This study was designed to investigate the physical characteristics and crystalline structure of 2-hydroxy-N-[3(5)-pyrazolyl]-1,4-naphthoquinone-4-imine (PNQ), a new active compound against Trypanosoma cruzi, the causative agent of American trypanosomiasis. Methods used included differential scanning calorimetry, thermogravimetry, hot stage microscopy, polarized light microscopy (PLM), Fourier-transform infrared (FTIR) spectroscopy, and high-resolution X-ray powder diffraction (HR-XRPD). According to PLM and HR-XRPD data, PNQ crystallized as red oolitic crystals (absolute methanol) or prisms (dimethyl sulfoxide [DMSO]-water) with the same internal structure. The findings obtained with HR-XRPD data (applying molecular location methods) showed a monoclinic unit cell [a = 18.4437(1) A, b = 3.9968(2) A, c = 14.5304(1) A, alpha = 90 degrees , beta = 102.71(6) degrees , gamma = 90 degrees , V = 1044.9(1) A(3), Z = 4, space group P2(1)/c], and a crystal structure (excluding H-positions) described by parallel layers in the direction of the b-axis, with molecules held by homochemical (phenyl-phenyl and pyrazole-pyrazole) van der Waals interactions. In addition, FTIR spectra displayed the NH-pyrazole stretch overlapped with the OH absorption at 3222 cm(-1), typical of -NH and -OH groups associated through H-bondings; and a carbonyl stretching absorption at 1694 cm(-1), indicating a nonextensively H-bonded quinonic C=O, which was in accordance with the solved crystal structure of PNQ. The existence of such cohesive forces shed light on the thermoanalytical data, which revealed that PNQ is a stable solid, unaffected by oxygen that decomposed without melting above 260 degrees C.  相似文献

1. We wished to further study the behavioral effects of -melanotropin (-MSH), melanin-concentrating hormone (MCH), and neuropeptide glutamine–isoleucine (NEI).2. To this effect we administered -MSH, MCH, and NEI in the ventral tegmental area of the rat, a structure where these neuropeptides are highly concentrated. To further elucidate the biochemical mechanisms of the behavioral effect of these neuropeptides, we determined the degree of grooming behavior and the levels of catecholamines, after neuropeptide administration.3. We preselected those animals responding to the central injection of -MSH with excessive grooming behavior. We administered the neuropeptides at the dose of 1 g/0.5 L, in each side of the ventral tegmental area, bilaterally. We studied grooming behavior, locomotor activity, and total behavior scores, 30 and 65 min after administration of the peptides.4. Three groups of animals were decapitated immediately after the injection of the neuropeptides, and 30 or 65 min after injection. We measured dopamine (DA), noradrenaline (NA), and the dopac/dopamine ratio (DOPAC/DA) to determine steady state levels of catecholamines and an indirect measure of DA release and metabolism, respectively.5. Injections of -MSH produced significant elevations in grooming behavior, locomotor activity, and total behavior scores, both 30 and 65 min after peptide administration. This was correlated with significant decreases in DA content, increases in DOPAC content, and increases in the DOPAC/DA ratio. In the caudate putamen, changes in catecholamines occurred both at 30 and 65 min after injection. In the nucleus accumbens, changes were present at 65 min after injection. Conversely, there were no alterations in NA content, either in the caudate putamen or in the nucleus accumbens, at any time after the injection.6. Injections of NEI resulted in significant elevations in grooming behavior, locomotor activity, and total behavior scores, both 30 and 65 min after peptide administration. This was correlated with increased DOPAC/DA ratio in the nucleus caudatus but not in the nucleus accumbens. Conversely, NEI produced increased NA concentrations in the nucleus accumbens, but not in the nucleus caudatus.7. Injections of MCH did not produce significant changes in behavior or significant changes in nucleus caudatus or nucleus accumbens catecholamines.8. Our results indicate (a) There is a correlation with alterations in behavior as induced for the neuropeptides injected here, and changes in extrapyramidal catecholamines. (b) There is a correlation between alterations in behavior and increases in DOPAC/DA ratio in the nucleus caudatus. (c) There is a correlation between alterations in behavior and alterations in catecholamines in the nucleus accumbens. In the nucleus accumbens, DOPAC/DA ratio is changed after -MSH, and NA ratio is changed after NEI injection. (d) Absence of alterations in extrapyramidal catecholamines, and in particular in catecholamines in the nucleus accumbens, correlates with absence of behavioral alterations after neuropeptide administration to the ventral tegmental area.9. In conclusion, the behavioral effect of exogenous administration of neuropeptides in the ventral tegmental area is peptide-specific, and is probably associated with alterations in catecholamine metabolism and release in the nucleus caudatus and the nucleus accumbens. Both -MSH and NEI seem to stimulate the nigrostriatal DA system. While -MSH appears to stimulate the mesolimbic DA system as well, NEI may exert its actions not through the DA, but through the NA mesolimbic system. The precise contribution of DA and NA, and the relative role of the nucleus caudatus and nucleus accumbens in these behaviors remain to be elucidated.  相似文献

Abstract

The Yacoraite Formation (Maastrichtian–Danian; Balbuena Subgroup; Salta Group), from Northwestern Argentina, represents a shallow epeiric unit which is the result of transgressions in the Andean basin of South America. Herein, we study the avian footprints from the Maimará locality, Jujuy province, and Quebrada del Tapón ichnosite, Salta province. The avian footprints from the Maimará locality is less diverse, made up: cf. Alaripeda isp., Avipeda isp., cf. Gruipeda filiportatis, and cf. Gruipeda isp. The avian track record from the Quebrada del Tapón ichnosite is composed of: cf. Alaripeda isp., Ardeipeda cf. egretta., Gruipeda filiportatis (=Yacoraitichnus avis) and others Gruipeda or compared to this ichnogenus, cf. Uhangrichnus isp., and several indetermined avian footprints. Both ichnological assemblages have tracks assigned to shorebirds or Charadriiformes. The facies distribution of trace fossils reinforces the overall model of a shorebird tracks dominance in moderate to low energy settings. This distribution can be divided into three sub-environments: (1) a moderate to high energy shoreline under wave action, (2) a supratidal body of ephemeral ponds, far away from direct wave influence and (3) a landward position, beyond the ephemeral ponds system. We have included all the ichnoassemblages within the shorebird ichnosubfacies as a subset of the Scoyenia Ichnofacies.  相似文献

The high impact of predators on breeding success has favored the evolution of diverse behavioral strategies that enhance both individual survivorship and reproductive success. Self‐ and nest‐defense against predators are inter‐related behaviors that vary according to life history traits and the risks involved in defense. In addition, interpopulation differences in the composition of predator guilds, and hence predation pressures, may result in geographic variation in antipredator defense strategies. It is generally accepted that the absence of predators could drive the loss of antipredator behavior. However, it is unclear how species respond if one type of predator (e.g., a specialized nest predator) is absent but others are present. The multipredator hypothesis suggests that the absence of only a few predators is not sufficient to drive a change or loss of antipredator behavior in a species. We examined interpopulation variation in defense behavior in the thorn‐tailed rayadito (Aphrastura spinicauda), a suboscine bird inhabiting a wide latitudinal range in southern South America. In populations where nest predators were either present or absent, we examined the responses of rayaditos to an adult predator (an owl) and to a nest predator (a marsupial). Owl‐defense behavior involved more intense and longer responses than marsupial‐defense. This suggests that adult rayaditos may allocate more energy to defend themselves than their young. However, because the presence of adult predators near the nest also prevents parents from feeding the nestlings, owl‐defense may simultaneously represent a form of nest‐defense. No differences in marsupial‐defense were found among the three sampled populations, despite the absence of native mammalian nest predators in one population. The lack of variation in nest‐defense behavior provides some support for the multipredator hypothesis, although the high interpopulation gene flow in this species could also contribute to this behavior's maintenance.  相似文献

Erythropoietin (Epo) is crucial for promoting the survival, proliferation, and differentiation of mammalian erythroid progenitors. The central role played by tyrosine phosphorylation of erythropoietin receptor (EpoR) in Epo-cell activation has focused attention on protein tyrosine phosphatases (PTPs) as candidates implicated in the pathogenesis of the resistance to therapy with human recombinant Epo. Prototypic member of the PTP family is PTP1B, which has been implicated in the regulation of EpoR signaling pathways. In previous reports we have shown that PTP1B is reciprocally modulated by Epo in undifferentiated UT-7 cell line. However, no information is available with respect to the modulation of this phosphatase in non-Epo depending cells or at late stages of erythroid differentiation. In order to investigate these issues we induced UT-7 cells to differentiate and studied their PTP1B expression pattern. Simultaneous observations were performed in TF-1 cells which can be cultured either with GM-CSF, IL-3 or Epo. We found that Epo induced PTP1B cleaveage in TF-1 and differentiated UT-7 cells. This pattern of PTP1B modulation may be due to an increased TRPC3/TRPC6 expression ratio which could explain the larger and sustained calcium response to Epo and calpain activation in Epo treated TF-1 and differentiated UT-7 cells.  相似文献

Background

Community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) is now the most common cause of skin and skin structure infections (SSSI) in several world regions. In Argentina prospective, multicenter clinical studies have only been conducted in pediatric populations.

Objective

Primary: describe the prevalence, clinical and demographic characteristics of adult patients with community acquired SSSI due to MRSA; secondary: molecular evaluation of CA-MRSA strains. Patients with MRSA were compared to those without MRSA.

Materials and Methods

Prospective, observational, multicenter, epidemiologic study, with molecular analysis, conducted at 19 sites in Argentina (18 in Buenos Aires) between March 2010 and October 2011. Patients were included if they were ≥14 years, were diagnosed with SSSI, a culture was obtained, and there had no significant healthcare contact identified. A logistic regression model was used to identify factors associated with CA-MRSA. Pulse field types, SCCmec, and PVL status were also determined.

Results

A total of 311 patients were included. CA-MRSA was isolated in 70% (218/311) of patients. Clinical variables independently associated with CA-MRSA were: presence of purulent lesion (OR 3.29; 95%CI 1.67, 6.49) and age <50 years (OR 2.39; 95%CI 1.22, 4.70). The vast majority of CA-MRSA strains causing SSSI carried PVL genes (95%) and were SCCmec type IV. The sequence type CA-MRSA ST30 spa t019 was the predominant clone.

Conclusions

CA-MRSA is now the most common cause of SSSI in our adult patients without healthcare contact. ST30, SCCmec IV, PVL+, spa t019 is the predominant clone in Buenos Aires, Argentina.  相似文献

The traditional method to obtain phycocolloids from seaweeds implies successive extraction steps with cold and hot water. The residual cake derived from phycocolloids obtaining process of red seaweed Porphyra columbina is a waste containing 27 % protein and 10.7-mg gallic acid equivalents (100 g)?1. Seaweeds contain functional proteins, and the enzymatic hydrolysis of these proteins has been shown to release bioactive peptides. The aims of this study were to extract bioactive peptides and polyphenols after enzymatic hydrolysis of the residual cake and to evaluate their ACE inhibitory and antioxidant capacities (TEAC, DPPH, and copper-chelating activity). Residual cake hydrolysate has low molecular weight peptides containing Asp, Glu, Ala, and Leu. Residual cake hydrolysate had higher protein solubility than residual cake. ACE inhibition (≈45 %) and radical scavenging activity (TEAC and DPPH inhibition) were attributed mainly to low molecular weight peptides (500 Da) and polyphenols compounds released during proteolysis. The 50 % inhibition protein concentration value (IC50) corresponded to residual cake hydrolysate was 1.01?±?0.02 and 0.91?±?0.01 g L?1, for ABTS and DPPH, respectively. Also, residual cake hydrolysate had high copper-chelating activity (≈97.5 %). Hydrolysis could be used as a means to obtain ACE inhibitory and antioxidant compounds (peptides and polyphenols) from algae protein waste and add value to the phycocolloids extraction process.  相似文献
