Mutagenicity of pan residues and gravy from fried meat   总被引:1,自引:0,他引:1  
Lean pork meat was fried with or without the addition of frying-fat at 200 or 250 degrees C. The pan residues were collected by washing the hot pan with boiling water. When producing thickened gravy the water was substituted by a mixture of water and flour, milk and flour or cream and flour. The basic extracts were tested for mutagenicity in Ames' Salmonella test on strain TA98 with the addition of S9 mix. High amounts of mutagenicity were found in all samples. The amounts of mutagenicity in the pan residues were at a comparable level of the amounts found in the meat crusts. Thickening of the gravy caused only small changes in the mutagenicity.  相似文献

The total fatty acids in human endothelial cells include approximately 5% each of 22:4(n-6), 22:5(n-3) and 22:6(n-3), whereas 22:5(n-6) is present only in trace amounts. This study evaluates the effect of three of these fatty acids bound to albumin on lipid composition and prostacyclin (prostaglandin I2) synthesis in primary cultures of endothelial cell monolayers. 22:4(n-6), 22:5(n-6) and 22:6(n-3) were all incorporated into total phospholipids. 20:4(n-6) was reduced in phospholipids in all cells incubated with the three different docosaenoic fatty acids. This reduction was abolished when equimolar concentrations of 20:4(n-6) and the separate docosaenoic fatty acid were added to the medium simultaneously. 22:4(n-6) incorporation into the free fatty acids was associated with an increase of 20:4(n-6) in this fraction. 22:4(n-6), 22:5(n-6) and 22:5(n-3) all reduced the synthesis of prostacyclin measured as 6-ketoprostaglandin F1 alpha. These effects were reversed by simultaneous incubation with 20:4(n-6). This study shows that three of the docosaenoic fatty acids present in human endothelial cells of the (n-6) and (n-3) family were all incorporated into endothelial cells with a simultaneous reduction in 20:4(n-6). The three fatty acids reduced the synthesis of prostacyclin.  相似文献

Microalgae, with their high lipid content, are a promising feedstock for renewable fuels. Traditionally, human and environmentally toxic solvents have been used to extract these lipids, diminishing the sustainability of this process. Herein, pulsed electric field technology was utilized as a process intensification strategy to enhance lipid extraction from Ankistrodesmus falcatus wet biomass using the green solvent, ethyl acetate. The extraction efficiency for ethyl acetate without PEF was lower (83–88%) than chloroform. In addition, the ethyl acetate exhibited a 2‐h induction period, while the chloroform showed no time dependence. Utilizing PEF technology resulted in 90% of the cells being lysed and a significant enhancement in the rate of lipid recovery using ethyl acetate. The increase in lipid recovery was due to the presence of the electric field and not due to temperature effects. The PEF technology uses less energy than other PEF systems reported in the literature. Abstract

Efficient cellular uptake is crucial for the success of any drug directed towards targets inside cells. Peptide nucleic acid (PNA), a DNA analog with a promising potential as a gene-directed drug, has been shown to display slow membrane penetration in cell cultures. We here used liposomes as an in vitro model of cell membranes to investigate the effect on penetration of a PNA molecule colvalently modified with a lipophilic group, an adamantyl moiety. The adamantyl attachment was found to increase the membrane-penetration rate of PNA three-fold, as compared to corresponding unmodified PNA. From the penetration behaviour of a number of small and large molecules we could conclude that passive diffusion is the mechanism for liposome-membrane passage. Flow linear dichroism (LD) of the modified PNA in presence of rod-shaped micelles, together with octanol-water distribution experiments, showed that the adamantyl-modified PNA is amphiphilic; the driving force behind the observed increased membrane-penetration rate appears to be an accumulation of the PNA in the lipid double layer.  相似文献

Newborn hooded seals (Cystophora cristata) have smaller weight-specific oxygen stores than adults, but nothing is known about how this affects their diving behaviour. Here, we present data on the diving behaviour and migrations of seven weaned hooded seal pups of the Greenland Sea stock during their first year of life, as collected by use of satellite telemetry. The pups started diving 1–2 days after tagging, and during a tracking period of 25–398 days they dispersed over vast areas of the Greenland and Norwegian Seas in a manner similar to adults. The initial development of diving depths and durations in April–May was rapid, and pups reached depths of >100 m and dived for >15 min within 3 weeks of age. During early summer (May–June) this development was temporarily discontinued, to be resumed throughout autumn and winter, during which time maximum depths and durations of >700 m and >30 min, respectively, were reached. Depths and durations were significantly related to age/season, location and time of day. The dive behaviour in early summer, with relatively shallow and short dives without diurnal variations, resembles that of adults and probably reflects the vertical distribution of prey rather than physiological constraints. Dives of pups were nevertheless shallower and shorter than those of adults, but relative to body mass both hooded seal pups and adults display a remarkable diving capacity which makes the species particularly suited for studies of defence mechanisms against hypoxia insult in mammals.  相似文献

The base excision repair (BER) pathway is mainly responsible for the repair of a vast number of non-bulky lesions produced by alkylation, oxidation or deamination of bases. DNA glycosylases are the key enzymes that recognize damaged bases and initiate BER by catalyzing the cleavage of the N-glycosylic bond between the base and the sugar. Many of the mammalian DNA glycosylases have been identified by a combination of biochemical and bioinformatics analysis. Thus, a mammalian family of three proteins (NEIL1, NEIL2 and NEIL3) that showed homology to the Escherichia coli Fpg/Nei DNA glycosylases was identified. Two of the proteins, NEIL1 and NEIL2 have been thoroughly characterized and shown to initiate BER of a diverse number of oxidized lesions. However, much less is known about NEIL3. The biochemical properties of NEIL3 have not been elucidated. This is mainly due to the difficulty in the expression and purification of NEIL3. Here, we describe the expression and partial purification of full-length human NEIL3 and the expression, purification and characterization of a truncated human core-NEIL3 (amino acids 1–301) that contains the complete E. coli Fpg/Nei-like domain but lacks the C-terminal region.  相似文献

Lebanon hosts the highest per capita refugee concentration worldwide. The Palestinian presence in Lebanon dates from 1948 and they remain a marginalized population. No information on their food security status has been reported previously. A survey of a representative sample of Palestinian refugee households in Lebanon (n = 2501) was conducted using a stratified two stage cluster sampling approach. We measured food insecurity using a modified USDA household food security module, locally validated. We collected data on household demographic, socioeconomic, health, housing, coping strategies and household intake of food groups and analysed these by food security status. About 41% (CI: 39-43) of households reported being food insecure and 20% (CI: 18-22) severely food insecure. Poor households were more likely to be severely food insecure (OR 1.41 (1.06-1.86)) while higher education of the head of household was significantly associated with protection against severe food insecurity (OR 0.66 (0.52-0.84)). Additionally, higher food expenditure and possession of food-related assets were significantly associated with food security (OR 0.93 (0.89-0.97) and OR 0.74 (0.59-0.92), respectively). After adjusting for confounders, households where at least one member suffered from an acute illness remained significantly more likely to be severely food insecure (OR 1.31(1.02-1.66)), as were households whose proxy respondent reported poor mental health (OR 2.64 (2.07-3.38)) and poor self-reported health (OR 1.62 (1.22-2.13). Severely food insecure households were more likely to eat cheaper foods when compared to non-severely food insecure households (p<0.001) and were more likely to rely on gifts (p<0.001) or welfare (p<0.001). They were also more likely to have exhausted all coping strategies, indicating significantly more frequently that they could not do anything (p = 0.0102). Food insecurity is a significant problem among Palestinian refugees in Lebanon and is likely to be exacerbated at this time when the Syrian crisis amplifies the problem.  相似文献

The purpose of this study was to answer the question of whether dehydrated harp seals (Phoca groenlandica) are able to obtain a net gain of water from the intake of seawater. Following 24 h of fasting, three subadult female harp seals were dehydrated by intravenous administration of the osmotic diuretic, mannitol. After another 24 h of fasting, the seals were given 1,000 ml seawater via a stomach tube. Urine and blood were collected for measurement of osmolality and osmolytes, while total body water (TBW) was determined by injections of tritiated water. In all seals, the maximum urinary concentrations of Na+ and Cl were higher than in seawater, reaching 540 and 620 mM, respectively, compared to 444 and 535 mM in seawater. In another experiment, the seals were given ad lib access to seawater for 48 h after mannitol-induced hyper-osmotic dehydration. In animals without access to seawater, the mean blood osmolality increased from 331 to 363 mOsm kg−1 during dehydration. In contrast, the blood osmolality, hematocrit and TBW returned to normal when the seals were permitted ad lib access to seawater after dehydration. In conclusion, this study shows that harp seals have the capacity to gain net water from mariposa (voluntarily drinking seawater) and are able to restore water balance after profound dehydration by drinking seawater.  相似文献

Inclusion of spatially explicit information on ecosystem services in conservation planning is a fairly new practice. This study analyses how the incorporation of ecosystem services as conservation features can affect conservation of forest biodiversity and how different opportunity cost constraints can change spatial priorities for conservation. We created spatially explicit cost-effective conservation scenarios for 59 forest biodiversity features and five ecosystem services in the county of Telemark (Norway) with the help of the heuristic optimisation planning software, Marxan with Zones. We combined a mix of conservation instruments where forestry is either completely (non-use zone) or partially restricted (partial use zone). Opportunity costs were measured in terms of foregone timber harvest, an important provisioning service in Telemark. Including a number of ecosystem services shifted priority conservation sites compared to a case where only biodiversity was considered, and increased the area of both the partial (+36.2%) and the non-use zone (+3.2%). Furthermore, opportunity costs increased (+6.6%), which suggests that ecosystem services may not be a side-benefit of biodiversity conservation in this area. Opportunity cost levels were systematically changed to analyse their effect on spatial conservation priorities. Conservation of biodiversity and ecosystem services trades off against timber harvest. Currently designated nature reserves and landscape protection areas achieve a very low proportion (9.1%) of the conservation targets we set in our scenario, which illustrates the high importance given to timber production at present. A trade-off curve indicated that large marginal increases in conservation target achievement are possible when the budget for conservation is increased. Forty percent of the maximum hypothetical opportunity costs would yield an average conservation target achievement of 79%.  相似文献
