In two previous quantitative trait locus (QTL) mapping studies conducted inLycopersicon esculentum x L. pimpinellifolium BC1 and BC2 populations we had localized a major QTL for fruit shape,fs8.1, to a ca. 20 cM interval on the short arm of chromosome 8, flanked by markers TG176 and CT92. At this QTL the allele from the wild species reduces the length of fruit, giving round-shaped fruit. In order to define more precisely the location offs8.1, near-isogenic lines (NILs) segregating for the region of interest were developed. The results from substitution mapping show that no recombination occurred betweenfs8.1 and the marker CD40 in 322 meioses. The gene action forfs8.1 was determined in a BC4F3 population to be partial dominance. The main effect offs8.1 is exerted on fruit length while fruit diameter is not significantly affected. A highly significant correlation (r=0.89;P<0.01) was found between fruit shape and ovary shape indicating that thefs8.1 gene product acts early in ovary development (preanthesis). Implications for the evolution of fruit shape and the feasibility of map-based cloning of this QTL are discussed.  相似文献

We investigate the effect of EGF on IP3 production, PLCγ phosphorylation, calcium transients in rat hepatocytes isolated in quiescent liver (G0 phase of cell cycle) and at 4 h (G1 phase of cell cycle) and 24 h (M phase of cell cycle) after partial hepatectomy. Our results show that EGF does not utilize IP3 and calcium as its signal transduction molecules when the hepatocytes are in vivo stimulated to entry in the cell cycle. In particular the growth factor does not phosphorylate PLCγ and induces a decrease in IP3 content. These data suggest that EGF utilizes different signal transduction to send information from receptor to nucleus during PH with respect to the quiescent liver.  相似文献

In order to estimate spatial distribution, temporal variation, and prevalence of Andes hantavirus antibody in the rodent community, and especially in Oligoryzomys longicaudatus populations, four different ecosystems were trapped seasonally between spring 2001 and winter 2002 in Neuquen, northwestern Argentinean Patagonia. Five peridomestic settings were sampled within the same period. The rodent O. longicaudatus had the widest distribution in Neuquen, as it was the only species captured at every sample site except for the High Andean steppe, and it was also the most common species captured. Rodents of 13 species were tested for hantavirus antibody prevalence, but O. longicaudatus and Abrothrix longipilis were the only seropositive species. Seropositive individuals were captured during spring and summer in the Subantarctic forest and in winter 2001 in a peridomestic setting in the Patagonian steppe. The dominant presence of O. longicaudatus throughout Neuquen must be incorporated into strategies to prevent human exposure to hantavirus.  相似文献

Summary A cytogenetic analysis of a lung metastasis of an adamantinoma of the tibia, a rare tumor of the long bones, revealed a karyotype 52, XY, t(7;13)(q32;q14), +7, +12, +13, +19, +der(7) t(7;13), +der(13) t(7;13). The t(7;13) was found to be constitutional in the patient and was also present in peripheral blood lymphocytes of his unaffected father. However, both subjects displayed normal levels of esterase D enzyme activity.  相似文献

Fluorescence analysis after quinacrine staining in squashes of Varese wild stock male larval ganglia confirmed that the Y chromosome has four characteristic sections of bright fluorescence. In one Y/X and in one Y/III translocation the section of bright fluorescence on the short arm of the Y is no longer bright when translocated onto the terminal portion of the X and on the right arm of the III chromosome, respectively. Fluorescence analysis has also permitted the identification of a structurally abnormal Y chromosome in a cell line of Drosophila melanogaster established in vitro. The findings in the two translocations call for caution in the interpretation of structural rearrangements by fluorescence analysis.  相似文献

Larval neuroblasts of four species of Drosophila (melanogaster, hydei, virilis and funebris) were treated with distamycin A, a DNA ligand which induces distinct undercondensation in AT-rich heterochromatin. For each species the patterns of undercondensation were correlated with distribution of quinacrine-bright regions and of satellite DNAs. An overlapping of distamycin A-sensitive and quinacrine-bright heterochromatic regions was demonstrated for D. melanogaster and D. virilis, but not for D. hydei and D. funebris. Distamycin A undercondensation is thus a further criterion for resolving heterochromatin into different parts and enables identification of the steps of the condensation process within heterochromatic regions.  相似文献

The technique of intracerebral dialysis in combination with a sensitive and specific radioenzymatic method was used for recovery and quantification of endogenous extracellular acetylcholine from the striata of freely moving rats. A thin dialysis tube was inserted transversally through the caudate nuclei, and the tube was perfused with Ringer solution, pH 6.1, at a constant rate of 2 microliter min-1. The perfusates were collected at 10-min intervals. In the presence of 1 and 10 microM physostigmine, acetylcholine release was 4.5 +/- 0.02 and 7.3 +/- 0.3 pmol/10 min, respectively (not corrected for recovery). The latter concentration of the acetylcholinesterase inhibitor was used in all experiments. Under basal conditions, acetylcholine output was stable over at least 4 h. A depolarizing K+ concentration produced a sharp, reversible 87% increase in acetylcholine output. Both the basal and K+-stimulated release were Ca2+ dependent. The choline uptake inhibitor hemicholinium-3 (20 micrograms intracerebroventricularly) reduced striatal acetylcholine output to 35% of the basal value within 90 min. Scopolamine (0.34 mg/kg s.c.) provoked a sharp enhancement of acetylcholine release of approximately 63% over basal values, whereas oxotremorine (0.53 mg/kg i.p.) transiently reduced acetylcholine release by 54%. These results indicate the physiological and pharmacological suitability of transstriatal dialysis for monitoring endogenous acetylcholine release.  相似文献

Summary Cytogenetic investigations for diagnostic purposes were performed on 1000 first trimester samples of chorionic villi (CVS) in two laboratories using similar techniques. Fetal karyotyping was the primary indication for CVS in 912 and maternal age was the major indication in 758 of them. The risk category previous child/fetus with chromosome abnormality included 74 diagnoses, while the category chromosome abnormality in one of the parents included 38 diagnoses. Sex determination was the primary indication for CVS in 53 pregnancies. The overall incidence of chromosomal abnormalities was 70, of which 47 were balanced and 23 unbalanced. The results are detailed for each of the risk categories and the incidence of abnormal karyotypes is given for each year of maternal age. In the maternal age of 35–37 years the incidence of unbalanced karyotypes was 2.9% and in the years 38 onwards it was 6.6%. The incidence of unbalanced karyotypes was about 4% when the sampling was made in the weeks 9 to 12 but six abnormal karyotypes were found among 39 CVS performed at the eight week of gestation. The 11 trisomies of the type not found at birth were clustered between the 8th and the 10th week of pregnancy. The technical problems encountered in this experience and the preliminary estimates of fetal loss are discussed.  相似文献

Three continuous cell lines (GM1, GM2 and GM3) were obtained from embryos of Drosophila melanogaster. Karyotypic analysis revealed characteristics distinguishing each line. Except for some minor variations GM1 cells had an X and a centric heterochromatic fragment (which is a portion of the Y). GM2 line was characterized by XO cells showing two new telocentric chromosomes while an autosome of the II pair was missing. GM3 cells were XY; the Y chromosome, however, was shorter than the normal, having a deletion of the terminal section of the short arm.
