The association of temporal lobe epilepsy with depression and other neuropsychiatric disorders has been known since the early beginnings of neurology and psychiatry. However, only recently have in vivo and ex vivo techniques such as Positron Emission Tomography, Magnetic Resonance Imaging and Magnetic Resonance Spectroscopy in combination with refined animal models and behavioral tests made it possible to identify an emerging pattern of common pathophysiological mechanisms. We now have growing evidence that in both disorders altered interaction of serotonergic and noradrenergic neurons with glutamatergic systems is associated with abnormal neuronal circuits and hyperexcitability. Neuronal hyperexcitability can possibly evoke seizure activity as well as disturbed emotions. Moreover, decreased synaptic levels of neurotransmitters and high glucocorticoid levels influence intracellular signaling pathways such as cAMP, causing disturbances of brain-derived and other neurotrophic factors. These may be associated with hippocampal atrophy seen on Magnetic Resonance Imaging and memory impairment as well as altered fear processing and transient hypertrophy of the amygdala. Positron Emission Tomography studies additionally suggest hypometabolism of glucose in temporal and frontal lobes. Last, but not least, in temporal lobe epilepsy and depression astrocytes play a role that reaches far beyond their involvement in hippocampal sclerosis and ultimately, therapeutic regulation of glial-neuronal interactions may be a target for future research. All these mechanisms are strongly intertwined and probably bidirectional such that the structural and functional alterations from one disease increase the risk for developing the other. This review provides an integrative update of the most relevant experimental and clinical data on temporal lobe epilepsy and its association with depression.  相似文献

The aim of this study was to investigate the use of items intended to provide enrichment during turnout, both for individual and group kept horses in an attempt to reduce the amount of passive behaviours. The study was divided into two parts, where study 1 involved eight horses rotated through eight individual paddocks, each containing one of seven enrichment items and one paddock being kept without item, functioning as a control. The horses’ item-directed behaviours; passive behaviours or other non-item related activities were scored using instantaneous sampling, every minute for 1 h at the beginning and the end of the turnout period. Study 2 involved six horse groups (3–6 horses) and the same scoring methods and ethogram as in study 1. The four items that the horses interacted the most with during study 1 (straw STRA, ball filled with concentrates CBALL, branches BRAN and scratching pole POLE) are investigated in study 2. In addition, the amount of social interactions was recorded.Both horses kept individually (P < 0.05) and in groups (P < 0.0001) performed significantly more item-directed behaviours towards edible items like STRA and CBALL than other objects. There was, however, no overall relation between the numbers of item-directed behaviours and the number of passive behaviours observed, indicating that the enrichment items did not alone reduce the amount of passive behaviours during turnout periods. Such a reduction was, however, only apparent when horses spent more time eating green leaves growing on the paddock surface (R = ?0.97 study 1, R = ?0.67 study 2, P < 0.0001). Access to STRA in group kept horses also seemed to reduce the amount of agonistic behaviours (P < 0.0001). In conclusion, if grass is not available in paddocks, the provision of roughage reduces the amount of passive behaviours in singly kept horses and it also reduces the risk of agonistic interactions between horses kept in group.  相似文献

The micro-flora of the proximal jejunum in healthy volunteers was compared with the micro-flora in patients with gastrointestinal symptoms suggestive of spontaneous bacterial overgrowth in the small intestine. Biopsies were taken distally to the ligament of Treitz with a Watson capsule. The samples were diluted and inoculated on selective and non-selective agar plates that were incubated aerobically and anaerobically. No major differences were found in the small jejunum micro-flora in healthy persons or in a heterogenous group of patients with gastrointestinal disorders. Oropharyngeal micro-organisms dominated the micro-flora in all subjects and colonic micro-organisms were found in low numbers in a few subjects from both groups. Streptococcus intermedius and Haemophilus parahaemolyticus were only found in the micro-flora of healthy subjects while Lactobacillus spp. was more frequently found in the samples from patients. Eight of 20 healthy subjects and five of 18 patients met the criterion of small intestinal overgrowth. Emerging evidence suggests that other factors are involved in the pathogenesis of the irritable bowel syndrome complex. There is a need for better understanding of the complicated interactions between the host and the endogenous micro-flora.  相似文献

Glycosylation of viral envelope proteins is important for infectivity and interaction with host immunity, however, our current knowledge of the functions of glycosylation is largely limited to N-glycosylation because it is difficult to predict and identify site-specific O-glycosylation. Here, we present a novel proteome-wide discovery strategy for O-glycosylation sites on viral envelope proteins using herpes simplex virus type 1 (HSV-1) as a model. We identified 74 O-linked glycosylation sites on 8 out of the 12 HSV-1 envelope proteins. Two of the identified glycosites found in glycoprotein B were previously implicated in virus attachment to immune cells. We show that HSV-1 infection distorts the secretory pathway and that infected cells accumulate glycoproteins with truncated O-glycans, nonetheless retaining the ability to elongate most of the surface glycans. With the use of precise gene editing, we further demonstrate that elongated O-glycans are essential for HSV-1 in human HaCaT keratinocytes, where HSV-1 produced markedly lower viral titers in HaCaT with abrogated O-glycans compared to the isogenic counterpart with normal O-glycans. The roles of O-linked glycosylation for viral entry, formation, secretion, and immune recognition are poorly understood, and the O-glycoproteomics strategy presented here now opens for unbiased discovery on all enveloped viruses.  相似文献

Suboptimal nitrogen (N) availability is a primary constraint for crop production in developing nations, while in rich nations, intensive N fertilization carries substantial environmental and economic costs. Therefore, understanding root phenes that enhance N acquisition is of considerable importance. Structural-functional modeling predicts that root cortical aerenchyma (RCA) could improve N acquisition in maize (Zea mays). We evaluated the utility of RCA for N acquisition by physiological comparison of maize recombinant inbred lines contrasting in RCA grown under suboptimal and adequate N availability in greenhouse mesocosms and in the field in the United States and South Africa. N stress increased RCA formation by 200% in mesocosms and by 90% to 100% in the field. RCA formation substantially reduced root respiration and root N content. Under low-N conditions, RCA formation increased rooting depth by 15% to 31%, increased leaf N content by 28% to 81%, increased leaf chlorophyll content by 22%, increased leaf CO2 assimilation by 22%, increased vegetative biomass by 31% to 66%, and increased grain yield by 58%. Our results are consistent with the hypothesis that RCA improves plant growth under N-limiting conditions by decreasing root metabolic costs, thereby enhancing soil exploration and N acquisition in deep soil strata. Although potential fitness tradeoffs of RCA formation are poorly understood, increased RCA formation appears be a promising breeding target for enhancing crop N acquisition.Nitrogen (N) deficiency is one of the most limiting factors in maize (Zea mays) production worldwide (Ladha et al., 2005). In developing countries such as those in sub-Saharan Africa, less than 20 kg N ha−1 is applied to fields of smallholder farmers due to high fertilizer cost (Azeez et al., 2006; Worku et al., 2007). In developed countries, intensive N fertilization is used to maintain satisfactory yield (Tilman et al., 2002). In the United States, N fertilizers are the greatest economic and energy cost for maize production (Ribaudo et al., 2011). However, less than half of the N applied to crops is actually acquired, and most of the remaining N becomes a source of environmental pollution (Raun and Johnson, 1999; Smil, 1999; Tilman et al., 2002). For example, N and phosphorus (P) effluents into marine systems from agriculture cause eutrophication and hypoxic zones (Diaz and Rosenberg, 2008; Robertson and Vitousek, 2009). Nitrate contamination in surface water and groundwater systems poses serious health risks, such as methemoglobinemia and N-nitroso-induced cancers (UNEP and WHRC, 2007). Emission of nitrous oxides from agricultural activities contributes to ozone damage and global warming (Kulkarni et al., 2008; Sutton et al., 2011). Furthermore, the production of N fertilizers requires considerable energy from fossil fuels, and since energy costs have risen in recent years, farmers face economic pressure from increasing N fertilizer costs, which are linked to higher food prices. It is estimated that a 1% increase in crop N efficiency could save more than $1 billion (U.S.) annually worldwide (Kant et al., 2011). Therefore, even a small improvement in N efficiency would have significant positive impacts on the environment and the economy.Soil N is heterogenous and dynamic. The bioavailability of soil N depends on the balance between the rates of mineralization, nitrification, and denitrification. These processes are determined by several factors, including soil composition, microbial activity, soil temperature, and soil water status (Miller and Cramer, 2004). The predominant form of soil N available to plants in most agricultural systems is nitrate, which is highly soluble in water and thus mobile in the soil (Barber, 1995; Marschner, 1995). Mineralization of organic matter and/or the application of N fertilizer at the beginning of the growing season followed by precipitation and irrigation create a pulse of nitrate that may exceed the N acquisition capacity of seedlings and leach below the root zone. Therefore, it has been proposed that increasing the speed of root exploration of deep soil strata could benefit N acquisition (Lynch, 2013). However, the structural investments and metabolic expenditures of root systems are substantial and can exceed half of daily photosynthesis (Lambers et al., 2002). Therefore, full consideration of the costs and benefits of root systems is crucial for identifying root traits to improve crop production, especially in water- and nutrient-deficient environments (Lynch, 2007). Taking rhizoeconomics and the spatiotemporal availability of soil N into account, Lynch (2013) proposed a root ideotype for enhanced N acquisition in maize called Steep, Cheap, and Deep, in which Steep refers to architectural phenes and Cheap refers to phenes that reduce the metabolic cost of soil exploration. One element of this ideotype is abundant root cortical aerenchyma (RCA).RCA consists of enlarged air spaces in the root cortex (Esau, 1977). RCA is known to form in response to hypoxia, and the role of RCA in improving oxygen transport to roots of many plant species under hypoxic conditions has been well researched (Vartapetian and Jackson, 1997; Jackson and Armstrong, 1999; Mano and Omori, 2007, 2013). Interestingly, RCA can also form in response to drought and edaphic stresses such as N, P, and sulfur deficiencies (Drew et al., 1989; Bouranis et al., 2003; Fan et al., 2003; Zhu et al., 2010a), which suggests that the benefit of RCA extends beyond facilitating oxygen transport. Several lines of evidence suggest that RCA enhances root metabolic efficiency under stress. Fan et al. (2003) found that RCA formation significantly reduced root segment respiration and P content of root tissue, which allowed greater shoot growth in soils with low P availability. Under drought, maize genotypes with high RCA formation had greater root length, deeper rooting, better leaf water status, and 8 times greater yield than closely related genotypes with low RCA (Zhu et al., 2010a). Effects of RCA on root respiration were more pronounced for large-diameter roots compared with small-diameter roots (Jaramillo et al., 2013). Results from the functional-structural plant model SimRoot showed that RCA formation could be an adaptive response to deficiency of N, P, and potassium by decreasing the metabolic cost of soil exploration. By reducing root respiration, RCA decreases the carbon cost of soil exploration, and by decreasing the N and P content of root tissue, RCA permits internal reallocation of nutrients to growing root tissue, which is particularly beneficial under conditions of low N and P availability (Postma and Lynch, 2011a). Under suboptimal P availability, RCA increased the growth of a simulated 40-d-old maize plant by 70% (Postma and Lynch, 2011b). In the case of N, RCA increased the growth of simulated maize plants up to 55% in low-N conditions, and plants benefit from RCA more in high-N-leaching environments than in low-N-leaching environments (Postma and Lynch, 2011a). In addition, the formation of RCA decreases critical soil nutrient levels, defined as the soil fertility below which growth is reduced, suggesting that cultivars with high RCA may require less fertilizer under nonstressed conditions. These in silico results suggest that RCA has potential utility for improving crop nutrient acquisition in both high- and low-input agroecosystems.The overall objective of this research was to assess the utility of RCA for N acquisition in maize under N-limiting conditions. Maize near-isophenic recombinant inbred lines (RILs) sharing a common genetic background (i.e. descending from the same parents) with common root phenotypes but contrasting in RCA formation were grown under N stress to test the hypothesis that RCA formation is associated with reduced root respiration, reduced tissue nutrient content, greater rooting depth, enhanced N acquisition, and therefore greater plant growth and yield under N limitation.  相似文献

Pike fry were kept in 0.054-m3 tanks at densities of 50, 100 and 150 (277, 555, 833 fry m−2) for seven weeks during which the development of social and feeding behaviour was observed. Zooplankton, macro-invertebrates and perch fry were provided sequentially as food; the pike fry were allowed to feed ad libitum.
Zooplanktivorous fry stopped growing at 22 mm whilst, in the presence of abundant suitable alternative prey, 1–4% turned cannibalistic at 5 weeks of age; cannibals subsequently grew rapidly (mean 1.88 mm day−1). Cannibalism ensued in all tanks when the ratio of predator size: prey size was c .2:1.
Fry tended to space-out evenly in the tanks with no overt aggression or territoriality. Behaviour was typified by remaining still for long periods, particularly subsequent to the onset of cannibalism. Cannibals were attracted by fry movements which often initiated attacks.
Daily per capita mortality rates showed no crowding effects before cannibalism but significant density-dependent mortality due to cannibals. Consumption rates of cannibals varied between 0.63 and 6.0 fry per cannibal per day. Cannibals accounted for 54–96% of daily mortality in the experimental tanks. These results are discussed in relation to proposed mechanisms of pike population density regulation.  相似文献

Muscle mass is related to higher bone mass and a reduction in fracture risk. However, the interactions between muscle tissues and bone metabolism are incompletely understood and there might be some humoral factors that are produced in muscle tissues and exhibit bone anabolic activity. We therefore investigated the role of FAM5C in osteoblast differentiation and the interactions between muscle and bone. A reduction of endogenous FAM5C by siRNA reduced the levels of osterix, alkaline phosphatase (ALP) and osteocalcin (OCN) mRNA as well as the levels of type 1 collagen and β-catenin in mouse osteoblastic MC3T3-E1 cells and mouse calvarial osteoblasts, although FAM5C overexpression significantly antagonized the levels of osterix, ALP and OCN mRNA induced by bone morphogenetic protein-2 in C2C12 cells. The conditioned medium from FAM5C-overexpressed and -suppressed C2C12 cells increased and decreased the levels of osterix, ALP and OCN mRNA in MC3T3-E1 cells, respectively. In conclusion, the present study is the first to show that FAM5C enhances osteoblast differentiation in differentiated osteoblasts, and that the effects of the conditioned medium from FAM5C-modulated myoblastic cells were positively correlated with the effects of FAM5C on osteoblast phenotype in osteoblasts. FAM5C might be an important humoral bone anabolic factor produced from muscle cells.  相似文献

Stream dwelling invertebrates are ideal candidates for the study of ecological speciation as they are often adapted to particular environmental conditions within a stream and inhabit only certain reaches of a drainage basin, separated by unsuitable habitat. We studied an atypical population of the stonefly Leuctra hippopus at a site in central Norway, the Isterfoss rapids, in relation to three nearby and two remote conspecific populations. Adults of this population emerge about a month earlier than those of nearby populations, live on large boulders emerging from the rapids, and are short‐lived. This population also has distinct morphological features and was studied earlier during the period 1975–1990. We reassessed morphological distinctness with new measurements and added several analyses of genetic distinctness based on mitochondrial and nuclear sequence markers, as well as AFLP fingerprinting and SNPs mined from RAD sequences. The Isterfoss population is shown to be most closely related to its geographical neighbors, yet clearly morphologically and genetically distinct and homogeneous. We conclude that this population is in the process of sympatric speciation, with temporal isolation being the most important direct barrier to gene flow. The shift in reproductive season results from the particular temperature and water level regime in the Isterfoss rapids. The distinct adult body shape and loss of flight are hypothesized to be an adaptation to the unusual habitat. Ecological diversification on small spatial and temporal scales is one of the likely causes of the high diversity of aquatic insects.  相似文献

Clathrin-dependent endocytosis is a main entry mechanism for the glycolipid-binding Shiga toxin (Stx), although clathrin-independent pathways are also involved. Binding of Stx to its receptor Gb3 not only is essential for Stx retrograde transport to the endoplasmic reticulum and toxicity but also activates signaling through the tyrosine kinase Syk. We previously described that Syk activity is important for Stx entry, but it remained unclear how this kinase modulates endocytosis of Stx. Here we characterized the effects of Stx and Syk on clathrin-coated pit formation. We found that acute treatment with Stx results in an increase in the number of clathrin-coated profiles as determined by electron microscopy and on the number of structures containing the endocytic AP-2 adaptor at the plasma membrane determined by live-cell spinning disk confocal imaging. These responses to Stx require functional Syk activity. We propose that a signaling pathway mediated by Syk and modulated by Stx leads to an increased number of endocytic clathrin-coated structures, thus providing a possible mechanism by which Stx enhances its own endocytosis.  相似文献

Background and Aims

Root cortical aerenchyma (RCA) increases water and nutrient acquisition by reducing the metabolic costs of soil exploration. In this study the hypothesis was tested that living cortical area (LCA; transversal root cortical area minus aerenchyma area and intercellular air space) is a better predictor of root respiration, soil exploration and, therefore, drought tolerance than RCA formation or root diameter.

Methods

RCA, LCA, root respiration, root length and biomass loss in response to drought were evaluated in maize (Zea mays) recombinant inbred lines grown with adequate and suboptimal irrigation in soil mesocosms.

Key Results

Root respiration was highly correlated with LCA. LCA was a better predictor of root respiration than either RCA or root diameter. RCA reduced respiration of large-diameter roots. Since RCA and LCA varied in different parts of the root system, the effects of RCA and LCA on root length were complex. Greater crown-root LCA was associated with reduced crown-root length relative to total root length. Reduced LCA was associated with improved drought tolerance.

Conclusions

The results are consistent with the hypothesis that LCA is a driver of root metabolic costs and may therefore have adaptive significance for water acquisition in drying soil.  相似文献
