Although food prices in major world markets are at or near a historical low, there is increasing concern about food security—the ability of the world to provide healthy and environmentally sustainable diets for all its peoples. This article is an introduction to a collection of reviews whose authors were asked to explore the major drivers affecting the food system between now and 2050. A first set of papers explores the main factors affecting the demand for food (population growth, changes in consumption patterns, the effects on the food system of urbanization and the importance of understanding income distributions) with a second examining trends in future food supply (crops, livestock, fisheries and aquaculture, and ‘wild food’). A third set explores exogenous factors affecting the food system (climate change, competition for water, energy and land, and how agriculture depends on and provides ecosystem services), while the final set explores cross-cutting themes (food system economics, food wastage and links with health). Two of the clearest conclusions that emerge from the collected papers are that major advances in sustainable food production and availability can be achieved with the concerted application of current technologies (given sufficient political will), and the importance of investing in research sooner rather than later to enable the food system to cope with both known and unknown challenges in the coming decades.  相似文献

In ongoing efforts to study the ontogeny of gonadotropin-releasing hormone (GnRH) neurons, we serendipitously observed that increasing times of incubation in antibodies enhanced signal detection. Here, we describe significant differences in the early migration pattern, population dynamics, and growth cone morphology from published reports. The first immunoreactive GnRH cells were detected in the mouse at E10.75 (7.6 ± 2.8 cells; morning after mating = E0.5), prior to the closure of the olfactory placode. Although half of these cells were in the medial wall of the olfactory pit, the other half had already initiated their migration, and approximately one quarter had reached the telencephalic vesicle. Although the migratory pattern of the GnRH cells after E11.00 was identical to that described previously, these earliest migrating cells traveled singly rather than in cords, with some reaching the presumptive preoptic area (posterior to the ganglionic eminence) by E11.75. The number of GnRH cells increased significantly (p < 0.05) to 777 ± 183 at E11.75 and peaked at 1949.6 ± 161.6 (p < 0.05) at E12.75. The adult population was approximately 800 cells distributed between the central nervous system (CNS) and the nasal region. Hence, the population of GnRH neurons during early development is much larger than previously appreciated; mechanisms for its decline are discussed. Neuritic extensions on the earliest GnRH neurons are short (30–50 μm) and blunt and may represent the leading edge of the moving cell. By E12.75, GnRH axons in the CNS had a ribboned or beaded morphology and increasingly more complex growth cones were noted from this time until the day of birth. The most complex growth cones were associated with apparent choice points along the axons' trajectory. By E13.75, GnRH axons were seen at the presumptive median eminence in all animals, and it was at this stage that the axons began to branch profusely. Branching, as well as the presence of growth cones, continued postnatally. Glucose aversion(Glu) is a naturally occurring behavioral mutant of the German cockroach,Blattella germanica. Earlier work suggested thatGlu is a semidominant autosomal trait. Further study was undertaken to place the mutant gene in the linkage map of the species and expand the information on its genetic basis. Linkage was investigated in test crosses with morphologic markers for 7 of the 11 autosomal linkage groups. Assays for amounts of glucose ingested per individual confirmed the expected low consumption ofGlu heterozygotes(Glu/+) and that continuous selection pressure on the parental strain had resulted in near- and probably complete homozygosity forGlu. Glu genotypes were identified by means of a discriminating ingestion that separated heterozygotes from wild type(Glu+). Variations in expression indicate that genetic factors influence food consumption inB. germanica. Positive results were obtained in linkage tests ofGlu with ruby eye(ru), indicating that theGlu locus lies in linkage group VIII (chromosome 9), a group notable for mutants that apparently result in the expression of latent, highly primitive development pathways.  相似文献

Membrane vesicles prepared from Halobacterium halobium extrude protons during illumination, and a pH difference (inside alkaline) and an electrical potential (inside negative) develop. The sizes of these gradients and their relative magnitudes are dependent on a complex interaction among the proton-pumping activity of bacteriorhodopsin, Na+ extrusion through an antiport system, and the ability of K+ and Cl? to act as counterions to the electrogenic movement of H+. The net result of these variable effects is that the electrical potential is relatively independent of external pH, whereas the pH difference tends toward zero when the pH is increased to 7.5–8. Although the light-induced pH difference is greater in KCl than in NaCl, and the electrical potential smaller, this is not caused by a high permeability of the vesicle membranes to K+. The vesicle membrane is poorly permeable to K+, as shown by: lack of a K+ diffusion potential in the absence of valinomycin, light-induced electrical potentials which are in excess of the chemical potential difference for K+, and direct measurements of the slow rate of K+ influx during illumination. The finding that the rate of K+ uptake is a linear function of external K+ concentration between 0 and 1 m is inconsistent with the existence of a specific K+ permeation mechanism in these vesicles. Since at external K+ concentrations < 1.4 m the extrusion of Na+ during illumination proceeds much more rapidly than K+ influx, it must be concluded that the vesicles also lose Cl? and water. Measurements of light-scattering changes confirm that under these conditions the vesicles collapse. The light-induced collapse is diminished only when the inward movement of K+ is increased, either by increasing the external K+ concentration or by adding valinomycin.  相似文献

Introduction

The FRAX calculator combines a set of clinical risk factors with country-specific incidence rates to determine the ten-year absolute risk of major osteoporotic fracture. However, regional or country-specific databases from Central American countries are not available. We compared the use of various FRAX databases and the Pluijm algorithm in determining risk of fracture.

Methods

We collected clinical risk factor data needed for the FRAX calculator and Pluijm algorithm of Hispanic women in Guatemala and calculated the FRAX absolute risk measures of major osteoporotic fracture and hip fracture. Subjects were postmenopausal women greater than age 40 with no history of using medication that affect bone. A random sample of 204 women in 34 different regions women in Guatemala City was visited in their homes to complete the surveys. The Pluijm risk score and FRAX risk score using the US Hispanic, Spain, and Mexican databases were calculated.

Results

We used the US NOF guidelines for treatment which suggest a treatment threshold for patients with a 10-year hip fracture probability ≥3% or a 10-year major osteoporotic fracture risk ≥20%. The number of patients meeting the suggested threshold limits for treatment using the Spain and Mexico calculators were identical. There was 100% conformity in threshold limits for both hip and major osteoporotic fracture risk. The mean conformity for any fracture risk between US Hispanic and the other two databases was 97.5%. Conformity was 99.0% based on major osteoporotic fracture and 97.5% based on risk of hip fracture. The Pluijm evaluation shows conformity of 87.2% and 83.3%, respectively, when compared to the US Hispanic and Spain/Mexico FRAX thresholds for risk of fracture.

Discussion

Although the different FRAX databases provide variations in the absolute risk of fracture, the overall conformity to treatment thresholds amongst the US Hispanic, Spain, and Mexico databases show the database used would have little effect as to the decision to treat. The Pluijm tool conforms to the FRAX thresholds and can be used as well. It does not matter which country-specific calculator or assessment tool is used, as there are a similar number of patients that would meet the intervention threshold.  相似文献

5a,6-Anhydrotetracycline was discovered to be unique among several tetracycline derivatives tested in its ability to inhibit RNA accumulation in vivo at low concentration (20 microgram/ml and less). In addition, in vivo protein, DNA, and guanosine 5'-diphosphate 3'-diphosphate (ppGpp) synthesis were completely inhibited by 20 microgram/ml 5a,6-anhydrotetracycline. ppGpp decay in a spoT strain was inhibited by 20 microgram/ml 5a,6-anhydrotef RNA synthesis by a 5a,6-anhydrotetracycline may be due, in part, to reduced UTP and CTP synthesis. The effects of tetracyclines on in vitro ppGpp synthesis by crude stringent factor in the absence of ribosomes were investigated. It was determined that of six tetracyclines tested, four strongly inhibited the reaction (oxytetracycline, chlorotetracycline, dedimethylaminotetracycline, and tetracycline) whereas 5a,6-anhydrotetracycline gave a moderate inhibition and alpha-6-deoxyoxytetracycline resulted in only a slight reduction in ppGpp synthesis. It is proposed that tetracyclines interfere with factors involved in ppGpp metabolism and function.  相似文献

Cultivated peatland (Histosol) in Southern Québec (Canada) is a rapidly declining non-renewable resource used to grow most Canadian lettuce (Lactuca sativa L., Asteraceae). Rolled-rye (Secale cereale L., Poaceae) cover crop is one of the conservation practices proposed to reach a more sustainable lettuce production, but the overall impact on the agroecosystem remains poorly studied in Histosols. We assessed multiple effects of rolled-rye cover crop on the trophic chain associated with the lettuce aphid, Nasonovia ribisnigri (Mosley) (Hemiptera: Aphididae), a major pest of lettuce. During one growing season and through two consecutive lettuce crops, we monitored in situ the impacts of rolled-rye cover crop on insect fauna and lettuce quality. We used visual scouting and yellow pan traps to assess plant colonization by N. ribisnigri, its natural enemies, and alternative prey. Exclusion cage experiments were also conducted to measure aphid fitness and population growth. Under greenhouse conditions, following cover crop removal, we examined potential lingering effects of rye within the soil on lettuce plants and N. ribisnigri. In situ experiments showed that rolled-rye cover crop has the potential to inhibit N. ribisnigri field colonization and recruit natural enemies and alternative prey in the first lettuce crop. Rye also reduced aphid fitness as well as lettuce foliar amino acid concentration and weight. For the second lettuce crop, an almost 50% reduction in N. ribisnigri abundance was observed with the use of rolled-rye cover crop. In greenhouse experiments, no persistent effect of rye was observed on the quality of lettuce grown with soil collected under a cover crop, nor on the fitness of aphids inoculated on these lettuces. This study highlights the diversity of trophic effects rolled-rye cover crop may have on lettuce production in Histosols and the potential of rolled-rye cover crop as a cultural practice to reduce aphid populations. Mechanisms at play while underlining agronomic challenges regarding proper rye termination must be further explored to maintain high-performing lettuce yields.  相似文献

Sweet basil (Ocimum basilicum) is an annual herb crop grown in polyethylene‐covered structures in Israel. It is Israel's leading herb crop, grown in warm regions of the country. Downy mildew (caused by Peronospora belbahrii) is a severe disease in Israel and in many other crop‐growing regions worldwide. Experiments were carried out to identify potential climate‐management techniques for suppression of this disease on basil in non‐heated greenhouses. Disease severity was evaluated under commercial‐like conditions in three experiments, with 8–10 walk‐in tunnels at each location. Pathogen inoculum was introduced into all walk‐in tunnels. Regression analysis was performed between the disease values and air temperature, relative humidity (RH) and soil temperature. Downy mildew severity was negatively related to high (>25°C) air temperature, RH in the range of 65–85% and high (>21°C) soil temperature. The increase in air temperature did not result in a significant increase in leaf temperature; canopy surface median temperatures only reached 30°C. Symptomless plants from relatively warmer tunnels (peak temperatures of 45–48°C) that were transferred to conditions that promote downy mildew (22 ± 2°C, RH > 95%) became severely diseased, showing sporulation of P. belbahrii, suggesting that infection occurred but at the high temperatures symptom expression/tissue colonisation was suppressed. Pot experiments in which aerial and subterranean plant organs were differentially heated revealed that treating the roots with a high temperature (26–31°C), similar to the soil temperatures in the warmer greenhouses, while maintaining the upper plant parts at ambient temperature (20°C), suppresses canopy downy mildew. The effect lasted for 1–2 weeks after the plants were removed from the heated soil treatments and maintained under optimal conditions for pathogen development. Furthermore, oospores were found in the symptomatic leaves. Oospores are minimally affected by high temperature, and therefore the high temperature presumably did not affect pathogen survival. In conclusion, the effect of high greenhouse temperature on basil downy mildew may not result from a direct negative effect of high temperature on the pathogen but from an indirect high‐temperature effect on the host, rendering it less susceptible to pathogen development.  相似文献

Genetic polymorphisms in DNA repair genes may impact individual variation in DNA repair capacity and alter cancer risk. In order to examine the association of common genetic variation in the base-excision repair (BER) pathway with bladder cancer risk, we analyzed 43 single nucleotide polymorphisms (SNPs) in 12 BER genes (OGG1, MUTYH, APEX1, PARP1, PARP3, PARP4, XRCC1, POLB, POLD1, PCNA, LIG1, and LIG3). Using genotype data from 1,150 cases of urinary bladder transitional cell carcinomas and 1,149 controls from the Spanish Bladder Cancer Study we estimated odds ratios (ORs) and 95% confidence intervals (CIs) adjusting for age, gender, region and smoking status. SNPs in three genes showed significant associations with bladder cancer risk: the 8-oxoG DNA glycosylase gene (OGG1), the Poly (ADP-ribose) polymerase family member 1 (PARP1) and the major gap filling polymerase-β (POLB). Subjects who were heterozygous or homozygous variant for an OGG1 SNP in the promoter region (rs125701) had significantly decreased bladder cancer risk compared to common homozygous: OR (95%CI) 0.78 (0.63–0.96). Heterozygous or homozygous individuals for the functional SNP PARP1 rs1136410 (V762A) or for the intronic SNP POLB rs3136717 were at increased risk compared to those homozygous for the common alleles: 1.24 (1.02–1.51) and 1.30 (1.04–1.62), respectively. In summary, data from this large case-control study suggested bladder cancer risk associations with selected BER SNPs, which need to be confirmed in other study populations. The middle medullary membrane of canine renal tissue was completely insensitive to parathyroid hormone (PTH) stimulation in the absence of GTP or tissue supernatant. In contrast, removal of endogenous GTP did not eliminate the PTH stimulation of cAMP formation in the renal cortical membrane preparation (P3). Addition of boiled cortical P3 to native cortical P3 enhanced PTH stimulation in a dose-dependent manner. The boiled cortical P3 was not active in middle medullary tissue. The minimum concentration of GTP which was required to cause stimulation in both cortical and middle medullary preparations was similar. The results suggest that (1) there are two classes of PTH-sensitive adenylate cyclase; one class is GTP dependent, while the other is GTP independent. (2) A new factor, as yet unidentified, in addition to GTP, is important in the regulation of adenylate cyclase by PTH in the renal cortex.  相似文献
