Studies demonstrate that GRM, expressed by >60% of human melanomas, may be a therapeutic target. We performed a phase II trial of 100 mg PO bid of riluzole, an inhibitor of GRM1 signaling, in patients with advanced melanoma with the primary endpoint of response rate. Thirteen patients with GRM1‐positive tumors were enrolled. No objective responses were observed, and accrual was stopped. Stable disease was noted in six (46%) patients, with one patient on study for 42 weeks. Riluzole was well tolerated, with fatigue (62%) as the most common adverse event. Downregulation of MAPK and PI3K/AKT was noted in 33% of paired tumor biopsies. Hypothesis‐generating correlative studies suggested that downregulation of angiogenic markers and increased leukocytes at the active edge of tumor correlate with clinical benefit. Pharmacokinetic analysis showed interpatient variability consistent with prior riluzole studies. Future investigations should interrogate mechanisms of biologic activity and advance the development of agents with improved bioavailability.  相似文献

From a single aflatoxin B1 oxime — bovine serum albumin conjugate, polyclonal and monoclonal antibody preparations were produced. The four rabbit polyclonal antisera were specific for aflatoxin Bi in a microtitration plate enzyme — linked immunosorbent assay. The monoclonal antibodies showed a wide range of differing specificities, recognizing, for example, aflatoxins B1, B2, G1 and G2; B1 and B2; B1 and G1; and G1 alone. No antibody preparations reacted with aflatoxin M1. The significance of these results to the strategy of anti-aflatoxin antibody production for use in quantitative enzyme immunoassays is discussed.  相似文献

Excised lettuce (Lactuca sativa L.) hypocotyl sections retain the ability to elongate in response to gibberellic acid (GA3) addition. In 48 hr at 30 C a GA3-treated segment more than doubles while a control segment elongates less than 50%. Auxin has no detectable effect on this system. Sensitivity to GA3 is not decreased by apex or root removal. Of the experimental variables tested, temperature, sucrose, and preincubation in water affect growth both with and without GA3. Blue and far red light inhibit growth without GA3; this inhibition is reversed by GA3. Potassium chloride stimulates growth of illuminated sections treated with GA3 but has no effect on control growth. When sections are incubated in the dark, KCl has a promotive effect on elongation.  相似文献

Densities of osmoticum and potassium were measured as a function of distance from the tip of the primary root of Zea mays L. (cv WF9 × mo17). Millimeter segments were excised and analyzed for osmotic potential by a miniaturized freezing point depression technique, and for potassium by flame spectrophotometry. Local deposition rates were estimated from the continuity equation with values for density and growth velocity. Osmotic potential was uniform, −0.73 ± 0.05 megapascals, throughout the growth zone of well-watered roots. Osmoticum deposition rate was 260 μosmoles per gram fresh weight per hour. Potassium density fell from 117 micromoles per gram in the first mm region to 48 micromoles per gram at the base of the growth zone. Potassium deposition rates had a maximum of 29 micromoles per gram per hour at 3.5 millimeters from the tip and were positive (i.e. potassium was being added to the tissue) until 8 millimeters from the tip. The results are discussed in terms of ion relations of the growing zone and growth physics.  相似文献

An equation is derived from transport theory to relate local growth rate to local water potential in an expanding tissue. For a noncompartmented continuum model, the relative elemental growth rate (L) equals the divergence of the tensor product of hydraulic conductivity () and the gradient of water potential, ψ, i.e. L = • [ · ψ]. This equation is solved numerically using published values of L and to show the water potential distribution which can sustain the observed growth pattern in the primary root of Zea mays L. The water potential required to sustain growth decreases from the outside to the inside of the root, and the longitudinal profile shows most negative values near the location of the highest growth rate. A cell originally located near the apex experiences a loss and then a gain in water potential as it is displaced through the growth zone.  相似文献

G protein regulation of human platelet membrane phospholipase A2 activity was investigated at pH 8.0 and 9.0 by studying the effects of the nonhydrolyzable GTP analogue, guanosine 5'-O-(3-thiotriphosphate) (GTP gamma S), and of F-/Al3+ ions on arachidonic acid (AA) release. The membrane acted as the source of the enzyme, the substrate, and the G protein. At pH 8.0, 10 and 100 microM GTP gamma S stimulated AA mobilization at least 6-fold. Optimum AA release conditions required 1 mM Ca2+ and 5 mM Mg2+. Nonspecific nucleotide effect was excluded since similar stimulatory effects on AA release were not observed by ATP, GTP, ADP, and NADP. Although at pH 9.0 the GTP gamma S-stimulated AA release was greater than at pH 8.0, it constituted only 26% of the total. At both pH values the effect of F- (10 mM) in the presence of Al3+ (2 microM) was similar to that of GTP gamma S. The G protein inhibitor, guanosine 5'-O-(2-thiodiphosphate), inhibited the GTP gamma S-stimulated AA release by about 80% at pH 8.0 and by 100% at pH 9.0. To determine a possible contribution to AA mobilization by the phospholipase C and diacylglycerol lipase pathway, the effects of neomycin, a phospholipase C inhibitor, were investigated. 100 microM neomycin did not inhibit the GTP gamma S-stimulated AA release at pH 8.0 and only slightly so (17%) at pH 9.0. At pH 8.0 in the presence of Ca2+ the released fatty acids consisted mainly of arachidonic and docosahexaenoic acids (80 and 8%, respectively). GTP gamma S had no effect on the fatty acid profile but only on their quantity. These results provide evidence of G protein regulation of phospholipase A2 activity in isolated platelet membranes.  相似文献

Trivers and Willard predicted that when parental condition has differential effects on the fitness of male and female offspring, parents who are in good condition will bias investment toward the sex that benefits most from additional investment. Efforts to test predictions derived from Trivers and Willard''s model have had mixed results, perhaps because most studies have relied on proxy measures of parental condition, such as dominance rank. Here, we examine the effects of female baboons condition on birth sex ratios and post-natal investment, based on visual assessments of maternal body condition. We find that local environmental conditions have significant effects on female condition, but maternal condition at conception has no consistent relationship with birth sex ratios. Mothers who are in poorer condition at the time of conception resume cycling significantly later than females who are in better condition, but the sex of their infants has no effect on the time to resumption of cycling. Thus, our findings provide strong evidence that maternal condition influences females'' ability to reproduce, but females do not facultatively adjust the sex ratio of their offspring in relation to their dominance rank or current condition.  相似文献

The purpose of the present paper is to evaluate the factors that contributed to annual variation in fertility and infant survival in a relatively undisturbed captive group of bonnet macaques (Macaca radiata) over a 16-year period. The size and composition of this population fluctuated over time, and these changes were consistently associated with changes in female fertility and infant survival. Female fertility was highest when there were relatively few adult females in the group and when there were relatively few adult females per adult male. Similarly, infant survival was highest in years when there were relatively few adult females present and when cohorts of infants were small. Since environmental factors, such as availability of food and vulnerability to predation, were unlikely to constrain population growth in captivity, the data suggest that other mechanisms may have affected demographic processes in this captive group.  相似文献

Rank, parity, and kinship influence the pattern and frequency of kidnapping interactions in a group of captive bonnet macaques,Macaca radiata. Kidnapping attempts are initiated by parous and nulliparous females toward the infants of lower ranking females. Contrary to the pattern documented in several other species, nulliparous females participate in kidnapping interactions at the same rate as lactating females. Kidnapping attempts are consistently resisted by mothers, and most attempts are unsuccessful. Both the rates and pattern of kidnapping interactions differ within and between lineages. The results suggest that kidnappers gain neither status nor maternal experience and that the mothers of kidnapped infants do not benefit at all. Kidnapping may represent a form of competition among females which is detrimental to both infants and their mothers.  相似文献
