beta-d-galactosyl- (1-->3)-N-acetylgalactosaminyl-(1-->3)-N-acetyl- d-glucosaminyl-(1-->4)-l-fucose. 3. Another oligosaccharide, which is not structurally related to the repeating pentasaccharide unit, has also been isolated and it is indistinguishable from an oligosaccharide obtained from Salmonella ;rough' (R) lipopolysaccharides. The isolation of this and similar core oligosaccharides from all chemotype VI lipopolysaccharides supports the view that Salmonella S-lipopolysaccharides have a common core that is probably identical with RII lipopolysaccharide. 相似文献

- Male bullfrogs at two different natural calling sites were presented with playbacks of synthetic advertisement calls differing in phase spectra. Sounds were presented in a ABA design to analyze the ability of the animals to perceive changes in repeated series of stimuli.
- The number of individual croaks in an answering call significantly increased over repeated presentations of two of the three stimulus phase types in condition A1. There were significantly fewer croaks to the third stimulus. These data suggest that two stimuli were perceived in a similar manner.
- Latency of calling to stimuli presented in conditions A and B changed in response to shifts in phase spectrum at a low density calling site. These differences were significant when comparing latency to playbacks where shifts in the phase spectrum changed the temporal fine-structure and waveform periodicity of the stimulus.
- The increase in number of croaks and decrease in response latency across condition A1 and the increase in latency in condition B suggest that discrimination may take the form of stimulus-specific sensitization. In this context, sensitization might reflect an increase in arousal due to repeated presentation of a salient stimulus.
- The operation of a hypothetical ‘mating call detector,’ based on linear summation of temporal responses from the eighth nerve, provides output similar to the behavioral results.

Contamination in a trace element laboratory can come from a variety of sources, including laboratory gloves. Therefore, vinyl
and latex gloves were obtained from as many manufacturers as would supply gloves. These gloves were either prepared for acid-washing
and subsequent soaking in an acid solution, or immersed in an acid solution for a duration of either 1 min or 1 h. Incubation
washes were analyzed for a variety of trace elements by flame atomic abosrption spectroscopy (AAS) or inductively coupled
mass spectrometry (ICP-MS). Results indicated that only three brands of vinyl gloves were acceptable for use in a trace element
laboratory, whereas others had contamination of different elements. Latex gloves contained such high levels of biologically
important elements that they were not considered suitable for routine trace element work. Vinyl gloves of choice should be
