Connexin mimetic peptides are widely used to assess the contribution of nonjunctional connexin channels in several processes, including ATP release. These peptides are derived from various connexin sequences and have been shown to attenuate processes downstream of the putative channel activity. Yet so far, no documentation of effects of peptides on connexin channels has been presented. We tested several connexin and pannexin mimetic peptides and observed attenuation of channel currents that is not compatible with sequence specific actions of the peptides. Connexin mimetic peptides inhibited pannexin channel currents but not the currents of the channel formed by connexins from which the sequence was derived. Pannexin mimetic peptides did inhibit pannexin channel currents but also the channels formed by connexin 46. The same pattern of effects was observed for dye transfer, except that the inhibition levels were more pronounced than for the currents. The channel inhibition by peptides shares commonalities with channel effects of polyethylene glycol (PEG), suggesting a steric block as a mechanism. PEG accessibility is in the size range expected for the pore of innexin gap junction channels, consistent with a functional relatedness of innexin and pannexin channels. mimetic peptide; polyethylene glycol; calcium wave 相似文献

We describe two new species of esthonioporate bryozoans, Revalotrypa inopinata sp. nov. and R. yugaensis sp. nov., from the Lower Ordovician, south-east of Lake Ladoga, north-western Russia. The colonies of the two species are very small and were extracted from limestone nodules found in the lower part of glauconitic sandstone in the Joa Member (lowermost Floian). Revalotrypa inopinata sp. nov. and R. yugaensis sp. nov. are the oldest known bryozoans from Baltoscandia. Study of the two species includes scanning electron microscopy (SEM) imaging and X-ray microtomography (micro-CT). The palaeogeographic distribution of the oldest known bryozoans from the Lower Ordovician and the advantages and disadvantages of micro-CT for the study of Palaeozoic bryozoans are discussed. The authorship of Revalotrypidae, which has been inconsistently allocated in the literature, is discussed. 相似文献

Injury to the central nervous system triggers glial calcium waves in both vertebrates and invertebrates. In vertebrates the pannexin1 ATP-release channel appears to provide for calcium wave initiation and propagation. The innexins, which form invertebrate gap junctions and have sequence similarity with the pannexins, are candidates to form non-junctional membrane channels. Two leech innexins previously demonstrated in glia were expressed in frog oocytes. In addition to making gap junctions, innexins also formed non-junctional membrane channels with properties similar to those of pannexons. In addition, carbenoxolone reversibly blocked the loss of carboxyfluorescein dye into the bath from the giant glial cells in the connectives of the leech nerve cord, which are known to express the innexins we assayed. 相似文献

EVI5 protein associates with the INCENP-aurora B kinase-survivin chromosomal passenger complex and is involved in the completion of cytokinesis 总被引:1,自引:0,他引:1
EVI5 has been shown to be a novel centrosomal protein in interphase cells. In this report, we demonstrate using immunofluorescence microscopy that EVI5 has a dynamic distribution during mitosis, being associated with the mitotic spindle through anaphase and remaining within the midzone and midbody until completion of cytokinesis. Knockdown of EVI5 using siRNA results in a multinucleate phenotype, which is consistent with an essential role for this protein in the completion of cytokinesis. The EVI5 protein also undergoes posttranslational modifications during the cell cycle, which involve phosphorylation in early mitosis and proteolytic cleavage during late mitosis and cytokinesis. Since the subcellular distribution of the EVI5 protein was similar to that characteristic of chromosomal passenger proteins during the terminal stages of cytokinesis, we used immunoprecipitation and GST pull-down approaches to demonstrate that EVI5 is associated with the aurora B kinase protein complex (INCENP, aurora B kinase and survivin). Together, these data provide evidence that EVI5 is an essential component of the protein machinery facilitating the final stages of cell septation at the end of mitosis. 相似文献

The 2002 discovery of the earliest European modern humans in the Peştera cu Oase (Romania) has led to systematic excavations of a bone bed (two levels), containing principally the remains of cave bears (ca. 5000 bones) and dated between 50 and 40 ka cal BP. The thanatocenosis corresponds mainly to a subadult and adult male utilization of the cavity. Levels 1 and 2 are distinguishable by different conservation processes demonstrating respectively low- and high-energy hydraulic transports. The heavy component of cannibalism may indicate bone processing for trace elements and/or a more omnivorous diet than is usually ascribed to cave bears. To cite this article: J. Quilès et al., C. R. The calyx of Held synapse is a giant axosomatic synapse that has a fast relay function within the sound localization circuit of the brainstem. In the adult, each principal neuron of the medial nucleus of the trapezoid body (MNTB) is contacted by a single calyx terminal. In rodents, the calyx of Held synapse forms around the third postnatal day (P3). Here, we studied the developmental changes in the intrinsic excitability of the principal neurons during the first postnatal week by making whole-cell recordings from brainstem slices. In slices from P0-1 rats, about 20% of the principal neurons were spontaneously active, whereas after P3, no spontaneously active cells were observed. Already at P0, principal neurons received both glutamatergic and GABAergic/glycinergic inputs. The occurrence of spontaneous action potentials depended upon the presence of spontaneous glutamatergic inputs; summation of only a few quanta was enough to reach action potential threshold. The main cause for this high excitability was a high resting membrane resistance, which decreased at least four-fold during the first postnatal week. A relatively slow decay of synaptic currents and a relatively depolarized membrane potential may have contributed as well. We conclude that the decrease in the excitability of principal neurons in the MNTB matches the increase of the strength of the synaptic inputs resulting from the formation and maturation of the calyx of Held synapse during the first postnatal week. This decrease in excitability will make it progressively more difficult for non-calyceal inputs to trigger action potentials. 相似文献

Efficient, practical and accurate estimates of population parameters are a necessary basis for effective conservation action to meet biodiversity targets. The brown hare is representative of many European farmland species: historically widespread and abundant but having undergone rapid declines as a result of agricultural intensification. As a priority species in the UK Biodiversity Action Plan, it has national targets for population increase that are part of wider national environmental indicators. Previous research has indicated that brown hare declines have been greatest in pastural landscapes and that gains might be made by focussing conservation effort there. We therefore used hares in pastural landscapes to examine how basic changes in survey methodology can affect the precision of population density estimates and related these to national targets for biodiversity conservation in the UK. Line transects for hares carried out at night resulted in higher numbers of detections, had better-fitting detection functions and provided more robust density estimates with lower effort than those during the day, due primarily to the increased probability of detection of hares at night and the nature of hare responses to the observer. Hare spring densities varied widely within a single region, with a pooled mean of 20.6 hares km(-2), significantly higher than the reported national average of hares in pastures of 3.3 hares km(-2). The high number of encounters allowed us to resolve hare densities at site, season and year scales. We demonstrate how survey conduct can impact on data quantity and quality with implications for setting and monitoring biodiversity targets. Our case study of the brown hare provides evidence that for wildlife species with low detectability, large scale volunteer-based monitoring programmes, either species specific or generalist, might be more successfully and efficiently carried out by a small number of trained personnel able to employ methods that maximise detectability. 相似文献

This paper presents two new experimental results: the protective effect of green light (GL) on ultraviolet (UV) denaturation
of proteins, and the effect of GL on protein macromolecular structures. The protective effect of GL was revealed on two serum
albumins, bovine (BSA) and human (HSA), and recorded by electrophoresis, absorption, and circular dichroism spectra. The effect
of GL irradiation on protein structure was recorded by using fluorescence spectroscopy and electrophoresis. These new effects
were modeled by quantum-chemistry computation using Gaussian 03W, leading to good fit between theoretical and experimental
absorption and circular dichroism spectra. A mechanism for these phenomena is suggested, based on a double-photon absorption
process. This nonlinear effect may lead to generation of long-lived Rydberg macromolecular systems, capable of long-range
interactions. These newly suggested systems, with macroscopic quantum coherence behaviors, may block the UV denaturation processes. 相似文献
