Although simple and low-cost interventions for sickle cell disease (SCD) exist in many developing countries, child mortality associated with SCD remains high, in part, because of the lack of access to diagnostic tests for SCD. A density-based test using aqueous multiphase systems (SCD-AMPS) is a candidate for a low-cost, point-of-care diagnostic for SCD. In this paper, the field evaluation of SCD-AMPS in a large (n = 505) case-control study in Zambia is described. Of the two variations of the SCD-AMPS used, the best system (SCD-AMPS-2) demonstrated a sensitivity of 86% (82–90%) and a specificity of 60% (53–67%). Subsequent analysis identified potential sources of false positives that include clotting, variation between batches of SCD-AMPS, and shipping conditions. Importantly, SCD-AMPS-2 was 84% (62–94%) sensitive in detecting SCD in children between 6 months and 1 year old. In addition to an evaluation of performance, an assessment of end-user operability was done with health workers in rural clinics in Zambia. Genetic variability within and among Bulinus truncatus of the Albertine Rift freshwater bodies were assessed to investigate the degree of inbreeding and gene flow in the snail
populations. The effect of ploidy on the genetic structuring of B. truncatus is also described. We characterized the genetic structure of seven B. truncatus populations from Lake Albert, Lake Kivu, and Katosho swamp in Tanzania using five polymorphic microsatellite loci. Genetic
differentiation was quantified using pairwise FST values and Nei’s standard genetic distances. Different alleles were observed
across all loci and genetic diversity was low although it varied greatly across populations; observed heterozygosity was,
however, higher than the expected heterozygosity in three of the populations studied. Significant heterozygote deficiencies
were observed coupled with significant linkage disequilibria in five populations for all the five loci examined in this study.
We found significant genetic differentiation among the seven freshwater bodies; private alleles were observed across all loci
indicating restricted or absence of gene flow between populations. Limited snail dispersal and the reproductive biology of

Gunnera manicata L. glands consist of up to nine separate papillae. Surgical removal of papillae showed that more than two papillae were needed for successful infection with Nostoc. Lactobacillus pentosus TV35b, isolated from the posterior fornix secretions of the vagina of a prenatal patient, produced a bacteriocin-like peptide (pentocin TV35b), which is inhibitory to Clostridium sporogenes, Cl. tyrobutyricum, Lact. curvatus, Lact. fermentum, Lact. sake, Listeria innocua, Propionibacterium acidipropionici, Propionibacterium sp. and Candida albicans. The mechanism of activity of pentocin TV35b is bactericidal, as shown by a decrease in the viable cell numbers of Lact. sake from approximately 4 x 108 to less than 10 cfu ml - 1 over a period of 4 h. Pentocin TV35b added to the growth medium of C. albicans stimulated the formation of pseudohyphae during the first 36 h, followed by a slight repression in cell growth. Production of pentocin TV35b was at its maximum towards the end of the logarithmic growth phase of strain TV35b. The peptide was purified by ammonium sulphate precipitation, followed by SP-Sepharose cation exchange chromatography. The molecular size of pentocin TV35b was estimated to be between 2.35 and 3.4 kDa, according to tricine-SDS PAGE. However, results obtained by electrospray ionization mass spectroscopy indicated that the peptide is 3930.2 Da in size. Amino acid analysis performed by using the Pico-Tag(R) method and a Nova-Pak C18 HPLC column indicated that pentocin TV35b consists of 33 amino acids with a total mass of 3929.63 Da. Pentocin TV35b is inactivated when treated with papain and Proteinase K, but remains active after incubation at pH 1-10 for 2 h at 25 degrees C, and when heat-treated for 30 min at 100 degrees C. 相似文献

Exogenous pyruvate added to cultures of the bluegreen alga, Anabaena cylindrica stimulated nitrogenase activity (measured by acetylene reduction) only in the dark under low pO2 (0.05 atmospheres). Under aerobic conditions or in the light, stimulation was absent and replaced by an inhibition of activity above 5 mM added pyruvate. The curve of nitrogenase activity versus oxygen concentration had a similar maximal value of ethylene production with or without added pyruvate, but in the presence of pyruvate this maximum occurred at 0.05 atmospheres O2, whilst in the absence of pyruvate the maximum occurred at 0.10 atmospheres O2. Malate, citrate, α-ketoglutarate, glucose and fructose were tested also, but none gave a similar effect to pyruvate. Addition of 14C-pyruvate and autoradiography indicated that exogenous pyruvate is metabolized through the interrupted Krebs cycle. These results are explained in terms of the activity of pyruvate: ferredoxin oxidoreductase and the ATP-induced oxygen sensitivity of nitrogenase. 相似文献

Frankia grown in batch culture was unable to maintain a high rate of nitrogenase activity and, once a peak level was reached, activity rapidly declined. Addition of 5 mM carbon source of cultures or transfer to fresh medium was followed by brief recovery of nitrogenase activity. The extent of recovery decreased as additions or transfers were made to progressively older cultures. Daily addition of fresh medium (dilution rate = 0.125 day-1) allowed Frankia to be maintained in continuous, derepressed culture with stable rates of growth and nitrogenase activity for more than 30 days. Satellite remote sensing offers a cost‐effective method for monitoring fire occurrence in savannah systems, for proper fire management. However, the ability of satellite fire products to detect active fire is known to vary depending on the terrestrial ecosystems and sensor characteristics. In this study, the overall accuracy, kappa coefficient of agreement and true skill statistic (TSS) were used to assess the accuracy of two MODIS fire products (MOD14A1 and MCD14ML) to detect active fire at two savannah woodland sites dominated by Baikiaea plurijuga and Brachystegia spiciformis in Zimbabwe. In both sites, MOD14A1 with a coarse spatial resolution of 1 km had a poor index of agreement with ground fire data (kappa = 0, TSS = 0 and overall accuracy ≤ 0.4). By contrast, a moderate to strong agreement between MCD14ML and active fires measured on the ground was observed at both study sites (overall accuracy ≥ 0.7, kappa ≥ 0.6 and TSS ≥ 0.6; Table 1 ). It was therefore concluded that MCD14ML, with a spatial resolution of 375 m, is a more suitable product for detecting active fires in both Baikiaea plurijuga and Brachystegia‐dominated savannah woodlands of southern Africa compared to MOD14A1. 相似文献

Nitrogenase activity in the Gunnera Nostoc symbiosis is shown to respond dramatically to the addition of glucose. H2 can replace glucose in stimulating nitrogenase activity, but there is no H2 stimulation in the presence of excess glucose. Net hydrogen evolution is strongly stimulated by addition of glucose. We postulate that carbohydrate supply and uptake hydrogenase can moderate the apparent activity of nitrogenase by supplying reductant and/or ATP. The recycling of a large proportion of the electron flux in nitrogenase through uptake hydrogenase maintains a high level of potential nitrogenase ready to take advantage of an influx of carbohydrate. 相似文献
